De overgang van een scarless littekens fenotype optreedt in het derde trimester van de zwangerschap bij de mens en ongeveer embryonale dag 18 (E18) bij muizen 2,3. In vergelijking met volwassen, foetale wondheling gekenmerkt door snelle epithelisatie, bindweefsel en fibroblast migratie. Vele studies hebben mogelijke verklaringen voor het fenomeen van scarless wondgenezing aangeboden tijdens de vroege ontwikkeling van de foetus. Ontsteking is een fundamenteel onderdeel van volwassen wondheling; echter, foetale wonden gekenmerkt door het ontbreken van acute ontsteking 4. Of dit is een gevolg van de functionele onvolwassenheid van het immuunsysteem tijdens de foetale stadia blijft onduidelijk. Een recente studie suggereert dat de verschillen in de overvloed, maturity en functie van mestcellen in E15 vs E18 foetale huid kan verantwoordelijk zijn voor de overgang van een scarless fenotype, althans in de muis 3 zijn. Longaker en collega&#39;s toonden aan dat wondvocht uit de foetus beschikt over een hoog niveau van hyaluronzuur stimulerende activiteit vergeleken met geen bij volwassen wondvocht 6. Bijgevolg kan hogere hyaluronzuur, een glycosaminoglycaan die een micro bevorderlijk celmotiliteit en proliferatie bevordert, in het foetale wondmilieu verantwoordelijk voor de scarless fenotype waargenomen tijdens de vroege foetale ontwikkeling. Andere bewijslijnen wijzen erop dat de fetal wondmilieu is relatief hypoxemisch en ondergedompeld in steriele vruchtwater rijk aan groeifactoren 7. Er is echter geen definitief antwoord is voorzien van een kritieke gebeurtenis of factor tijdens de embryogenese dat de overgang van scarless regeneratie tot fibrotische reparatie triggers. Daarnaast kunnen kleine wijzigingen van dit model worden gebruikt om een aantal verdere studies, zoals genexpressie analyse van foetale wonden en huid 8,9 voeren. Gezien het feit dat precies getimede zwangerschappen zijn van cruciaal belang voor een succesvolle recapitulatie van deze foetale scarless wondgenezing model, hebben we ook detail ons protocol voor superovulatie getimed zwangerschappen.

Early in utero, but not in postnatal life, cutaneous wounds undergo regeneration and heal without formation of a scar. Scarless fetal wound healing occurs across species but is age dependent. The transition from a scarless to scarring phenotype occurs in the third trimester of pregnancy in humans and around embryonic day 18 (E18) in mice. However, this varies with the size of the wound with larger defects generating a scar at an earlier gestational age. The emergence of lineage tracing and other genetic tools in the mouse has opened promising new avenues for investigation of fetal scarless wound healing. However, given the inherently high rates of morbidity and premature uterine contraction associated with fetal surgery, investigations of fetal scarless wound healing in vivo require a precise and reproducible surgical model. Here we detail a reliable model of fetal scarless wound healing in the dorsum of E16.5 (scarless) and E18.5 (scarring) mouse embryos.
