Den presenterade mikrokosmos installationen visade lämpligt att studera biotillgängligheten av rumsligt separerade kemikalier till förnedrande organismer efter upptag och transport av mycel. Potentiell transport av delvis flyktiga föreningar gasfas förhindras och bakterie bioreporter celler kan visualiseras utan arbetade provberedning och därmed med minimal störning av känsligt system. Samtidigt, kan kemisk analys av provet lätt bedrivas möjliggör en god kontroll av uppnådda resultat och för kvantifiering a...

Marken är tätt befolkat av ett brett spektrum av mikroorganismer 1,2 såsom bakterier. Men förhållandena i denna livsmiljö är utmanande, särskilt när det gäller tillgången på vatten 3. Bakterier permanent behöver söka efter optimala förhållanden i heterogena miljöer fyra, men frånvaron av kontinuerliga vatten filmer leder till begränsad rörlighet 5 hindrar dem att sprida sig fritt. Dessutom är diffusionshastigheter av lösta ämnen (t.ex. näringsämnen) sänks vid omättade förhållanden 6. Således är bakterier och näringsämnen ofta fysiskt separerade och närings tillgängligheten är begränsad 3. Som en konsekvens, kan en transportvektor för kemiska föreningar som inte kräver en kontinuerlig vatten-fasen bidrar till att övervinna dessa begränsningar. I själva verket har många mikroorganismer såsom svampar och oomyceter utvecklat en fintrådiga tillväxt formulär som gör det möjligt för dem att växa genom luftfyllda porutrymmen därigenom nå och mobilizing även fysiskt separerade näringsämnen 7 och kolhaltiga 8 ämnen över långa avstånd. De kan även fungera som biologiska transport vektorer som levererar socker och andra energikällor till bakterier 9. Upptag och transport i mycelie organismer har även visats för hydrofoba organiska föroreningar såsom polycykliska aromatiska kolväten (PAH) i Pythium ultimum 10 eller i arbuskulära mykorrhizasvampar 11. Eftersom PAH är allestädes närvarande och dåligt vattenlösliga föroreningar 12 i jord, kanske mycel-medierad transport bidra till att öka förorenings biotillgänglighet för potentiella bakterie nedbrytare. Det totala beloppet för föroreningsspridning kan kvantifieras direkt genom kemisk väg 10, biotillgänglighet av föroreningar som transporteras av mycel till nedbrytande bakterier och andra organismer kan inte bedömas lätt. Följande protokoll presenterar en metod för att utvärdera effekten av mycelia på förorening biotillgänglighet för bakteriella nedbrytare i ett direkt sätt; det tillåter samla information om Spatiotemporal effekterna av föroreningar på mikrobiella ekosystem. Mer i detalj, var tre-ring PAH fluoren, den myceliala organismen Pythium ultimum och bioreporter bakterien Burkholderia sartisoli RP037-mChe 13 appliceras i de beskrivna mikrokosmos uppställningar. Bakterien B. sartisoli RP037-mChe byggdes ursprungligen för att studera fenantren flöden till cellen 14 och uttrycker förbättrade grönt fluorescerande protein (EGFP) som ett resultat av PAH flödet tillcellen, medan den röda fluorescerande mCherry uttrycks konstitutivt. Detaljerad information om reportern konstruktionen ges av al. Tecon et 13 I preliminära tester visade bakterien ingen simning och endast mycket långsamt myllrande förmåga. Det kunde migrera långsamt på hyfer av Pythium ultimum när den appliceras som en tät suspension ovanpå hyfer. Eftersom bakterier inbäddade i agaros i följande protokoll, gjorde migration på hyfer inte förekomma. Använda konfokal laserscanningsmikroskopi (CLSM) kan bioreporter bakterier visualiseras direkt i mikrokosmos och expression av EGFP kan kvantifieras i förhållande till mängden av celler (i proportion till mCherry signal) med hjälp av programvaran ImageJ. Detta gör att man jämför biotillgängligheten kvalitativt i olika scenarier (dvs högre eller lägre). FLU befanns vara biotillgängligt efter mycel transporter av P. ultimum (dvs, detvar högre än i en negativ kontroll). Vidare beskriver protokollet hur man kvantifiera den totala mängden mycel-medierad transport via kemiska medel och att kontrollera förorenings biotillgänglighet använder silikonbelagda glasfibrer (SPME fibrer) i identiska mikrokosmos. Resultat som använder detta mikrokosmos inställning har publicerats och diskuterats för kombinationen av P. ultimum, fluoren och B. sartisoli RP037-mChe 15. Här ligger fokus på en detaljerad metodbeskrivning och identifiering av potentiella fallgropar i protokollet för att tillhandahålla denna kunskap för potentiella nya ansökningar. Ytterligare tillämpningar kan innebära olika svamp-, bakteriearter (t.ex., från förorenade områden), och andra föroreningar (t.ex. bekämpningsmedel) eller förorening-försörjning (t.ex. åldern jordar).

a whole cell bioreporter approach to assess transport and bioavailability of organic contaminants in water unsaturated systems

Bioavailability of contaminants is a prerequisite for their effective biodegradation in soil. The average bulk concentration of a contaminant, however, is not an appropriate measure for its availability; bioavailability rather depends on the dynamic interplay of potential mass transfer (flux) of a compound to a microbial cell and the capacity of the latter to degrade the compound. In water-unsaturated parts of the soil, mycelia have been shown to overcome bioavailability limitations by actively transporting and mobilizing organic compounds over the range of centimeters. Whereas the extent of mycelia-based transport can be quantified easily by chemical means, verification of the contaminant-bioavailability to bacterial cells requires a biological method. Addressing this constraint, we chose the PAH fluorene (FLU) as a model compound and developed a water unsaturated model microcosm linking a spatially separated FLU point source and the FLU degrading bioreporter bacterium Burkholderia sartisoli RP037-mChe by a mycelial network of Pythium ultimum. Since the bioreporter expresses eGFP in response of the PAH flux to the cell, bacterial FLU exposure and degradation could be monitored directly in the microcosms via confocal laser scanning microscopy (CLSM). CLSM and image analyses revealed a significant increase of the eGFP expression in the presence of P. ultimum compared to controls without mycelia or FLU thus indicating FLU bioavailability to bacteria after mycelia-mediated transport. CLSM results were supported by chemical analyses in identical microcosms. The developed microcosm proved suitable to investigate contaminant bioavailability and to concomitantly visualize the involved bacteria-mycelial interactions.
