Desenvolvimento craniofacial requer, interacções celulares e teciduais moleculares complexas para conduzir a proliferação celular, migração e diferenciação 1,2, 3. Este processo bem regulado e complexo está sujeito a perturbações genéticas e ambientais, de tal forma que deformidades craniofaciais estão entre as malformações congénitas mais comuns 1-9. Enquanto intervenções cirúrgicas continuam a ser a base do tratamento para anomalias craniofaciais, compreender a base de desenvolvimento é essencial para inovar futuras terapias. Portanto, estudar a morfogênese e os mecanismos para a convergência e extensão e integração de células proporciona novos insights sobre a formação do esqueleto craniofacial 1. Cranial migração e de crista neural preencher o primeiro arco faríngeo, processos mandibulares então formar pares que se estendem para formar cartilagem de Meckel, o que prefigura a mandíbula. Morfogênese of cartilagem de Meckel exige organização dos condrócitos via proliferação direcional, polarização e diferenciação celular 1,10. Zebrafish embriões oferecem muitas vantagens de desenvolvimento e genéticos para estudo detalhado da cartilagem de Meckel morfogênese. Sua rastreabilidade genética, transparência, ex vivo e rápido desenvolvimento são vantagens poderosas emprestando-lhe bem para observação do movimento celular e organização de imagens ao vivo 6. Usando ferramentas de rastreamento de linhagem, tais como: SOX10 linhagem transgênica kaede, nós e outros delinearam as origens da crista neural do esqueleto craniofacial embrionário 1,5. Using a SOX10: ERT2-Cre com o ubi: linhagem transgênica Zebrabow-M, é agora possível explorar detalhes de movimentos celulares durante o desenvolvimento craniofacial. O Zebrabow-H, é uma linha transgénica desenvolvida com o promotor da ubiquitina que conduz a expressão de fluoróforos diferentes, cada um flanqueado por locais lox 8. O fluoróforo padrão Zebrabow-M é Vermelho, expressando RFP. Após indução da expressão de Cre, o Zebrabow-H construir recombina e células expressam uma combinação de diferentes fluoróforos (RFP, CFP e YFP), criando expressão multi-espectral no embrião. Todas as células filhas que dividem a partir das células marcadas após o evento de recombinação são então clonalmente marcado, de modo que as populações de células que derivam de diferentes progenitoras justapostas são clonalmente marcado. Por esta marcação celular clonagem, células proliferação e migração com resolução clonal pode ser seguido (Figura 1 e 2).

Desenvolvimento das estruturas craniofaciais vertebrados requer uma coordenação precisa de migração celular, a proliferação, a adesão e a diferenciação. Padronização da cartilagem de Meckel, um primeiro arco derivado da faringe, envolve a migração das células da crista neural cranial (CNC) e do particionamento progressiva, proliferação e organização dos condrócitos diferenciados. Vários estudos têm descrito a migração CNC durante inferior morfogênese mandíbula, mas os detalhes de como alcançar os condrócitos organização no crescimento e extensão da cartilagem de Meckel permanece obscuro. O SOX10 restrito e Cre recombinase mediada por recombinação induzida quimicamente gera permutações de proteínas fluorescentes diferentes (RFP, YFP e PCP), criando, assim, um rótulo multi-espectral de células progenitoras e a sua descendência, que reflecte as populações clonais distintos. Usando fotografia confocal time-lapse, é possível observar os condrócitos behaviou durante o desenvolvimento da cartilagem do peixe-zebra de Meckel. Rotulagem célula Multispectral permite aos cientistas demonstrar extensão de condrócitos de Meckel. Durante a fase de extensão da cartilagem de Meckel, o que prefigura a mandíbula, condrócitos intercalar para efetuar extensão como eles se comportam em uma fileira em camadas de uma única célula organizada. A falha deste processo de intercalação organizado para mediar extensão célula fornece a explicação mecanicista celular para mandíbula hipoplásica que observamos em malformações mandibulares.
