Aufgrund der jüngsten schnellen technologischen Entwicklungen in der Mikroskopie, einschließlich optischer Bildverarbeitung, ist die Analyse von Zellen im Rahmen des gesamten Gewebes für Immunologen immer zugänglich. Die Charakterisierung der einzelnen Zellen in Suspension eine wertvolle und unverzichtbare Methode, um zelluläre und molekulare Funktion zu verstehen. Jedoch ist die Analyse der Zellen in ihrem (Mikro-) -anatomical Umgebung wesentlich für das Verständnis der Wechselwirkungen zwischen verschiedenen Zelltypen, die in komplexen Prozessen zusammenarbeiten wie die Entwicklung von Immunantworten. Während es relativ einfach für Mikroskopikern qualitative Informationen aus Bildern erhalten bleibt es eine Herausforderung, diese Daten zu quantifizieren, teilweise aufgrund der Tatsache, dass die Analyse-Methoden in diesem Bereich hinter Vergleich zu dem, was in der Bildaufnahme möglich zurückbleibt. Viele Forscher verlassen sich immer noch auf zeitraubende manuelle Zellzählung in der Histologie Bilder,damit die Einführung einer Vorspannung zwischen verschiedenen Rater und behindern die Replikation von anderen Gruppen. Oft wird ein repräsentatives Bild gewählt, um eine Aussage über zelluläre Position oder Co-Lokalisation in einer Publikation zu unterstreichen, dass es schwer für den Leser auf die statistische Relevanz eines solchen Ereignisses zu beurteilen. Zusammen mit der Tatsache, dass das volle Informationsgehalt der Bilddaten wird nur selten genutzt, unterstreicht dies die Notwendigkeit für eine unvoreingenommene, schneller und umfassender Ansatz zur histologischen Bilder analysieren. Das Knochenmark ist ein komplexes Gewebe, die auf wichtige Lebensfunktionen als Organ der Hämatopoese bei erwachsenen Wirbeltieren findet. Abgesehen davon, dass der Geburtsort für hämatopoetische Zellen 1,2 und spielen eine wichtige Rolle in B-Lymphozyten-Entwicklung 3, wirkt es auch als ein Ort, an dem Immunreaktionen ausgelöst werden 4 und unterstützt reifen, rezirkulierenden B-Zellen 5. Zusätzlich ihre Rolle bei der Aufrechterhaltung immunological Speicher wird zunehmend in den letzten zehn Jahren geschätzt, weil verschiedene Arten von Zellen Immungedächtnis bilden haben sich dort aufzuhalten, 6-9. Die Beziehung zwischen der komplexen Gewebearchitektur des Knochenmarks und seine Funktionen nach wie vor schwer. Im Gegensatz zu sekundären lymphatischen Organe, die in Makro Fächer wie T- und B-Zell-Zonen organisiert sind, fehlt dem Knochenmark eine klare Makro Kompartimentierung. Bisher getrennte Kompartimente im Knochenmark sind durch ihre Nähe zu der Knochenrinde bzw. an Vaskulatur definiert. Die Bedeutung der verschiedenen resident stromalen Zellpopulationen im Knochenmark für eine Reihe von Prozessen, wie Stütz Stammzellen, die Entwicklung von B-Zellen oder die Wartung von Gedächtnis-Immunzellpopulationen (wie langlebige Plasmazellen (PCs), CD4 + und CD8 + Gedächtnis-T-Zellen), zeigt deutlich, dass es einen gewissen Grad von Mikro-Kompartimentierung im Knochenmark. <pclass = &quot;jove_content&quot;&gt; Diese Beobachtungen haben zu dem Konzept der unterschiedlichen microanatomical Nischen, die in bestimmten Funktionalitäten (Erhaltung von Stammzellen, B-Zell-Entwicklung bei verschiedenen Stufen, und die Wartung des immunologischen Gedächtnisses) im Knochenmark spezialisierten geführt. Zwar scheint es eine gewisse Heterogenität zwischen den Nischen, die verschiedenen Funktionen dienen, einige der Faktoren, die von Stromazellen, wie CXC-Chemokin-Liganden 12 (CXCL12) oder Interleukin 7 (IL-7), hergestellt werden, sind wichtige Komponenten für einige dieser Nischen 10. Die Visualisierung und Charakterisierung von Stromazellen des Knochenmarks ist schwierig aufgrund ihrer morphologischen Merkmale mit langen, dünnen dendritischen Fortsätze bilden ein Netzwerk in der gesamten Knochenmark und das Fehlen geeigneter Marker stromal Subpopulationen zu unterscheiden. Es ist noch nicht klar, inwieweit diese Nischen gemeinsame Merkmale hinsichtlich ihrer zellulären und molekularen composition, und welche Elemente machen eine bestimmte Nische einzigartig. Neben Stromazellen wurden hämatopoetischen Zelltypen wurde gezeigt, dass eine entscheidende Rolle, indem sie bestimmte Signale zumindest für einige der Nischen spielen. Offensichtlich ist die Komplexität der Nische Zusammensetzung erfordert deren Analyse in situ, und es wird immer wichtiger für Immunologen und Hämatologen zum Vergrößern auf das Knochenmark-Mikroarchitektur, zum Beispiel durch die Analyse der räumlichen Beziehungen zwischen ihren zellulären Komponenten. Hier wird eine Strategie, um zelluläre Kolokalisation und Nachbarschaftsbeziehungen im Knochenmark in einem automatisierten und unvoreingenommene Weise zu quantifizieren wird vorgestellt. Eine detaillierte Arbeitsablauf einschließlich der Erzeugung von chimären Mäusen, Beherbergung Fluoreszenz Stromazellen und nichtfluoreszierenden hämatopoetischen Zellen, Herstellung von histologischen Schnitten von entkalkte Knochen, Akquisition von konfokalen Bildern, die den gesamten Knochen, sowie die automatische Bildanalyse der zellulären co-Lokalisierung und dessen Validierung / Diskriminierung aus zufälligen Positionierung durch ein Simulationstool zur Verfügung gestellt (Abbildung 8).

