Wij hebben echter gevonden dat de LM LEC sneller prolifereren en zijn beter bestand tegen apoptosis dan voorhuid LEC 15 suggereert dat er een primair defect in de LM LEC&#39;s. Bij LM LEC worden geïmplanteerd in een muis xenograft model, vormen ze structuren denken aan lymfatische misvormingen 15. Dit ondersteunt de hypothese dat lymfatische misvormingen kunnen worden veroorzaakt door een of meer somatische mutaties ontstaan ​​in LM LEC tijdens de foetale ontwikkeling. Inderdaad hebben recente rapporten zo&#39;n mutatie in het p110α katalytische subeenheid van fosfoinositide 3-kinase (PIK3CA) genen 20 geïdentificeerd. Gezien de vorderingen in DNA sequencing technologie, relevante mutaties cOuld gemakkelijker worden geïdentificeerd in geïsoleerde LM LEC&#39;s, het begeleiden van toekomstige studies van deze voorwaarden. De isolatie van levensvatbare LEC zou vergelijkingen tussen abnormale en normale LECs in assays zoals migratie, proliferatie, buis die vermogen en overleving in respons op verminderde beschikbaarheid van voedingsstoffen of pro-apoptotische middelen 15 te vergemakkelijken. Geïsoleerde LEC zou ons in staat te stellen cel-specifieke genexpressie en proteomics studies uitgevoerd om nieuwe LEC subpopulaties bakenen en ontdekken van nieuwe farmacologische middelen die geschikt zijn voor klinische behandeling van lymfatische misvormingen. We hebben eerder publiceerde een LEC isolatie methode gebaseerd op magnetische kraal scheiding van LEC&#39;s van neonatale voorhuid en lymfatische misvormingen 15. We meldden een strategie voor het scheiden normale en zieke LEC&#39;s van vasculaire endotheliale cellen op basis van de afwezigheid van CD34 expressie, gevolgd door het onderwerpen CD34 Neg celfractie positieve selectie van CD31.Echter, deze werkwijze belemmerd door de aanwezigheid van achtergebleven niet-endotheelcellen. Dit was onafhankelijk van de epidermis te verwijderen vóór verder bindweefsel digestie. Deze verontreinigingen algemeen prolifereerden sneller en dus uiteindelijk overgrew de endotheliale celculturen ondanks latere pogingen om LEC isolatie herhalen. De rationale voor de selectie van deze merkers werd gebaseerd op de rapporten die tijdens LEC en bloed vasculaire endotheliale cells hebben veel celoppervlaktemerkers gemeen, zoals CD31, LEC&#39;s tonen fenotypische variatie in hun expressie van CD34, podoplanin en VEGFR-3 celoppervlak marker in vergelijking met bloed vasculaire endotheliale cellen 21-23. CD31 is een 130 kDa transmembraan glycoproteïne ook bekend als bloedplaatjes endothelial cell adhesion molecule 1 (PECAM-1). Het wordt beschouwd als een pan-endotheelcel merker omdat het tot expressie wordt gebracht op alle soorten bloed- en lymfevaten 21,24,25. CD34 is 110 kDa transmembraan glycoproteïne aanwezig op de meeste hematopoietische moedercellen en stamcellen, vasculaire endotheelcellen en enkele lymfevaten 26. VEGFR-3, de receptor voor vasculaire endotheliale groeifactoren C en D, aanvankelijk aanwezig op de ontwikkelende aders in het muizenembryo, maar na lymfatische specificatie gereguleerd door de transcriptiefactoren SRY-gerelateerde HMG-box (SOX) -18, c hicken o valbumin u pstream p romoter t ranscription f actor 2 (COUPTF-II) en PROX-1, is VEGFR-3 veneuze expressie verloren en het wordt beperkt tot embryonale LECs 25,27. Podoplanin, een 38 kDa membraan mucoprotein, wordt eerst opgemerkt op de lymfevaten op ongeveer embryonale dag 11 (~ E11.0) van de muis embryonale ontwikkeling 28 en terwijl het sterk wordt uitgedrukt door microvasculaire lymfevaten, podoplanin expressie door macrocystic lymfatische endotheel in lymfatische misvormingen meer variabel 15. Flowcytometrie experimenten suggereren dat ten minste enkele CD34 Hoge CD31 Pos endotheelcellen drukken de lymfatische marker podoplanin 29. Hoewel systematische evaluatie van LYVE-1 en podoplanin kleuring in humane lymfatische malformaties gebleken dat beide effectief zijn bij het ​​kleuren lymfatisch endothelium misvorming 30, in normale weefsels, LYVE-1 werd gemeld sterk aanwezig in de initiële lymfatische zijncapillaire endotheel maar verminderd en zelfs afwezig bij het ​​verzamelen van lymfatische endotheel 31. Omdat ons doel is om te isoleren zowel de initiële en het verzamelen van lymfatische endotheelcellen we hebben ervoor gekozen niet te LYVE-1 gebruiken als onderdeel van onze mobiele strategie selectie. Tenslotte werd de beslissing om deze markers ook gebruikmaken basis van de beschikbaarheid van antilichamen die diagnostisch zijn voor lymfevaten labelen voor microscopische beeldvorming, een eigenschap die verband tussen flowcytometrie en immunofluorescentie studies zou toestaan. Lymfestelsel aandoeningen zoals primaire lymfoedeem, lymfatische misvormingen en lymfatische tumoren zijn zeldzame omstandigheden die aanzienlijke morbiditeit veroorzaken, maar er is weinig bekend over hun biologie. Het isoleren van zeer zuiver menselijke lymfatische endotheelcellen (LEC&#39;s) van zieke en gezonde weefsel zou studies van het lymfatische endotheel bij genetische, moleculaire en cellulaire niveau te vergemakkelijken. Verwacht wordt dat deze onderzoeken targets kunnen openbaren nieuwe therapieën die de klinische behandeling van deze aandoeningen kunnen veranderen. Een protocol beschrijft de isolatie van menselijke voorhuid LEC&#39;s en lymfatische misvorming lymfatische endotheelcellen (LM LEC&#39;s) wordt gepresenteerd. Voor het verkrijgen van een enkele celsuspensie weefsel werd fijngehakt en enzymatisch behandeld met collagenase en dispase II II.
