Hierin beschrijven we de voorbereiding en uitvoering van intravitale microscopie van mesenterium aderen van muizen in vivo. Deze methode geeft ons de mogelijkheid om leukocyten-endotheel en bloedplaatjes-endotheel interacties 11 direct waar te nemen in het levende organisme. Als een vroege reactie op het endotheel ontsteking wordt geactiveerd en interageert met leukocyten en bloedplaatjes resulteert in rollen, adhesie en transmigratie 12. Maar leukocyten ook in...

Het doel van dit protocol is een vereenvoudigde methode van intravitale microscopie van mesenterium aderen beschrijven levende muizen directe observatie van leukocyt-endotheel leukocyt en bloedplaatjes interacties onder ontstekingsaandoeningen. Intravitale microscopie werd ontwikkeld om leukocyt-endotheel interacties in vivo te bestuderen onder ontstekingsaandoeningen 1,2, wat niet triviaal, maar belangrijk voor het begrijpen van inflammatoire leukocyten rekrutering en functie. De werkwijze beschrijven we in dit protocol is ontwikkeld op basis van eerder gepubliceerde publicaties. Soortgelijke visualiseren bloedplaatjes bijmenging in een groeiende trombus 3 en parallel aan wat eerder is gepubliceerd 4, een geëxterioriseerde mesenterium ader wordt onderzocht door doorvallend licht microscopie. Er zijn diverse andere modellen van intra-vitale microscopie zoals de cremaster spier van ratten 5 of lever van muizen en ratten 6. Intravitale microscopie van mesenterium ader is ontwikkeld en in eerdere studies toegepast door verscheidene groepen. We hebben deze techniek gebruikt om de verschillen in de rekrutering van leukocyten tussen wild type muizen en tryptofaan hydroxylase1 (Tph1) te observeren - / - muizen, die een tekort van niet-neuronale serotonine en vergeleken de resultaten met muizen waarvan de plaatjes serotonine werd farmacologisch uitgeput door de lange termijn zijn de toepassing van de selectieve serotonine heropname remmers fluoxetine 7. We hebben ook onderzocht leukocyten-endotheel interacties na de acute behandeling met fluoxetine 8. In dit protocol richten we ons op intravitale microscopie van mesenterium aders, omdat dit model snel kan worden uitgevoerd en maakt valide metingen van leukocyt-endotheel en bloedplaatjes-leukocyt interacties. Dit is veel moeilijker en tijdrovender in intravitale microscopie van andere organen, zoals bot, lever, huid of spier cremaster. The model beschreven ideaal voor reproduceerbare beoordeling van inflammatoire interactie cel-cel na challengeing met een inflammatoire stimulus, zoals intraperitoneale injectie van lipopolysaccharide.

Intravitale microscopie is een werkwijze die kan worden gebruikt om verschillende processen in verschillende regio&#39;s en schepen in levende dieren te onderzoeken. In dit protocol beschrijven we intravitale microscopie van mesenterium aderen. Dit kan worden uitgevoerd in een korte tijd met reproduceerbare resultaten tonen leukocyt-endotheel interacties in vivo. We beschrijven een inflammatoire instelling na LPS uitdaging van het endotheel. Maar in dit model kan men veel verschillende soorten van toepassing van inflammatoire aandoeningen, zoals bacteriële, chemische of biologische en onderzoek de toediening van geneesmiddelen en hun directe effecten op het leven van dieren en de impact ervan op leukocyten werving. Dit protocol is met succes toegepast op een aantal verschillende behandelingen muizen en hun effect op ontstekingsreactie in vaten. Hierin beschrijven we de visualisatie van leukocyten en bloedplaatjes door fluorescent labelen die met rhodamine 6G. Bovendien kan een specifieke pe beeldvorming zijnrformed met behulp van gericht fluorescerend gemerkte moleculen.

Een overzicht van de experimentele instelling in dit protocol beschreven in figuur 1. Het toont een muis met een geëxterioriseerde ileum-lus en de vaten, die worden waargenomen in opklaren. Figuur 2 toont zekere activatie in onbehandelde dieren wegens de procedure zelf. Er is bijna geen langzame walsen of stevige adhesie van leukocyten vergeleken met LPS behandelde muizen.
