Entwicklung von Thymozyten machen etwa 98% des Thymus, die restlichen 2% bestehen aus einer Vielzahl von Zellen, die zusammen bilden den Thymus-Stroma (dh Epithelzellen, dendritischen Zellen, Makrophagen, B-Zellen, Fibroblasten, Endothelzellen). Die äußeren kortikalen Epithelzellen (CTEC) beschaffen Einwanderung von Pro-T-Zellen aus dem Knochenmark, T-Zell-Linie Induktion in multi pre-T-Zellen und positive Selektion von selbst MHC beschränkt unreifen Thymozyten. Die inneren Mark Thymusepithelzellen (mTEZ) in Toleranzinduktion dieser Thymozyten mit einer hochaffinen TCR Selbst Peptid / MHC-Komplexen durch entweder induzierende negative Selektion oder deren Abweichung in den regulatorischen T-Zelllinie beteiligt. Im Zusammenhang mit der zentralen Toleranzinduktion sind mTEZ, daß sie ein breites Spektrum von gewebe beschränkt Selbst-Antigene (TAA), die so Spiegeln des peripheren selbst exprimieren einzigartig. Dieses Phänomen wird als Promiscuous Genexpression (pGE)1,2. Die meisten aktuellen Studien zu diesem faszinierenden Zelltyp verlassen sich auf ex vivo isolierten Zellen, wie verschiedene kurzfristige 2D-Kultursystemen unweigerlich zum Verlust von PGE und Schlüsselregulator Moleküle wie MHC-Klasse II, FOXN1 und Aire in den ersten 2 Tagen 3-6 geführt . Es blieb jedoch unklar, welche bestimmte Komponenten und Funktionen des intakten 3D Geflecht des Thymus in 2D-Modelle fehlt. Die Reaggregation Thymus-Organkultur (RTOC) bisher das einzige 3D-System, das die Untersuchung der T-Zell-Entwicklung ermöglicht, einerseits und stromal cell biology, auf der anderen Seite war, in einer intakten Thymus Mikro 7. Dennoch haben RTOCs bestimmte Beschränkungen, das heißt sie enthalten bereits eine komplexe Mischung von Zellen, erfordern die Eingabe von fötalen Stromazellen und ertragen einer maximalen Kultivierungsdauer von 5 bis 10 Tagen. Der Mangel an reduktionistischen in vitro Kultursystemen hat das Studium behindertmehrere Aspekte der T-Zell-Entwicklung und Thymus Organogenese nicht zuletzt die molekulare Regulation von PGE und seiner Beziehung zu der Entwicklungsbiologie mTEZ. Durch die enge Bezogenheit der strukturierten Organisation der Epithelzellen der Haut und der Thymus, entschieden wir uns für ein 3D organotypische Kultur (OTC) System, das ursprünglich entwickelt worden war, um die Differenzierung von Keratinozyten in vitro nachzuahmen und so eine dermale Äquivalent. Die OTC-System besteht aus einem inerten Gerüstmatrix mit dermalen Fibroblasten, die in einem Fibrin-Gel, auf die Keratinozyten ausgesät 8,9 gefangen sind überlagert. Hier ersetzt wir Keratinozyten mit gereinigtem mTEZ. Und dabei die grundlegenden Merkmale dieses Modells optimierten wir bestimmte Parameter. In der angenommenen OTC Modell mTEZ vermehrt unterzog terminalen Differenzierung und gepflegt mTEC Identität und PGE, somit eng in vivo mTEZ Entwicklung nachahmt <sup&gt; 10 ist. Dieses Datenblatt ein detailliertes Protokoll, das die schrittweise Einrichtung von Thymus OTCs.

Intra-Thymus-T-Zell-Entwicklung erfordert ein kompliziertes dreidimensionales Maschenwerk aus verschiedenen Stromazellen, dh Nicht-T-Zellen. Thymocyten durchqueren dieses Gerüst in einem hoch koordinierten zeitliche und räumliche Ordnung, während sequentiell vorbei obligatorischen Kontrollpunkte, dh T-Zelllinie Engagement, gefolgt von T-Zell-Rezeptor-Repertoire und Auswahl vor der Ausfuhr in die Peripherie. Die beiden wichtigsten ansässigen Zelltypen bilden dieses Gerüst sind kortikalen (CTEC) und Mark Thymusepithelzellen (mTEZ). Ein Schlüsselmerkmal der mTEZ ist die sogenannte Promiscuous Expression zahlreicher gewebe beschränkten Antigenen. Diese gewebe beschränkten Antigenen präsentiert werden, um Thymozyten direkt oder indirekt durch unreife mTEZ oder Thymus-dendritische Zellen, die jeweils resultierenden Selbsttoleranz. Geeignete in vitro Modelle emuliert die Entwicklungswege und Funktionen der CTEC und mTEZ liegen noch lacKönig. Dieser Mangel an angemessenen experimentellen Modellen hat zum Beispiel beeinträchtigt die Analyse der Promiscuous Genexpression, die immer noch schlecht auf zellulärer und molekularer Ebene verstanden wird. Wir angepasst 3D organotypischen Co-Kultur-Modell zur Kultur ex vivo isolierten mTEZ. Dieses Modell wurde ursprünglich entwickelt, um Keratinozyten derart zu kultivieren, um eine Hautäquivalents in vitro zu erzeugen. Das 3D-Modell beibehalten wichtigsten Funktionsmerkmale mTEC Biologie: (i) die Proliferation und terminale Differenzierung von CD80 lo, Aire-negative in CD80 hallo, Aire-positive mTEZ, (ii) Reaktionsfähigkeit auf RANKL, und (iii) nachhaltige Ausdruck FOXN1, Aire und gewebe eingeschränkt Gene in CD80 hallo mTEZ.

Wir nahm eine 3D organotypischen Co-Kultur-Modell (3D OTC), die ursprünglich für die in-vitro-Langzeitkultur von Keratinozyten 9 entwickelt hatte. MACS-angereicherten mTEZ (siehe MACS Anreicherung Schema Abbildung 1) wurden auf ein Gerüst aus einer Fibrin-Gel und eingeschlossene Fibroblasten ausgesät. Studying medullary thymic epithelial cells in vitro has been largely unsuccessful, as current 2D culture systems do not mimic the in vivo scenario. The 3D culture system described herein - a modified skin organotypic culture model - has proven superior in recapitulating mTEC proliferation, differentiation and maintenance of promiscuous gene expression.
