We hebben met succes aangepast nauwkeurig en gekalibreerd licht afgiftesysteem een mechanisme voor onderzoek optimalisering van R / NIR-LT in vitro. Golflengte en intensiteitswaarden van R / NIR-LT kunnen nauwkeurig en efficiënt worden gemanipuleerd met dit systeem. We stelden vast dat de lichtbehandeling van de cellen niet tot celdood, alhoewel ROS niet golflengten en doseringen geleverd verlaagd, in de celtypen getest. De maximale bereikt door het huidige systeem op 670nm (20.11W / m 2) intensitei...

Reactieve zuurstof species (ROS) zijn vereist voor verschillende signaaltransductieroutes en normale reacties van cellulair metabolisme, waaronder neuroprotectie 1. Wanneer echter endogene antioxidant mechanisme in staat zijn de productie van ROS controleren kunnen cellen bezwijken oxidatieve stress 2,3. Na letsel aan het CZS, zijn de stijgingen van de aanwezigheid van ROS en oxidatieve stress gedacht een belangrijke rol in de progressie van schade 4,5 spelen. Ondanks het grote aantal strategieën voor het verzwakken van oxidatieve stress die ervan zijn er nog geen volledig effectief klinisch relevante anti-oxidant strategieën voor het verzwakken ROS productie en geassocieerde oxidatieve stress in klinisch gebruik na neurotrauma 6. Daarom is de demping van oxidatieve stress blijft een belangrijk doel voor therapeutische interventie 7. Verbeteringen volgening R / NIR-LT gemeld bij diverse verwondingen en ziekten waaronder vermindering cardiale infarctgrootte, nier- en hepatische complicaties bij diabetes, retinale degeneratie, CNS letsel en beroerte 8, bijvoorbeeld door het verminderen van oxidatieve stress. Met bijzondere aandacht voor CNS letsel, hebben preklinische studies van werkzaamheid van 670nm licht goede effecten in modellen van retinale degeneratie 9-11, dwarslaesie 12, neuronale dood 13 getoond. Klinische studies zijn uitgevoerd voor de droge leeftijdsgebonden maculaire degeneratie en zijn momenteel aan de gang voor een beroerte 14, maar de uitkomsten van deze onderzoeken niet lijken veelbelovend, misschien te wijten aan een gebrek aan effectieve behandeling in dienst parameters 15. Als zodanig R / NIR-LT niet wijd goedgekeurd als onderdeel van de normale klinische praktijk in neurotrauma, ondanks een gemakkelijk toe te dienen, niet-invasieve en relatief goedkope behandeling. Belemmeringen voor de klinische vertaling verschillende ontbreken van een clvroeg begreep werkingsmechanisme en het ontbreken van een gestandaardiseerd effectieve behandeling protocol 16,17. Huidige literatuur over lichttherapie blijkt dat er een overvloed aan variatie in de behandeling parameters met betrekking tot de bestraling bronnen (LED of laser), golflengte (bijvoorbeeld 630, 670, 780, 810, 830, 880, 904nm), totale dosis (joules van bestraling / oppervlakte-eenheid), de duur (belichtingstijd), timing (pre- of post- belediging), behandeling frequentie en wijze van levering (puls of continue) 8. De variabiliteit in behandeling parameters tussen studies maakt vergelijking moeilijk en heeft bijgedragen aan de scepsis ten aanzien van de werkzaamheid 16. Daarom wordt de optimalisatie van R / NIR-LT duidelijk vereist, met celkweek systemen in staat om de high-throughput screening mechanisme nodig om de meerdere variabelen te vergelijken bieden. Er zijn echter weinig in de handel verkrijgbaar verlichting systemen die voldoende flexibiliteit en controle over wa kan biedenvelength en intensiteit van dergelijke optimalisatie experimenten. Commercieel verkrijgbare LED inrichtingen algemeen niet in staat om meerdere golflengten of intensiteiten, waardoor onderzoekers gebruik meerdere LED-inrichtingen van verschillende fabrikanten, die kunnen variëren niet alleen de intensiteit leveren, maar ook het spectrum van de golflengte van het uitgestraalde licht. We hebben dit probleem aangepakt door het gebruik van een breedband Xenon lichtbron gefilterd door smalbandinterferentie filters, waardoor een gebied van golflengten en invloeden van licht genereren, waardoor afsluiten, nauwkeurige regeling van de parameters van R / NIR-LT. Het is belangrijk op te merken dat de therapeutische dosis van de behandeling wordt bepaald door het aantal fotonen interactie met de photoacceptor (chromofoor), die, in het geval van R / NIR-LT is gepostuleerd om cytochroom c oxidase (COX) 18. Foton energie geleverd varieert met de golflengte; betekent gelijke doses van energie bij verschillende golflengten zal comgewaardeerde verschillende aantallen fotonen. Daarom is het licht uitgezonden door de inrichting werd geijkt met een gelijk aantal fotonen uitzenden voor elk van de gekozen golflengten te testen. We hebben een systeem dat kan worden gebruikt om R / NIR-LT leveren tegen een gebied van golflengten en intensiteiten cellen in vitro ontwikkelde en de mogelijkheid aangetoond om de effecten van de geleverde R / NIR-LT op ROS productie in cellen blootgesteld meten glutamaat stress.

