El advenimiento de las etiquetas de proteínas fluorescentes, tales como foto-activable-GFP (paGFP), ha proporcionado nuevas herramientas para examinar el tráfico intracelular. Photo-activable-GFP es una forma de GFP que es casi invisible después de la síntesis, pero desarrolla verde fuerte (GFP) de fluorescencia después de ser activado por luz láser 413 nm y esta señal es estable por día 25,26. Las construcciones utilizando paGFP se han utilizado para demostrar el tráfico intralisosomal de la proteína lisosómica proteína de membrana 1 (LAMP1) 25, la entrega de la superficie celular del virus de estomatitis vesicular glicoproteína (VSVG, un marcador de la ruta secretora) 27, y el volumen de los peroxisomas 28 y 29 autofagosomas. Con el fin de visualizar la clasificación de APP desde el TGN en células vivas, hemos diseñado plásmido que expresa los últimos 112 aminoácidos de APP acoplados a la foto-activable (paGFP) en el C-terminal (referido como βAPP-paGFP) 30. También hemos realizado estos experimentos con APP de longitud completa y ha logrado resultados similares; el constructo ΒAPP se utiliza aquí, ya que proporciona imágenes más brillantes. A continuación, la foto-activación y# 946; APP-paGFP sólo en el TGN, como demarcada por el TGN marcador galactosiltransferasa (GalT). Aunque APP ha sido etiquetado con paGFP para visualizar el transporte axonal rápido de la APP y el aclaramiento de la región perinuclear, esta es la primera demostración de tráfico de APP de un compartimento a otro cuidadosamente definido 11,31,32. Aquí, demostramos exacta foto-activación dentro del TGN y la salida de APP en compartimentos de bajada y posterior escisión y el visto bueno de los lisosomas 30. La foto-activación precisa dentro del Golgi es ampliamente aplicable a otros sistemas de proteínas.

Beta-amiloide (Aß) es el constituyente principal de las placas seniles que se encuentran en los cerebros de pacientes con enfermedad de Alzheimer. Aß se deriva de la escisión secuencial de Proteína Precursora de Amiloide (APP) por β y gamma-secretasas. A pesar de la importancia de Aß a la patología AD, la localización subcelular de estas divisiones no está bien establecida. El trabajo en nuestro laboratorio y otros implican el sistema endosomal / lisosomal en el procesamiento de APP después de la internalización de la superficie celular. Sin embargo, el tráfico intracelular de APP es relativamente poco estudiada. Mientras que las proteínas de la superficie celular son modificables a muchas técnicas de etiquetado, no hay métodos simples para seguir el tráfico de proteínas de la membrana del Golgi. Para ello, hemos creado construcciones APP que fueron etiquetadas fotoactivable GFP (paGFP) en el C-terminal. Después de la síntesis, paGFP tiene baja fluorescencia basal, pero puede ser estimulado con 413 nm luz to producir una fluorescencia de color verde fuerte y estable. Al utilizar el aparato de Golgi marcador galactosiltransferasa acoplado a cian proteína fluorescente (GalT-PPC) como un objetivo, estamos en condiciones de precisión fotoactivar APP en la red trans-Golgi. Activada por fotos APP-paGFP puede entonces ser seguido, ya que trafica a los compartimentos posteriores identificados con fluorescencia etiquetados proteínas marcadoras de compartimentos para el endosoma temprano (Rab5), a finales del endosoma (Rab9) y el lisosoma (LAMP1). Por otra parte, el uso de inhibidores de procesamiento de APP incluyendo cloroquina o el inhibidor de la γ-secretasa L685, 458, estamos en condiciones de realizar experimentos de pulso-caza para examinar el procesamiento de APP en las células individuales. Nos encontramos con que una gran parte de APP se mueve rápidamente hacia el lisosoma sin aparecer en la superficie celular, y luego se elimina del lisosoma por divisiones-secretasa similares. Esta técnica demuestra la utilidad de paGFP para seguir el tráfico y el procesamiento de las proteínas intracelulares from el Golgi a los compartimentos aguas abajo.

Los resultados típicos muestran βAPP dejando el TGN y parece tráfico rápidamente a LAMP1 (Figura 1a y b).
