Siden ubiquitinering er en grunnleggende celleaktivitet, kunne forstå de regulatoriske mekanismer være nøkkelen til å undergrave prosesser av sykdom og patologi. Den ubiquitin-proteasom system (UPS) fungerer som en av de viktigste nedbrytningsmekanismer i pattedyrcelle. Underlag bundet for nedbrytning i proteasome er kovalent merket med polymerer av ubiquitin (Ub) 1. Før den måls substratet kommer inn i proteasome for nedbrytning, må poly-ubiquitin merkelappen fjernes. En klasse av enzymer som kalles deubiquitinating enzymer (Dubs) er ansvarlig for fjerning og gjenvinning av ubiquitin molekyler to. Det har blitt spådd fra det menneskelige genom at det er nesten hundre Dubs arbeider i cellen tre. Med et så stort antall Dubs kontrollerende Ub-mediert cellulære prosesser, studerer disse enzymene presenterer en utfordring siden mRNA teknikker ikke gir informasjon om aktivitet og western blotting bare gir informasjon om uttrykket nivåer. Bruken av influensa hemagglutinin (HA) merket, Ub-avledet aktive sete-rettet sonder gjør det mulig for en kovalent modreduser fuktighet av de funksjonelle Dubs og derfor gir en direkte visualisering av aktiviteten til disse enzymene på en Western blot-4. Sondene har en C-terminal tiol-reaktiv gruppe som fungerer som et substrat for selvmord det aktive sete cystein-rest 5. Med disse sonder, er det mulig å studere aktiviteten og potensialet ekspresjon av mange Dubs under begge patologiske og fysiologiske tilstander av cellen. Endringer i DUB-aktivitet har vært implisert i en rekke patologiske tilstander, slik som Parkinsons, Alzheimers, anemi og ulike kreftformer 6-10. Denne teknikken gir et kraftig verktøy for studier av sykdommen. I det foreliggende papir, viser vi anvendelsen av denne teknikk i HeLa og M17-celler som er lysert ved hjelp av glassperler. I tillegg vi skissere hvordan du bruker denne metoden i museryggmargs vevsprøver. Informasjon fra denne teknikken kan brukes som et utgangspunkt for identifiseringterapeutiske mål samt etablere modeller for studiet av ulike sykdomstilstander. Den sanne nytten av denne teknikken ligger i dens evne til å gi informasjon om flere Dubs i en enkelt analyse.

Den ubiquitin-proteasom-system er nylig blitt implisert i forskjellige sykdomstilstander, inkludert neurodegenerative sykdommer og kreft. I lys av dette, teknikker for å studere reguleringsmekanisme av dette systemet er avgjørende for å belyse de cellulære og molekylære prosesser i de nevnte sykdommene.
