De schaalbaarheid van de 3&#39;LIFE assay maakt het screenen van een grote bibliotheek 3&#39;UTR voor het richten van een bepaalde miRNA zonder voorspannen van het scherm richting bioinformatically geïdentificeerde genen. detection of mirna targets in high-throughput using the 3'life assay

Lichtgevende Identificatie van functionele elementen in 3&#39;UTRs (3&#39;LIFE) kan de snelle identificatie van doelwitten van specifieke miRNAs in een reeks van honderden bevraagd 3&#39;UTRs. Target identificatie is gebaseerd op de dual-luciferase assay, die detecteert binding bij het mRNA niveau door meting translationele uitgang, die een functionele uitlezing van miRNA richten. 3&#39;LIFE maakt gebruik van niet-merkgebonden buffers en reagentia, en publiekelijk beschikbare verslaggever bibliotheken, waardoor genoom-brede schermen haalbaar en rendabel. 3&#39;LIFE kan worden uitgevoerd in een standaard lab instelling of opgeschaald met behulp van vloeibare handling robots en andere high-throughput instrumentatie. We illustreren de benadering met behulp van een dataset van menselijke 3&#39;UTRs gekloond in 96-well platen en twee test-miRNAs, laat-7c en miR-10b. We demonstreren hoe DNA bereiding, transfectie, celcultuur en luciferase assays in 96 putjes format uit te voeren en zorgen voor gegevensanalyseanalyse. Tot slot 3&#39;LIFE is zeer reproduceerbare, snelle, systematische, en identificeert groot vertrouwen doelen.

De luminometer output bestand bevat ruwe metingen voor zowel vuurvlieg en Renilla luciferase eiwitten.
