Photo-activatable-GFP é uma forma de GFP que é quase invisível após a síntese, mas desenvolve verde forte (GFP) de fluorescência depois de ser ativado por 413 nm de luz laser e este sinal é estável durante dias 25,26. Constructos usando paGFP têm sido utilizados para demonstrar o tráfico intralisossomal da proteína lisossómica proteína de membrana 1 (LAMP1) 25, a entrega da superfície da célula do vírus da estomatite vesicular glicoproteína (VSVG, um marcador da via secretora) 27, e o volume de peroxissomas 28 e 29 autofagossomas. Aqui, nós demonstramos foto-ativação exata dentro do TGN ea saída de APP em compartimentos jusante e subsequente clivagem e apuramento de lisossomos 30. A foto-activação preciso dentro do Golgi é amplamente aplicável a outros sistemas de proteínas.

Beta-amilóide (Ap) é o constituinte principal das placas senis encontradas nos cérebros de pacientes com doença de Alzheimer. Ap é derivado da clivagem sequencial da Proteína Precursora Amilóide (APP) por β e y-secretases. Apesar da importância de Ap a patologia da DA, a localização subcelular destas clivagens não está bem estabelecida. Trabalhar em nosso laboratório e outros implicam o sistema endossomal / lisossomal no processamento de APP após a internalização da superfície da célula. No entanto, o tráfego intracelular da APP é relativamente pouco estudada. Embora as proteínas da superfície celular são alteráveis ​​a muitas técnicas de marcação, não existem métodos simples para seguir o tráfico de proteínas de membrana a partir do Golgi. Para este fim, criada construções de APP que foram marcados com foto-activável GFP (paGFP) na extremidade C-terminal. Após a síntese, paGFP tem baixa fluorescência basal, mas pode ser estimulado com luz de 413 nm, to produzir uma fluorescência verde forte, estável. Os resultados típicos mostram pAPP deixar o TGN e parece tráfego rapidamente para LAMP1 (Figura 1a e b).
