Трансплантация сердца модель часто используется для исследования I / R-травмы, отторжения трансплантата, после трансплантации васкулопатии и эффективность иммуномодулирующих агентов. Мышиные модели сочетают много преимуществ, как известный иммунологических и генетических фонов, наличие многих инбредных и трансгенных линий и относительно низких затрат экспериментальных. В 1973 году методика брюшной трансплантации сердца у мышей была впервые описана Corry и др 1. В этой модели трансплантаты реваскуляризированных по конец в бок анастомоза восходящей аорты в брюшной аорты получателя и главного легочной артерии в нижнюю полую вену. В 1991 году методика шов шейки трансплантации сердца у мышей была описана Чен и др 2. В этой модели ретроградной перфузии коронарных артерий устанавливается анастомоза сонную артерию получателя, чтобы восходящей аорты ГРКормовая. В венозных стоков крови через коронарный синус в правое предсердие, правый желудочек и выбрасывается через легочную артерию в получателя внешнего яремной вены (рис 1). По сравнению с брюшной трансплантата имплантации шейки модель является менее инвазивным и поверхностное расположение трансплантата позволяет очень легкий доступ для дополнительных последующих испытаний (например, пальпации, эхокардиография, прижизненный флуоресцентной микроскопии) 3,4. Несмотря на технические усовершенствования 5,6, более широкое использование этой модели было ограничено из-за технических трудностей, чтобы выполнить пришитых анастомозов мелких сосудов с высокой частотой утечки анастомоза и тромбоза. В 1991 году Мацуура и др. представил методику манжеты, в котором стенка сосуда выворачивают через синтетического цилиндра, что значительно облегчает модель трансплантации сердца шейки 7. Техника была фуrther принят на различных экспериментальных моделях трансплантации почек в том числе 8, поджелудочной железы, 9 конечности 10, печеночная 11,12, легких и сосудистой 13,14 трансплантации 15. По сравнению с методом шовного требуемое время анастомозов и, таким образом, время тепловой ишемии короче и есть значительно меньше, сосудистых осложнений с использованием метода манжеты 16. Кроме того, она позволяет хирургам с небольшим обучения в микрохирургии, чтобы выполнить операцию с высокой скоростью успеха. heterotopic cervical heart transplantation in mice

The heterotopic cervical heart transplantation in mice is a valuable tool in transplant and cardiovascular research. The cuff technique greatly simplifies this model by avoiding challenging suture anastomoses of small vessels thereby reducing warm ischemia time. In comparison to abdominal graft implantation the cervical model is less invasive and the implanted graft is easily accessible for further follow-up examinations. Anastomoses are performed by pulling the ascending aorta of the graft over the cuff with the recipient&#8217;s common carotid artery and by pulling the main pulmonary artery over the cuff with the external jugular vein. Selection of appropriate cuff size and complete mobilization of the vessels are important for successful revascularization. Ischemia-reperfusion (I/R) injury can be minimized by perfusing the graft with a cardioplegic solution and by hypothermia. In this article, we provide technical details for a simplified and improved cuff technique, which should allow surgeons with basic microsurgical skills to perform the procedure with a high success rate.
