De homopentameric vorm is verantwoordelijk voor kanaal activiteit in vlakke lipidendubbellagen 22,23. De correcte oriëntatie van de TM monomeren in de lipide bilaag werd eerst bepaald middels plaatsspecifieke infrarood dichroïsme 23, waaruit bleek His-22 in een lumen, dicht bij inter-spiraalvormige oriëntatie. Dezelfde TM domein oriëntatie werd bevestigd door NMR studies dat de pentamere reconstrueerde-helix bundel van het eiwit van volledige lengte in dodecylphosphocholine (DPC) micellen 22. In deze &quot;micel-model, werd een a- schroeflijnvormige TM domein N-terminaal geflankeerd door een a-helix en C-terminaal van een uitgebreide B-haarspeld. De twee protoneerbare residuen van SH eiwit, His-22 en His-51 worden in de TM domein (lumenally georiënteerd), en aan het uiteinde van de extramembrane C-terminale β haarspeld (ver van de zender porie), respectievelijk. In een bicellar Envirooverschrijdende aanpak echter de TM α-helix strekt zich uit tot His-51, en ​​beide zijn resten zijn toegankelijk voor het lumen van het kanaal 24. De kanaalstructuur neemt een trechtervormig architectuur 22, waarbij het ​​smallere gebied (Ser-29, Cys-45) 22 is bekleed met hydrofobe zijketens (Ile-32, Ile-36, Ile-40 en Leu-44), en definieert Ile-36 het smalste punt in het kanaal lumen. His-22 bevindt zich op de grootste opening van de trechter, terwijl His-51 aan het uiteinde van de kleinste opening. In dit document, is analytische centrifugatie in een sedimentatie evenwicht modus gebruikt om te bepalen of zijn protonering beïnvloedt de stabiliteit van de SH eiwit pentameer. In dit geval werd SH eiwit opgelost in C14-betaine detergent, die eerder gewend SH eiwit vormt oligomeren pentameer 22 tonen.

sedimentation equilibrium of a small oligomer-forming membrane protein: effect of histidine protonation on pentameric stability

Analytische ultracentrifugatie (AUC) kan worden gebruikt om reversibele interactie tussen macromoleculen over een breed scala van interactie sterke en onder fysiologische omstandigheden te bestuderen. Dit maakt AUC een methode van keuze om kwantitatief te beoordelen stoichiometric en thermodynamica van homo- en hetero-vereniging die van voorbijgaande aard en omkeerbaar in biochemische processen zijn. In de modaliteit van sedimentatie evenwicht (SE), een evenwicht tussen diffusie en sedimentatie levert een profiel als functie van de radiale afstand die afhankelijk is van een specifiek associatiemodel. Hierin wordt een gedetailleerd SE protocol beschreven om het formaat en monomeer-monomeer associatie energie van een klein membraaneiwit oligomeer met een analytische ultracentrifuge bepalen. AUC-ES-label vrij, enkel op basis van fysische principes, en kan worden gebruikt zowel wateroplosbare en membraaneiwitten. Een voorbeeld wordt getoond van de laatste kleine hydrofoob (SH) eiwit in het menselijk respiratoir syncytieel virus (HRSV), Een 65 aminozuur polypeptide met een α-helix transmembraan (TM) domein dat pentameer ionenkanalen vormen. NMR-gebaseerde structurele gegevens blijkt dat SH eiwit twee protoneerbare His residuen in het transmembraan domein dat georiënteerd tegenover het lumen van het kanaal. SE experimenten werden ontworpen om te bepalen hoe pH beïnvloedt associatieconstante en de oligomere grootte van SH eiwit. Terwijl de pentamere vorm werd bewaard in alle gevallen werd de associatieconstante gereduceerd bij lage pH. Deze gegevens zijn in overeenstemming met een vergelijkbare pH-afhankelijkheid waargenomen voor SH kanaal activiteit, consistent met een lumenale oriëntatie van de twee Zijn residuen in SH-eiwit. Dit laatste kan bij lage pH elektrostatische afstoting en verminderde stabiliteit oligomeer ervaren. Kortom, deze werkwijze geldt wanneer kwantitatieve gegevens over subtiele eiwit-eiwit associatie veranderingen in fysiologische omstandigheden moeten worden gemeten.

De radiale verdeling profiel van C14SB detergens micellen in 50 mM Tris, 100 mM NaCl pH 7,3 vormt een zeer ondiepe exponentiële dat een lineair model (figuur 7A) worden aangebracht.
