Luminiscentes Identificación de elementos funcionales en 3&#39;UTRs (3&#39;LIFE) permite la rápida identificación de los objetivos de miRNAs específicos dentro de una matriz de cientos de 3&#39;UTRs consultados. La identificación del objetivo se basa en el ensayo de luciferasa dual, que detecta la unión a nivel de mRNA mediante la medición de la salida de la traducción, dando una lectura funcional de los genes miARN focalización. 3&#39;LIFE utiliza buffers no propietarios y reactivos, y bibliotecas reportero públicamente disponibles, por lo que las pantallas de todo el genoma viable y rentable. 3&#39;LIFE se puede realizar en un laboratorio estándar o ampliarse el uso de robots de manipulación de líquidos y otros instrumentos de alto rendimiento. Se ilustra el método utilizando un conjunto de datos de 3&#39;UTRs humanos clonados en placas de 96 pocillos, y dos miRNAs prueba, vamos-7c y miR-10b. Demostramos cómo realizar la preparación de ADN, transfección, cultivo celular y ensayos de luciferasa en formato de 96 pocillos, y proporcionar herramientas para datosanálisis. En conclusión 3&#39;LIFE es altamente reproducible, rápido, sistemático, e identifica objetivos de alta confianza.