Konfokale Mikroskopie ist das Verfahren der Wahl für die Analyse der Lokalisierung von mehreren Zelltypen in komplexen Geweben, wie Knochenmark. Jedoch ist die Analyse und Quantifizierung der zellulären Lokalisation schwierig, da es in vielen Fällen beruht auf manueller Zählung, wodurch damit das Risiko der Einführung einer Bewerterabhängigen Bias und Verringerung Interraterreliabilität. Darüber hinaus ist es oft schwierig, ob die Co-Lokalisation von zwei Zellen ergibt sich aus beliebiger Positionierung beurteilen, insbesondere wenn die Zelltypen unterscheiden sich stark in der Häufigkeit ihres Auftretens. Hier wird ein Verfahren zur Quantifizierung der zellulären unvoreingenommene Kolokalisation im Knochenmark eingeleitet. Das Protokoll beschreibt die Probenvorbereitung zur histologischen Schnitten der gesamten murinen langen Knochen wie dem Knochenmark zu erhalten, sowie die Färbungsprotokoll und den Erwerb von hochauflösenden Bildern. Eine Analyse Workflow spannt sich von der Anerkennung der blutbildenden und nicht-Hematopoietic Zelltypen in 2-dimensionale (2D) Knochenmark Bilder in der Quantifizierung der direkt zwischen diesen Zellen präsentiert wird. Dies beinhaltet auch eine Nachbarschaftsanalyse, um Informationen über die zelluläre Mikroumgebung eines bestimmten Zelltyps zu erhalten. Um zu bewerten, ob die Co-Lokalisierung von zwei Zelltypen ist das bloße Ergebnis zufälliger Zellpositionierung oder reflektiert bevorzugte Verbindungen zwischen den Zellen, ein Simulationswerkzeug, das für die Prüfung dieser Hypothese im Falle von hämatopoetischen sowie Stromazellen, ist verwendet. Dieser Ansatz ist nicht auf die Knochenmark beschränkt und kann auf andere Gewebe verlängert werden, um reproduzierbare quantitative Analyse der histologischen Daten ermöglichen.

Automated Quantification of Hematopoietic Cell ÃƒÂ¢Ã¢â€šÂ¬Ã¢â‚¬Å“ Stromal Cell Interactions in Histological Images of Undecalcified Bone

A strategy to quantitatively analyze histological data in the bone marrow is presented. Confocal microscopy of fluorescently labeled cells in tissue sections results in 2-dimensional images, which are automatically analyzed. Co-localization analyses of different cell types are compared to data from simulated images, giving quantitative information about cellular interactions.
