Deze singlet zuurstof radicalen kan worden gebruikt voor foto-oxideren diaminobenzidine polymeren aan en nabij het ​​oppervlak van de gelabelde eiwitten, die aanzienlijk dichter dan immunogoud gemerkte antilichamen 11 te vormen. Veel eiwitten die betrokken zijn bij DNA dubbele breuken reparatie ophopen in brandpunten die monteren rond een DNA dubbele streng te breken 12-14. Hoewel hun functie niet bekend is, vertegenwoordigen zij de plaats in de kern die de DNA dubbele streng breuk bevat en de plaats van DNA dubbele streng breuk reparatie. DNA-reparatie foci (DRF) zijn gekarakteriseerd door fluorescentiemicroscopie en zij dienen als biomarkers voor DNA schade 12,15. Ze zijn enorm ten opzichte van de grootte van het dubbelstrengs breuken en waren betrekkelijk homogeen beschouwd zolang Language superresolutietechnieken studies toonden aanwijzingen van sub-compartimentering van moleculen binnen iedere 16 gericht. Om te begrijpen hoe deze plaatsen zijn georganiseerd, dient te visualiseren alle onderliggende biologische structuren ten opzichte van elkaar. Dit kan niet worden bereikt door fluorescentiemicroscopie maar kan door elektronenmicroscopie 17,18. Hier wordt een methode beschreven die elektronen spectroscopische beeldvorming combineert met de miniSOG methode om het potentieel van deze gecombineerde aanpak illustreren aan de ultrastructuur van DNA double-strand break repair verkennen.

De grenzen van optische resolutie en de uitdaging van het identificeren van specifieke eiwit bevolkingsgroepen in transmissie elektronenmicroscopie zijn obstakels in de celbiologie geweest. Veel verschijnselen kunnen niet verklaard worden door in vitro analyse in vereenvoudigde systemen en aanvullende structurele informatie nodig in situ, in het bijzonder in het traject tussen 1 nm en 0,1 urn, teneinde te worden begrepen. Hier, elektronen spectroscopische beeldvorming, een transmissie elektronenmicroscopie techniek die gelijktijdig in kaart brengen van de verdeling van eiwitten en nucleïnezuren mogelijk maakt, en een uitdrukking tag, miniSOG, worden gecombineerd om de structuur en organisatie van DNA double-strand break repair foci bestuderen.

Electron spectroscopic imaging can image and distinguish nucleic acid from protein at nanometer resolution. It can be combined with the miniSOG system, which is able to specifically label tagged proteins in transmission electron microscopy samples.
