Als een monoklonaal muis primair antilichaam - en dus een anti-muis secundair antilichaam - wordt gebruikt, wordt een tweede blokkerende stap endogene muis immunoglobulinen omvatten die noodzakelijk (stap 3.4) om niet-specifieke achtergrondsignaal te verlagen. Gepaste microscopie en beeldverwerking technieken zijn cruciaal voor het verkrijgen van biologisch accurate informatie 21. Het histogram toont de verdeling van pixels op elk intensiteitsniveau (0-65.535 niveaus beeld een 16-bit) voor elke kleur. Achtergrond, kan de helderheid en het contrast worden aangepast door het instellen van een intensiteit weergave bereik dat het histogram piek flankeert; zinvolle vergelijkingen tussen de condities te maken, moeten dezelfde instellingen tussen controle en experimentele omstandigheden worden gebruikt. Dit protocol voorziet in een betrouwbare methode om cardiomyocyte rijping en ontwikkeling te analyseren in de moedertaal embryonale muis hart. Terwijl immunofluorescentie cardiomyocyt-eiwitten wordt vaak gebruikt om hartspiercellen tijdens de ontwikkeling markeren weinig studies met technieken die hoge-resolutie analyse van myofibril structuur en de opkomst geïntercaleerde schijven en costameres 12-14,22,23 mogelijk. Deze techniek kan worden gebruikt voor in vivo beoordeling van mutaties die ontwikkelingsstoornissen hartafwijkingen veroorzaken, als een middel ter identificatie van veranderingen in cardiomyocyte rijping die licht kunnen werpen op de mechanismen van structurele afwijkingen.

Tijdens de ontwikkeling, hart weeën beginnen kort na hartspiercellen migreren naar de middellijn en vormen de lineaire hart buis 1,2. De sarcomeer is de fundamentele contractiele eenheid binnen de cardiomyocyte; deze zeer georganiseerde cytoskeletstructuur bevat actine filamenten verankerd aan de Z-disc sarcomeer α-actinine (s-α-actinine) en myosine vezels verankerd aan de M-lijn (figuur 1). Zoals de cardiomyocyte rijpt, sarcomeres assembleren in serie te myofibrils die zich uitstrekken over de cel te vormen. Myofibrillen zijn verankerd aan de einden van de cardiomyocyten van de geïntercaleerde schijf, de cel-cel junctional structuur die een over- kruising met een deelverzameling van Z schijfvormige elementen zoals s-α-actinine 3, adherens bevat junction eiwitten zoals N-cadherine en β catenine, gap junction eiwitten en desmosomen (figuur 1) 4. Langs de lengterichting membraan, de Z-schijven van perifere myofibrillen ookhechten aan het celmembraan via costameres; deze gespecialiseerde focale adhesies vormen een anker tussen de myofibril, plasmamembraan en extracellulaire matrix aanvullende structurele steun aan de cardiomyocyten voorzien (figuur 1) 4. Vroeg in de ontwikkeling van het hart, zijn hartspiercellen gerangschikt in vinger-achtige projecties bekend als trabeculae die uitsteken in de ventriculaire ruimte en bevatten relatief volwassen myofibrils 5. Zoals ontwikkeling van het hart opbrengst, de hartspiercellen in de sub-epicardial regio woekeren met de compacte myocard dat de ventriculaire muren bestaat, maar sarcomeer en myofibril assemblage zijn vertraagd in vergelijking met trabeculair myocard 5,6 te vormen. Modellen van sarcomeer en myofibril samenstel zijn grotendeels het gevolg immunofluorescentie studies op gekweekte cardiomyocyten 7-10, die eenvoudig zijn maar missen een driedimensionale omgeving, de bloedstroom, en contacten met andere hartcels aanwezig in vivo. Hoge-resolutie structurele studies met behulp van immunofluorescentie in de muis embryonale hart zijn technisch uitdagend, en weinig studies hebben de opkomst van geïntercaleerd schijven en costameres tijdens muis ontwikkeling van het hart onderzocht. De adherens junction eiwit β catenine lijkt te lokaliseren om geintercaleerde schijven van embryonale dag (E) 17,5 11, N-cadherine lokaliseert om lineaire structuren die geïntercaleerd schijven door E18.5 12 versus postnatale dag 0 13 kan vertegenwoordigen, en costameres zijn geconstateerd E18.5 14, maar deze eiwitten weer te diffuus en meer continue membraan distributie in eerdere ontwikkelingstijd punten 11-13. We beschrijven hier een eenvoudige en reproduceerbare werkwijze voor immunokleuring en fluorescentie microscopie van doorsnede muis embryonale hart dat zorgt voor gedetailleerde analyse van myofibril en cardiomyocyten ontwikkeling, waaronder de opkomst van intercalated discs zo vroeg E12.5 en ontluikende costameres op E16.5. Dit protocol kan nuttig zijn voor het sonderen van de effecten van de mutaties op de sarcomeer vorming evenals myofibril en cardiomyocyte rijping zijn.

analysis of cardiomyocyte development using immunofluorescence in embryonic mouse heart

