Hurtighet av leukocytter mobilisering bestemmer positive utfallet av immunresponser. Disse intrikate prosesser er avhengige av spesifikke molekyler (f.eks selectins, endotel-bundet chemokiner) til stede på den betente endotelet som bidrar til etablering av selvklebende kontakter mellom sirkulerende leukocytter og endotelet 1-3 .For få innsikt i molekylene innblandet i leukocytter rekruttering kaskade, er det viktig å visualisere de kinetikken til celle rekruttering og å spore oppførselen til hver celle / populasjon. Effektiv immunresponsen er avhengig av rask mobilisering av blodleukocytter til stedet for infeksjon eller skade. Time-lapse filmer samlet fra 30 min til flere timer med bildebehandling tillate analyse av leukocytter atferd i tarmens blodårer under begge stabile og betennelsestilstander. Vi beskriver også fremgangsmåten for å lokalt indusere blodåre betennelse med TLR2 / TLR1 agonist Pam3SK4 og overvåke subsequent rekruttering av nøytrofile granulocytter og monocytter. Den presenterte teknikken kan også brukes til å overvåke andre populasjoner av leukocytter og undersøke molekyler involvert i leukocytt rekruttering eller trafficking ved hjelp av andre stimuli eller transgene mus.
