Dos regiones del cerebro generar nuevas neuronas constitutivamente largo de la vida, la zona subventricular de los ventrículos laterales y la zona subgranular (SGZ) del giro dentado del hipocampo (DG). Las neuronas recién nacidas se derivan de las células progenitoras neurales y pasan por diferentes etapas de desarrollo morfológico y fisiológico antes de alcanzar la madurez 1,2. Desde un dividiendo lentamente-glía radial como células madre (tipo 1) etapas consecutivas de tránsito amplificar surgen células progenitoras intermedias. Los subtipos más indiferenciados (tipo 2a y 2b tipo) tienen una forma irregular con cortos, procesos tangenciales. Generan neuroblastos (tipo 3) que salen gradualmente del ciclo celular para convertirse en neuronas inmaduras (con dendritas extienden hacia la capa molecular) y finalmente integrar en la red del hipocampo células granulares como maduros. Debido a sus características fisiológicas estas células proporcionan los circuitos con una mejora de la plasticidad 3 sugeting un papel único en la función del hipocampo. Análogos de timidina incorporan en el ADN durante la fase S del ciclo celular y permiten nacimiento de citas, la cuantificación y el destino análisis de las células recién nacidos 6-8. La aplicación secuencial de los diferentes análogos de timidina (por ejemplo, CldU, EdU o IDU) puede ser usado para estudiar la renovación celular o poblaciones de células nacidos en diferentes puntos de tiempo durante el curso de un experimento 9. Una alternativa, marcador endógeno para la proliferación celular es Ki67. Para analizar el fenotipo de las poblaciones de células recién nacidas en el adulto dentate gyrus varios marcadores moleculares específicos de la etapa se pueden utilizar como GFAP, nestina, DCX y NeuN 1,6. GFAP es un marcador de astrocitos maduros pero también se expresa en células gliales-como radiales en el cerebro anterior adulto. Nestin es un filamento intermedio específico para células gliales como radiales y células progenitoras intermedios tempranos. DCX es una proteína asociada a microtúbulos expresado en progenitores intermedios, los neuroblastos y las neuronas inmaduras. Sobre la base de la (co) expresión de estos tres marcadores y las características morfológicas de las células marcadas cuatro subtipos de células progenitoras se pueden identificar diferentes: tipo 1 (GFAP +, nestina +, DCX -), tipo 2a (GFAP -, nestina + , DCX -), el tipo 2b (GFAP -, nestina +, DCX +) y tipo 3 (GFAP -, nestina -, DCX +) 1. Co-etiquetado de DCX junto con NeuN, que se expresa en las neuronas postmitóticas, permite la diferenciación de immature (DCX +, NeuN +) y maduros (DCX -, NeuN +) neuronas granulares. Los marcadores mencionados anteriormente se utilizan con frecuencia para inmunofluorescencia co-etiquetado y microscopía confocal posterior para analizar el número y la identidad de las células recién nacidos. Esto requiere típicamente anticuerpos de diferentes especies huésped para evitar anticuerpo no deseado reactividad cruzada. Además, algunos antígenos son susceptibles a la formalina enmascaramiento epítopo inducida por la fijación (por ejemplo, Ki67, Nestina). Esta revisión se describen las modificaciones de los protocolos immunolabeling múltiple y simple clásicos (por ejemplo, la recuperación de epítopo, inmunotinción secuencial múltiple, uso de nestin-GFP ratones transgénicos 12) que superar muchas de estas cuestiones. En particular, el protocolo de inmunofluorescencia secuencial múltiple permite tinción contra un máximo de cuatro antígenos diferentes, incluso si una parte de los anticuerpos se deriva de la misma máquina. immunohistochemistry and multiple labeling with antibodies from the same host species to study adult hippocampal neurogenesis

Neurogénesis adulta es un proceso multi-etapa altamente regulado en el que las nuevas neuronas se generan a partir de una célula madre neural activado a través de cada vez más comprometida subtipos progenitoras intermedias. Cada uno de estos subtipos expresa un conjunto de marcadores moleculares específicos que, junto con los criterios morfológicos específicos, pueden ser utilizados para su identificación. Por lo general, las técnicas de inmunofluorescencia se aplican implica anticuerpos específicos de subtipo en combinación con marcadores de proliferación exo o endógenos. Nosotros describimos en la presente memoria métodos para la detección y cuantificación de todas las etapas de neurogénesis en el hipocampo adulto immunolabeling. Estos comprenden la aplicación de los análogos de timidina, perfusión transcardial, procesamiento de tejido, de recuperación de epítopo inducida por calor, ABC de inmunohistoquímica, inmunofluorescencia indirecta múltiple, microscopía confocal y cuantificación de células. Además se presenta un protocolo de inmunofluorescencia secuencial múltiple que evita problemas usually que surge de la necesidad de la utilización de anticuerpos primarios planteados en la misma especie hospedadora. Se permite una identificación precisa de todos los subtipos progenitoras del hipocampo, junto con un marcador de proliferación dentro de una sola sección. Estas técnicas son una poderosa herramienta para estudiar la regulación de los diferentes subtipos progenitoras en paralelo, su implicación en patologías cerebrales y su papel en las funciones específicas del cerebro.

Se aplicaron los métodos descritos anteriormente para cuantificar y caracterizar las células recién nacidas en el hipocampo postnatal y adulto. Por lo tanto, hemos utilizado de tipo salvaje y la neurogénesis deficiente ciclina D2 knock out (KO) CCND2 ratones alojados en condiciones conocidas por afectar la tasa de neurogénesis (es decir, el medio ambiente enriquecido, EE) 13,14.