Rood / nabij-infrarood licht therapie (R / NIR-LT), laser of licht emitterende diode (LED) afgeleverd, verbetert de functionele en morfologische resultaten bij verschillende centraal zenuwstelsel letsels in vivo, waarschijnlijk door het verminderen van oxidatieve stress. Echter, zijn effecten van R / NIR-LT op oxidatieve stress is aangetoond variëren naargelang de golflengte of sterkte van de straling. Studies behandeling parameters te vergelijken ontbreken, door het ontbreken van commercieel beschikbare apparaten die meerdere golflengtes of intensiteiten, geschikt voor high-throughput in vitro optimalisatie studies leveren. Dit protocol beschrijft een techniek voor de levering van licht bij verschillende golflengten en intensiteiten therapeutische doses die voor een bepaalde traumamodel optimaliseren. Onze hypothese was dat een werkwijze voor het afgeven licht, waarbij golflengte en intensiteit parameters eenvoudig kunnen worden veranderd, kan bepaling van een optimale dosis van R / NIR-LT voor het verminderen reactieve zuurstof species bevorderen(ROS) in vitro. Niet-coherente Xenon licht werd gefilterd door smalband interferentie filters aan variërende golflengten (centrum golflengtes van 440, 550, 670 en 810nm) en invloeden te leveren (8,5 x 10 -3 tot 3,8 x 10 -1 J / cm2) van het licht gekweekte cellen. Lichtopbrengst van het apparaat werd gekalibreerd om therapeutisch relevante, gelijk quantale doses van licht uitzenden bij elke golflengte. Reactieve soorten werden gedetecteerd in glutamaat gewezen cellen behandeld met het licht, met DCFH-DA en H 2 O 2 fluorescente kleurstoffen. We succesvol afgeleverd licht bij diverse fysiologisch en therapeutisch relevante golflengten en intensiteiten gekweekte cellen blootgesteld aan glutamaat als een model van CNS letsel. Hoewel de invloeden van R / NIR-LT gebruikt in de onderhavige studie geen effect op ROS geproduceerd door de gekweekte cellen te oefenen, de wijze van licht levering voor andere systEMS waaronder geïsoleerde mitochondria of meer fysiologisch relevante organotypische slice cultuur modellen, en kan worden gebruikt om effecten op verschillende uitkomstmaten van oxidatief metabolisme te beoordelen.

De lichtopbrengst afgegeven bij een golflengte van 670 nm werd gekalibreerd met grijsfilters om cellen te bestralen met verschillende invloeden omvat een dosis van 670 nm licht eerder gebleken bij vivo (0,3 J / cm2) 20 zijn.
