In realtà, poche proteine ​​hanno avuto il loro traffico intracellulare ripreso. L&#39;avvento di tag proteina fluorescente, come foto-attivabili-GFP (paGFP), ha fornito nuovi strumenti per esaminare il traffico intracellulare. Foto-attivabili-GFP è una forma di GFP che è quasi invisibile dopo la sintesi, ma sviluppa forte verde (GFP) fluorescenza dopo l&#39;attivazione da 413 nm di luce laser e questo segnale è stabile per giorni 25,26. I costrutti che utilizzano paGFP sono stati utilizzati per dimostrare il traffico intralysosomal della proteina Lysosomal proteine ​​di membrana 1 (LAMP1) 25, la consegna superficie cellulare della stomatite virus vescicolare glicoproteina (VSVG, un marker di via secretoria) 27, e il fatturato dei perossisomi 28 e 29 autofagosomi. Per visualizzare lo smistamento di APP dal TGN in cellule vive, abbiamo progettato plasmide esprimente ultimi 112 amminoacidi di APP accoppiati a foto-attivabile (paGFP) sul terminale C (denominato βAPP-paGFP) 30. Abbiamo anche eseguito questi esperimenti con lunghezza APP e ha raggiunto risultati simili; il costrutto ΒAPP è usata qui perché fornisce immagini più luminose. Abbiamo poi photo disattivare &amp;# 946; APP-paGFP solo nel TGN, come delimitate dalla TGN marcatore galattosiltransferasi (GalT). Nonostante APP è stato taggato con paGFP di visualizzare un rapido trasporto assonale di APP e la clearance della regione perinucleare, questa è la prima dimostrazione di traffico di APP da un compartimento accuratamente definito ad un altro 11,31,32. Qui, dimostriamo accurata foto-attivazione entro il TGN e la fuoriuscita di APP in compartimenti a valle e la successiva scissione e liquidazione di lisosomi 30. L&#39;accurata fotoattivazione all&#39;interno del Golgi è ampiamente applicabile ad altri sistemi proteici.

Beta-amiloide (A?) È il principale costituente delle placche senili presenti nel cervello dei pazienti con malattia di Alzheimer.
