Hepatocellulair carcinoom (HCC) is de 3 belangrijkste oorzaak van kankersterfte ter wereld 1. Nog slechts 30% van de HCC patiënten komen in aanmerking voor curatieve chirurgische behandelingen 2 en systemische chemotherapie niet effectief 3. Daarom is er dringend onvervulde medische behoefte aan nieuwe therapieën HCC en de ontwikkeling van modelsystemen geschikt voor het efficiënt testen van nieuwe middelen. De chick chorion-membraan (CAM) assay levert een reproduceerbare, rendabele en snelle middellange-throughput werkwijze voor het testen van potentiële anti-tumor drugs in vivo. De CAM-assay is uitgebreid toegepast om angiogenese te bestuderen 4. Het is ook met succes ontwikkeld tot een tumor xenograft model van kanker, zoals glioblastoom 5, 6 pancreaskanker, melanoom 09/07, 11/10 en osteosarcoom. Zowel in ovo 12 en ex ovo13 technieken zijn toegepast in de literatuur met gegevens variërend van protocol protocol. Een belangrijke uitdaging voor de CAM xenotransplantaatmodel is de relatief hoge incidentie van embryonale sterfte na manipulatie van het ei, met gepubliceerde kippenembryo sterftecijfers variërend 25-50 procent 11-14. In dit artikel beschrijven we de ontwikkeling van een in ovo xenotransplantaatmodel van HCC die betrouwbaar produceert groei van drie-dimensionale, gevasculariseerde tumoren die histologisch lijken ongedifferentieerde HCC. We hebben een protocol voor het eerst beschreven door Ossowski et al. 14 aangepast en chick embryonale overleving tot 93% met een extreem hoge tumor implantatie hebben bereikt.

De chick chorion-membraan (CAM) begint te ontwikkelen op dag 7 na de bevruchting en rijpt op dag 12. De cam is natuurlijk immunodeficiënte en sterk doorbloed, waardoor het een ideaal systeem voor tumor implantatie. Voorts bevat de CAM extracellulaire matrixeiwitten zoals fibronectine, laminine, collageen, integrine alfa (v) beta3 en MMP-2, waardoor het een aantrekkelijk model voor tumor invasie en metastase te onderzoeken. Wetenschappers hebben lang gebruik gemaakt van de fysiologie van de CAM door het te gebruiken als een model van angiogenese. Meer recent heeft de CAM-assay aangepast om te werken als een in vivo xenograft model voor verschillende kankers die de kloof tussen basic in vitro werk en complexere dierlijke modellen van kanker overbrugt. De CAM-assay maakt de studie van tumorgroei, antitumor therapieën en pro-tumor moleculaire pathways in een biologisch systeem waarin zowel kosten- en tijd-effectief. We beschrijven hier de ontwikkeling van de CAM xenograft model van hepatocellulair carcinoom (HCC) embryonale overleving tot 93% en betrouwbare tumor nemen leidt tot groei van driedimensionale, gevasculariseerde tumoren.
