Echter, live cellulaire migratie en organisatie tijdens de organogenese is al lang de hypothese en indirect aangetoond, maar nooit gevisualiseerd. Craniofaciale ontwikkeling vereist complexe moleculaire, cellulaire en weefsel interacties naar cel proliferatie, migratie en differentiatie 1,2, 3 rijden. Dit strak gereguleerd en complex proces is onderworpen aan genetische en omgevingsfactoren storingen, zodat craniofaciale misvormingen behoren tot de meest voorkomende geboorte misvormingen 1-9. Terwijl chirurgische ingrepen blijft de steunpilaar van de behandeling van craniofaciale afwijkingen, het begrijpen van de ontwikkeling basis is essentieel voor toekomstige therapieën te innoveren. Daarom is het bestuderen van de morfogenese en de mechanismen van de convergentie en extensie en cellen integratie verschaft nieuwe inzichten in de vorming van het craniofaciale skelet 1. Craniale neurale lijst migreren en bevolken de eerste faryngeale boog, vormen dan gepaarde onderkaak processen die zich uitstrekken tot de Meckel&#39;s kraakbeen, waarbij de onderkaak voorafschaduwing vormen. Morfogenese of de Meckel&#39;s kraakbeen vereist chondrocyten organisatie via gerichte proliferatie, cel polarisatie en differentiatie 1,10. De ingewikkeldheid van chondrocyte organisatie in de groei en uitbreiding van Meckel kraakbeen blijft onduidelijk. Inzicht in de dynamische cel gedrag is van cruciaal belang voor het begrijpen van aangeboren afwijkingen van invloed zijn onderkaak grootte, zoals hypoplastic onderkaak fenotypes 11. Zebravis embryo&#39;s bieden vele ontwikkelings-en genetische voordelen voor de gedetailleerde studie van Meckel&#39;s kraakbeen morfogenese. Hun genetische traceerbaarheid, transparantie, ex vivo en snelle ontwikkeling zijn krachtige voordelen goed uitlenen voor observatie van de cel beweging en organisatie door het live-imaging 6. Met behulp van lineage tracing hulpmiddelen, zoals sox10: kaede transgene lijn, hebben wij en anderen de neurale oorsprong van de embryonale craniofaciale skelet 1,5 afgebakend. Using de sox10: ERT2-Cre met de Ubi: Zebrabow-M transgene lijn, is het nu mogelijk om de details van cellulaire bewegingen verkennen tijdens craniofaciale ontwikkeling. De Zebrabow-M, is een transgene lijn ontworpen met de ubiquitine promotor die de expressie van verschillende fluoroforen, elk geflankeerd door Lox plaatsen 8. De Zebrabow-M standaard fluorofoor is Rood, uiten RFP. Na inductie van Cre expressie, de Zebrabow-M te construeren recombineert en cellen brengen een combinatie van verschillende fluoroforen (RFP, GVB en YFP) het creëren van multi-spectrale expressie in het embryo. Alle dochtercellen die scheiden van de gemerkte cellen na de recombinatie gebeurtenis worden daarna klonaal gemerkt, zodat celpopulaties die voortkomen uit verschillende naast elkaar geplaatste progenitors klonaal gelabeld. Door deze klonen cel labeling, kunnen cellen proliferatie en migratie klonale resolutie volgen (figuur 1 en 2).

Ontwikkeling van de gewervelde craniofaciale structuren vereist een nauwkeurige afstemming van de cel migratie, proliferatie, adhesie en differentiatie. Patroonvorming van de Meckel&#39;s kraakbeen, een eerste faryngeale boog derivaat, omvat de migratie van craniale neurale (CNC), cellen en de progressieve partitionering, proliferatie en de organisatie van gedifferentieerde chondrocyten. Verschillende studies hebben CNC migratie beschreven in onderkaak morfogenese, maar de details van hoe de chondrocyten organisatie bereiken in de groei en uitbreiding van Meckel kraakbeen blijft onduidelijk. De sox10 beperkte en chemisch geïnduceerde Cre-recombinase gemedieerde recombinatie genereert permutaties van verschillende fluorescerende eiwitten (RFP, YFP en CFP), waardoor een multi-spectrale kenmerken van stamcellen en hun nageslacht, als gevolg van verschillende klonale populaties. Via confocale time-lapse fotografie is het mogelijk om de chondrocyten observeren behaviof tijdens de ontwikkeling van de zebravis Meckel kraakbeen. Multispectrale cel etikettering kunnen wetenschappers uitbreiding van de Meckel&#39;s chondrocyten tonen. Tijdens de uitbreiding fase van het Meckel&#39;s kraakbeen, waarbij de onderkaak voorafschaduwing, chondrocyten intercaleren verlenging effect als ze stapelen in een georganiseerde eencellige gelaagde rij. Mislukking van deze georganiseerde intercalerend proces cel uitbreiding bemiddelen biedt de cellulaire mechanistische verklaring voor hypoplastic onderkaak die we waarnemen in de onderkaak misvormingen.

Traditionele kraakbeen visualisatie door hele berg Alcian blauwe vlekken is van onschatbare waarde in het observeren van de ontwikkeling van Meckel&#39;s kraakbeen en vaak gebruikt voor de uiteindelijke cellulaire organisatie 12 (figuur 1A) visualiseren geweest.
