electroretinogram analysis of the visual response in zebrafish larvae

De electroretinogram (ERG) is een niet-invasieve elektrofysiologische methode voor het bepalen van retinale functie. Door het plaatsen van een elektrode op het oppervlak van de cornea, elektrische activiteit gegenereerd in reactie op licht kan worden gemeten en gebruikt om de activiteit van retinale cellen in vivo te evalueren. Dit manuscript beschrijft het gebruik van de ERG om de visuele functie te meten in de zebravis. Zebravis zijn al lange tijd gebruikt als een model voor gewervelde ontwikkeling als gevolg van het gemak van het gen onderdrukking door morfolino- oligonucleotiden en farmacologische manipulatie. 5-10 dpf, alleen kegels zijn functioneel in de larvale retina. Daarom is de zebravis, in tegenstelling tot andere dieren, is een krachtig model voor de studie van kegel visuele functie in vivo. Dit protocol maakt gebruik van standaard anesthesie, micromanipulatie en stereomicroscopie protocollen die gangbaar zijn in de laboratoria die de zebravis onderzoek uit te voeren. De geschetste methoden maken gebruik van standaard elektrofysiologie equipment en een weinig licht camera om de plaatsing van de opname micro-elektrode te begeleiden op de larvale hoornvlies. Tenslotte tonen we hoe een commercieel verkrijgbaar ERG stimulator / recorder oorspronkelijk ontworpen voor gebruik met muizen gemakkelijk worden aangepast voor gebruik bij zebravis. ERG van larvale zebravis biedt een uitstekende methode voor het bepalen van kegel visuele functie bij dieren die zijn gewijzigd door morfolino- oligonucleotide injectie, alsook nieuwere genoom engineering-technieken zoals Zinc Finger Nucleasen (ZFNs), transcriptie Activator-Like Effectorcellen Nucleasen (Talens), en Regelmatig Gegroepeerde afgewisseld met korte Palindromic Repeats (CRISPR) / Cas9, die allemaal sterk verhoogde de efficiëntie en effectiviteit van gen targeting in zebravis. Bovendien benutten we het vermogen van farmacologische middelen aan zebravis larven de moleculaire componenten die bijdragen aan de photoresponse evalueren dringen. Dit protocol beschrijft een opstelling die kan worden gemodificeerd en gebruikt door onderzoekersmet diverse experimentele doelen.

Gewoonlijk ERG worden geregistreerd van zebravis larven 5 dpf, aangezien een aantal studies ERG opnames gepubliceerd in dit stadium 9, 16,20. Larvale reacties werden gemeten onder donker aangepaste voorwaarden zonder achtergrond verlichting met behulp van een 20 msec stimulans van wit LED licht.