Tijdens de ontwikkeling van het hart, de generatie van myocardiale specifieke structurele en functionele eenheden zoals sarcomeres, contractiele myofibrils, tussengevoegde schijven en costameres vereist de gecoördineerde assemblage van meerdere componenten in tijd en ruimte. Verstoring in het installeren van deze componenten leidt tot ontwikkelingsstoornissen hartafwijkingen. Immunofluorescentiekleuring technieken worden algemeen gebruikt in gekweekte cardiomyocyten te myofibril rijping sonde, maar ex vivo benadering wordt beperkt door de mate waarin myocyten volledig differentiëren in cultuur ontbreken van normale in vivo mechanische input, en afwezigheid van endocardiale signalen. Toepassing van immunofluorescentie technieken voor de studie van ontwikkeling muizenhart is wenselijk maar technisch uitdagend en methoden vaak niet voldoende gevoeligheid en resolutie te sarcomeren zichtbaar in de vroege stadia van ontwikkeling van het hart. Hier beschrijven we een robuuste en reproduceerbare methode samen immunostain multiple eiwitten of om samen te visualiseren een fluorescerend eiwit met immunofluorescentiekleuring in de embryonale muis hart en gebruiken deze methode te ontwikkelen myofibrils, geïntercaleerd schijven, en costameres analyseren. Deze werkwijze kan verder worden toegepast cardiomyocyten structurele veranderingen veroorzaakt door mutaties die leiden tot ontwikkelingsstoornissen hartafwijkingen beoordelen.

De figuren 2 tot en met 6 tonen typische resultaten voor co-kleuring van verschillende eiwitten in een snap-bevroren en aceton gefixeerde hart. Het antilichaam tegen B-α-actinine reproduceerbaar gemerkte Z-schijven en geïntercaleerde schijven met hoge specificiteit en minimale achtergrond (Figuren 2A, 3A, 4A, 5A, 6A en 6C) Figuur 6 toont dat de anti-muis IgG (H + L) monovalente Fab fragment blokkeert effectief de endogene muis IgG binding van anti-muis secundaire antilichamen. Het antilichaam tegen adherens junction eiwit β catenine gebonden het membraan zowel cardiomyocyten als niet-cardiomyocyt cellen en co-lokalisatie met s-α-actinine opgetreden bij veronderstelde tussengevoegde schijven op E16.5 (figuur 2C en D), zoals verwacht van de β catenine kleuringspatroon in het volwassen hart 18. β1 integrine immunofluorescentie in het embryonale hart is bijzonder uitdagend en vaak niet in slaagt om focale adhesies 14 te identificeren, maar β1 integrine kleuring in deze studies is gebleken signaal met dezelfde frequentie als s-α-actinine label Z-schijven, mogelijk als gevolg van ontluikende costameres vormen op E16.5 (Figuur 3D). Bij E12.5, s-α-actinine en tropomyosine (sarcomeer dunne filament eiwit) immunofluorescentie bleek een kleuringspatroon met regelmatige periodiciteit in trabeculaire hartspiercellen consistent met volwassen myofibrils in deze cellen (Figuren 4A en 5A voor s-α-actinine; Figuur 4B voor tropomyosine). N-cadherine kleuring trabeculaire cardiomyocyten op E12.5 harten verzorgen colocaliseren met gebieden van intense s-α-actinine kleuring (figuur 5B - D en figuur 6A - C) eventueel voor tussengevoegde schijven. Incontrast aan trabeculair myocytes, s-α-actinine in de compacte zone was meer punctate dan lineair, en tropomyosine kleuring was diffuus plaats van lineair (Figuur 4A en 4B). Aldus kan sarcomeer assemblage later in compact voorkomen opzichte trabeculaire myocardium. Bovendien differentiële patronen van s-α-actinine en tropomyosine in de compacte zone suggereren dat s-α-actinine organiseert in puncta en onvolwassen Z-discs vroeg, terwijl tropomyosine opname in de dunne gloeidraad een later gebeurtenis myofibril samenstel kan zijn. Figuur 7, Film 1 en Film 2 tonen typische resultaten van een PFA-vaste E12.5 embryonale hart. In deze voorbeelden werd een LifeAct-RFPruby transgeen embryo voor beeldvorming; de LifeAct-RFPruby transgen 19 etiketten draadvormige actine maar vereist PFA fixatie. Z-schijven gelabeld met s-α-actinine waren gemakkelijk te visualiseren in de meeste gebieden, maar de signaal-ruisverhouding Decre<html> ased vergelijking met ingevroren hart secties (figuur 7A); Dit signaal was typerend voor s-α-actinine immunofluorescentie in PFA-gefixeerd weefsel, waarin epitopen kunnen worden gemaskeerd door eiwit cross-links. Figuur 7B toont co-visualisatie van draadvormige actine en immunolabeled s-α-actinine binnen myofibrils (pijlpunten) en filamenteuze actine binnen endocardiale cellen naast trabeculaire myocyten (pijlen). Drie-dimensionale beeldreconstructie onthulde aanvullende gegevens: individuele cardiomyocyten werden gemakkelijker onderscheiden, myofibrils binnen cardiomyocyten werden ongeveer parallel aan elkaar, maar individuele cardiomyocyten werden georiënteerd onder verschillende hoeken met elkaar (Figuur 7C en D, film 1 en film 2 ).
