immunohistochemistry and multiple labeling with antibodies from the same host species to study adult hippocampal neurogenesis

Нейрогенез является строго регулируется, многоступенчатый процесс, в котором новые нейроны генерируются из активированного нервной стволовой клетки с помощью более совершенных промежуточных подтипов предшественников. Каждый из этих подтипов выражает набор конкретных молекулярных маркеров, которые, вместе с конкретными морфологических критериев, могут быть использованы для их идентификации. Как правило, иммунофлуоресцентные методы применяются с участием подтипов специфических антител в сочетании с экзо- или эндогенных маркеров пролиферации. Здесь мы опишем иммунного методы для обнаружения и количественного определения всех этапах взрослого гиппокампа нейрогенеза. Они включают в себя применение тимидина аналогов, transcardial перфузии, обработки ткани, тепло-индуцированной поисковых эпитоп, ABC иммуногистохимических, рассеянный непрямой иммунофлюоресценции, конфокальной микроскопии и сотовый количественной оценке. Кроме того, мы представляем очередную несколько протоколов иммунофлюоресценции, которая обходит проблемы ˙Usually, вытекающие из необходимости использования первичных антител, поднятые в одних и тех же видов хозяев. Это позволяет точно идентифицировать все гиппокампа подтипов предшественников вместе с маркером пролиферации в пределах одного раздела. Эти методы являются мощным инструментом для изучения регуляции различных подтипов предшественников параллельно, их участие в патологий головного мозга и их роль в специфических функций головного мозга.
