Chemische synaptische transmissie tussen neuronen is een belangrijk mechanisme van communicatie in het zenuwstelsel. Zij beroept zich op de afgifte van neurotransmitters door middel van een dynamische cyclus van vesikel fusie en de afhaling aan het presynaptische website. Veel van de betrokken blaasje dynamiek eiwitten zijn geïdentificeerd; Hun specifieke bijdrage aan het verschijnsel nog worden opgehelderd 1. Ons begrip wordt gedeeltelijk beperkt door het feit dat de meest gebruikte testen voor exo / endocytose niet altijd de meest geschikte. Verschillende studies met betrekking tot de vesikel fusie en dynamiek vertrouwen op elektrofysiologische technieken. tirfm and ph-sensitive gfp-probes to evaluate neurotransmitter vesicle dynamics in sh-sy5y neuroblastoma cells: cell imaging and data analysis

Synaptische blaasjes los neurotransmitters bij chemische synapsen door middel van een dynamische cyclus van fusie en ophalen. Monitoring synaptische activiteit in real time en het ontleden van de verschillende stappen van exo-endocytose bij de enkele vesicle niveau cruciaal voor het begrijpen synaptische functies in gezondheid en ziekte. Genetisch gecodeerde pH-gevoelige sondes direct gericht aan synaptische blaasjes en totale interne reflectie fluorescentiemicroscopie (TIRFM) zorgen voor de ruimtelijke resolutie nodig is om blaasje dynamiek volgen. Het oneindig kleine veld door totale interne reflectie alleen prikkelen fluoroforen in een dunne laag (&lt;150 nm) boven de glasplaat waarop cellen hechten, waar de processen van exo-endocytose plaatsvinden. De resulterende beelden met hoog contrast zijn bij uitstek geschikt voor het bijhouden van blaasjes en kwantitatieve analyse van fusie-evenementen. In dit protocol, SH-SY5Y menselijke neuroblastoma cellen worden voorgesteld als een waardevol model voor het bestuderen van neurotransmitter afgifte bij de enkele-vesikel niveau TIRFM vanwege hun platte oppervlak en de aanwezigheid van gedispergeerde vesicles. De methoden voor het kweken SH-SY5Y als hechtende cellen en het transfecteren hen synapto-pHluorin voorzien, alsook de techniek om TIRFM en beeldvorming uitvoeren. Tenslotte wordt een strategie voor selecteren, tellen en analyseren fusiegebeurtenissen in whole-cell en single-vesikel niveaus gepresenteerd. Om de beeldvorming procedure en de data-analyse benadering te valideren, zijn de dynamiek van pHluorin-tagged blaasjes geanalyseerd onder rust en gestimuleerd (depolariserende kalium concentraties) voorwaarden. Membraandepolarisatie verhoogt de frequentie van fusie evenementen en veroorzaakt een parallelle verhoging van de netto fluorescentie signaal opgenomen in hele cel. Single-blaasje analyse blijkt modificaties van fusie-event gedrag (verhoogde piek hoogte en breedte). Deze gegevens suggereren thop kalium depolarisatie niet alleen leidt tot een massale afgifte van neurotransmitters, maar wijzigt ook het mechanisme van de vesikel fusie en recycling. Met de juiste fluorescente probe, kan deze techniek worden toegepast in verschillende cellulaire systemen om de mechanismen van constitutieve en gestimuleerde secretie ontleden.

De TIRF beeldvorming en data-analyse procedures beschreven zijn ontworpen om blaasjes dynamiek in cellulaire systemen te bestuderen. Deze techniek kan worden gebruikt om de effecten van signaalmoleculen en drugs op fusiegebeurtenissen en neurotransmitter blaasje dynamiek 17 bepalen.
