La souris est actuellement le modèle prédominant pour la recherche sur l&#39;obésité. Le court intervalle de génération et des outils génétiques disponibles chez la souris ont été inégalée à ce jour. Cependant, le poisson zèbre a aussi un intervalle de génération court (3 - 4 mois) et dépasse même la souris dans la facilité de la manipulation génétique 1,2. Le poisson zèbre maintient près de 90% des gènes de mammifères, tout en dépassant de loin la souris dans nombre de descendants et le potentiel pour une utilisation dans les écrans génétiques et drogues 3. Pour exploiter le potentiel du modèle de poisson zèbre pour les études à l&#39;obésité, les dosages doivent être développées pour étudier les facteurs qui influencent la régulation du poids corporel, y compris les dépenses d&#39;énergie. Comme métabolites sont traités par β-oxydation et l&#39;oxygène du cycle de l&#39;acide tricarboxylique est consommé et 2 NADH est produit. Ainsi, le NADH 2 est un indicateur direct du flux de métabolites par l&#39;intermédiaire des voies métaboliques (oxydation) de métabolite. Dans poikilotherms, H + à travers la fuite la membrane mitochondriale interne, qui découple oxydation de NADH deux la synthèse d&#39;ATP, est de 4 à 5 fois plus faible que dans les homéothermes 4. En outre, la manipulation génétique ciblée, y compris effet de choc et knock-in, peuvent être appliquées rapidement. Nous avons précédemment montré que ce dosage peut être combiné avec l&#39;application de la drogue bain et knock-down génétique ou knock-out pour identifier des composés qui modifient les gènes et les taux de 5 métabolique. Poisson zèbre sont une importante organisme modèle avec des avantages inhérents qui ont le potentiel pour faire un poisson zèbre modèle largement appliqué pour l&#39;étude de l&#39;homéostasie énergétique et l&#39;obésité. La petite taille du poisson zèbre permet des dosages sur les embryons soient menées dans un format de plaque à 96 ou 384 puits, les technologies basées Morpholino et CRISPR promouvoir la facilité de manipulation génétique, et de traitement de la toxicomanie par l&#39;application de bain est viable.
