1. Bereid geactiveerde menselijke T-cellen <ol><li> Verzamel 10 ml gehepariniseerd volbloed dat bij een donor door venapunctie. <ol><li> Verkrijgen geïnformeerde toestemming van donoren van tevoren venapunctie. Venapunctie moet worden uitgevoerd door gekwalificeerd en opgeleid personeel. </li><li> Handvat verkregen bloedmonsters met steriele apparatuur en na een passende bioveiligheid richtlijnen. </li></ol></li><li> Verdun 10 ml gehepariniseerd volbloed met 10 ml D-PBS (-). </li><li> Put 15 ml Ficoll-oplossing (Ficoll-Paque PREMIUM) in een nieuwe 50-ml conische buis. </li><li> Laag het 20-ml bloed bereid in stap 1.2 op de Ficoll-oplossing in stap 1,3. Met behulp van een elektrische pipet, laat bloed bereid in stap 1.2 run langzaam langs de binnenwand van de buis. Wees niet naar de bloed-oplossing en Ficoll- oplossing te mengen. </li><li> Centrifugeer bij 400 xg gedurende 30 min bij kamertemperatuur. Voeg 10 ml D-PBS (-) aan de mononucleaire cellen en centrifugeer bij 200 xg gedurende 5 minuten bij kamertemperatuur. </li><li> Verwijder het supernatant, voeg 1 ml medium KBM502 de verzamelde mononucleaire cellen en telling van de cellen met een hemocytometer. Meer dan 5 x 10 6 mononucleaire cellen kunnen worden verkregen uit 10 ml gehepariniseerd volbloed met behulp van Ficoll passende scheiding. </li><li> Bereid een anti-humaan CD3-antilichaam oplossing met een concentratie van 10 ug / ml in D-PBS (-). Voeg anti-humaan CD3-antilichaam oplossing voor een 6-wells plaat, geniet de brandingace van elk putje en incubeer de schaal bij 37 ° C in een 5% CO2 incubator gedurende ten minste 30 min. </li><li> Verwijder het anti-humaan CD3-antilichaam oplossing uit elk putje en eenmaal wassen met D-PBS (-) net voor de cellen te zaaien. </li><li> Seed mononucleaire cellen die werden bereid in Stap 1,9 bij een dichtheid van 2,5 x 10 5 cellen / cm2 op het anti-humaan CD3-antilichaam beklede 6-putjesplaten in KBM502 medium. Infect menselijke T-cellen met SeV Vectoren <ol><li> Na 3-7 dagen cultuur, het verzamelen van de geactiveerde T-cellen met bestaande medium door pipetteren. Overbrengen naar een 15 ml conische buis en centrifugeer bij 200 xg gedurende 5 minuten bij kamertemperatuur. </li><li> Verwijder de bovenstaande vloeistof en voeg 1 ml vers KBM502 medium. Tel de cellen en de plaat 1.5 x 10 6 cellen (in 2 ml vers medium KBM502) in elk putje van een anti-CD3-antilichaam beklede 6-wells plaat. </li><li> Ontdooi de SeV oplossingen van Oct3 / 4-SeV / TSΔF, SOX2-SeV / TSΔF, KLF4-SeV / TSΔF, en c-MYC (HNL) -SeV / TS15ΔF op ijs. </li><li> Voeg de SeV oplossingen die Oct3 / 4-SeV / TSΔF, SOX2-SeV / TSΔF, KLF4-SeV / TSΔF en c-MYC (HNL) -SeV / TS15ΔF afzonderlijk aan de putjes, elk bij een multipliciteit van infectie (MOI) 10. Incubeer bij 37 ° C in een 5% CO 2 incubator voor een verdere 24 uur. </li></ol> 3. Verwijder SeV vectoren van menselijke T-cellen <ol><li> 24 uur na infectie, het verzamelen van de geïnfecteerde cellen met een pipet. Als er hechtende cellen, kunnen ze gemakkelijk worden losgemaakt ze herhaaldelijk pipetteren en neer. Overbrengen naar een 15 ml conische buis en centrifugeer bij 200 xg gedurende 5 minuten bij kamertemperatuur. </li><li> Verwijder de bovenstaande vloeistof met de SeV vectoren en voeg 2 ml vers KBM502 medium. Replate cellen in each van de putjes. Incubeer bij 37 ° C in een 5% CO 2 incubator gedurende nog 24 uur. </li></ol> 4. reseed Cellen op een Matrigel Layer <ol><li> Bereid basaalmembraan matrix oplossing ontdooien matrix gel (zie de lijst Materialen voor het specifieke merk in deze studie) bij 4 ° C en oplossen van 0,2 ml in 10 ml DMEM / F12 medium. Opmerking: Matrix gel (zie de lijst Materialen voor het specifieke merk gebruikt in deze studie) moet ontdooien O / N bij 4 ° C volledig worden ontdooid. Houd het op ijs tot vlak voor gebruik. </li><li> Plate 5 ml basaalmembraan matrix oplossing per plaat in 100-mm schalen en laat bij kamertemperatuur gedurende ten minste 30 min voor het zaaien cellen. Tenminste twee 100-mm schalen zijn nodig voor een experiment. </li><li> Verzamel SeV-geïnfecteerde cellen door pipetteren, overbrengen naar een 15 ml conische buis en centrifugeer bij 200 xg gedurende 5 minuten bij kamertemperatuur. </li><li> Verwijder de bovenstaande vloeistof en voeg 1 ml vers KBM502 medium. Tel het aantal ceLLS met behulp van een hemocytometer, verwijder het basaal membraan matrix-oplossing van 100-mm gerechten bereid in stap 4.2 en plaat 1 x 10 5 en 1 x 10 6 cellen (in 10 ml vers mTeSR gemiddeld) op een 100-mm basaal membraan matrix-gecoat gerechten. Tot slot, gebruik maken van de plaat waarin kolonies gemakkelijk worden geplukt. </li><li> Incubeer de schotel bij 37 ° C in een 5% CO 2 incubator O / N. </li><li> Verander het medium tot 10 ml vlees mTeSR medium elke andere dag. Opmerking: De kolonies van de SER en iPSCs zijn rond en plat, zoals weergegeven in figuur 1B. </li><li> Transfer geselecteerde kolonies in de 96-well plaat gevuld met mTeSR medium in stap 5,3. </li><li> Voeg 200 ul mTeSR medium aan elk putje en zorgvuldig pipet op en neer om de kolonie breken in kleine klontjes onder de stereomicroscoop met een pipet 200 ul. Verwijder de basaalmembraan matrix oplossing uit de goed bereid in 5,2 en zet elke kolonie in een putje van de basaalmembraan matrix beklede 6-wells plaat met 2 ml mTeSR medium per putje. </li><li> Verander de middellange tot 2 ml vlees mTeSR medium elke andere dag. </li></ol> 6. Zorg voor T-cel afgeleid iPSCs De T-cel afgeleide iPSCs kan worden gehandhaafden opgeslagen met behulp van dezelfde technieken als voor menselijke iPSCs en menselijke SER. We describe a protocol for generating iPSCs from human peripheral T cells in serum-free and feeder-free conditions using a combination of matrigel, mTeSR medium, and SeV vectors. For clinical applications of iPSCs, it is important to have a protocol for stably generating iPSCs and a less-invasive method for cell sampling. Although generating iPSCs with the combination of matrigel and mTeSR medium showed lower reprogramming efficiency than that with knockout serum replacement (KSR) medium and feeder-cells, this combination achieves stable generation of iPSCs from donors16. Stable iPSC generation and less-invasive cell sampling has the advantage of being able to increase the number of donors for iPSC generation. 
SeV vectors, a minus-strand RNA virus, is not integrated into the host genome. Therefore the risks of tumorigenesis can be avoided at the step of reprogramming factor induction20-24. Additionally, the feeder-free conditions, which use a combination of defined culture medium and matrigel instead of a feeder layer, make it possible to minimize the potential risks of exposure to unknown exogenous factors. The fusion of these techniques provides a less invasive and safer iPSC technology for regenerative medicine16.
Important steps in this protocol are the step of activating human T cells (Step 1) and infecting them with SeV vectors (Step 2). When no ESC-like colony is obtained in the culture dish at an optimal time after SeV infection, the following should be considered. First, the confluency of the mononuclear cells before T cell activation may not be appropriate because optimal activation of T cells is critical for SeV infection25,26. Too high a density of mononuclear cells leads to cell death, interfering with the proper activation of T cells. Too low a density of mononuclear cells also disturbs the proper activation and proliferation of T cells. Therefore, the confluency of mononuclear cells should be checked and adjusted accordingly. Second, the dosage of SeV vectors may not be sufficient. The induction efficiency of iPSC colonies depends on the dosage of the virus6. If no ESC-like colony is observed after SeV infection, the option of increasing the virus dosage up to an MOI of 15-20 should be considered. If signs of iPSC colony differentiation are observed, the frequency at which medium is changed may be increased up to every other day, or to every day when colonies are larger. 
The limitation in this protocol consists of this method not being virus free. Although SeV for cell reprogramming is commercially available with ease, users need to prepare equipment according to the appropriate biosafety level. As another method for generating integration-free iPSCs, episomal vectors have been used until now27-29. Although episomal vectors can be used with equipment with a lower level of biosafety, episomal vectors might insert into the host genome at extremely low rates. Therefore, additional checks are required to confirm the disappearance of transgenes, as when using SeV.
There is another limitation in that this technique is not absolutely free from animal-derived products. Some substrates, such as matrigel, anti-CD3 mAb, dissociation solution and SeV solution are derived from animal products and are therefore associated with a risk of transferring xenogeneic pathogens. However, the reduction of animal-derived substrates in the culture system is meaningful for the clinical application of iPSC technology due to the lower risk.

iPSCs hebben aanzienlijke aandacht getrokken als een baanbrekende bron van cellen voor regeneratieve geneeskunde 1-3. Tot op heden hebben diverse werkwijzen voor het genereren iPSCs gemeld 4,5. Hiervan iPSCs gegenereerd uit humane T-cellen zijn van bijzonder belang vanwege de minder invasieve methode van celbemonstering 6-8. Bovendien iPSCs afgeleid van T-cellen bevatten herrangschikkingen van het T cel receptor (TCR) genen en derhalve een bron van antigen specifieke T-cellen 9,10. Daarom is het genereren van T-cel afgeleid iPSCs veilig is nuttig voor vordert regeneratieve geneeskunde. Deze methode is gebaseerd op het concept van het risico van onvoorspelbare pathogeniciteit. Voor een veilige klinische toepassing van iPSCs is het belangrijk om het risico op blootstelling aan pathogenen 11. Eerder in verschillende kweeksystemen van pluripotente stamcellen hebben foetaal runderserum en voedingscellen gebruikt essentieel reagenss 12. Het wegnemen van beide reagentia uit het kweeksysteem voorkeur dat iPSC genereren om het risico op onvoorspelbare pathogeniciteit verminderen. Bovendien heeft deze werkwijze het voordeel dat invasieve celbemonstering van patiënten en moeizame bereiding van voedingscellen. Omdat T cel afgeleid iPSCs reeds met succes toegepast bij ziekteonderzoek 13,14, deze werkwijze is ook toepasbaar en nuttig voor het genereren van ziektespecifieke iPSCs van patiënten. Onder T cel herprogrammering werkwijzen, met het Sendai-virus (SeV) vector een gen voertuig een methode die iPSCs kan genereren met een hoog rendement 7,16. Bovendien, omdat SeV is een enkelstrengs RNA-virus en geen DNA fase nodig voor replicatie, zijn gebruik in iPSC generatie voorkomt het breken van het gastheergenoom 17-19. Daarom hebben we vastgesteld protocollen voor het genereren iPSCs van humane perifere T-cellen in serum-free en voedingsvrije met gebruikmaking van een combinatie van Matrigel, mTeSR medium en SeV vectoren.

Onlangs hebben iPSCs aandacht getrokken als een nieuwe bron van cellen voor regeneratieve therapieën. Hoewel de eerste werkwijze voor het iPSCs aangevoerde huidfibroblasten verkregen door invasieve biopsie en retrovirale genoom inbrengen van transgenen zijn er veel pogingen om deze nadelen te vermijden zijn. Humane perifere T-cellen zijn een unieke cel bron voor het genereren van iPSCs. iPSCs afgeleid van T-cellen bevatten herrangschikkingen van het T cel receptor (TCR) genen en zijn een bron van antigen specifieke T-cellen. Daarnaast T-celreceptor omlegging in het genoom heeft het potentieel om afzonderlijke cellijnen label en onderscheid maken tussen getransplanteerde en donorcellen. Voor een veilige klinische toepassing van iPSCs, is het belangrijk om het risico van blootstelling van nieuw gegenereerde iPSCs aan schadelijke agentia minimaliseren. Hoewel foetaal runderserum en voedingscellen essentieel voor pluripotente stamcellen celkweek zijn, verdient het de voorkeur om ze van het kweeksysteem te verwijderen om het risico te verminderenonvoorspelbare pathogeniciteit. Om dit aan te pakken, hebben we een protocol opgesteld voor het genereren iPSCs uit humane perifere T-cellen middels Sendai virus het risico van blootstelling iPSCs op undefined ziekteverwekkers te verminderen. Hoewel de behandeling van Sendai-virus vereist apparatuur met de juiste bioveiligheid niveau Sendai virus infecteert geactiveerde T-cellen zonder genoom inbrengen, maar met een hoog rendement. In dit protocol tonen we het genereren van iPSCs uit humane perifere T-cellen in voedingsvrije met gebruikmaking van een combinatie van geactiveerde T-celkweek en Sendai virus.

<html> Met behulp van dit protocol, kunnen gebruikers iPSCs uit menselijke perifere T-cellen stabiel te genereren. iPSC opwekking van T-cellen met SeV en matrigel vertoonden ongeveer 0,002% -. 0,005% rendement cel herprogrammering tussen verschillende donoren zaken en SeV werden niet gedetecteerd na een aantal passages 16 Figuur 1A toont een schema van het protocol voor het genereren van T cel-afgeleide iPSCs in feeder-vrije omstandigheden met behulp matrigel en mTeSR medium.
