Een eenvoudig en efficiënt protocol voor zowel expressie en zuivering van een grote hoeveelheid recombinant muizen Shadoo eiwit wordt gepresenteerd. De beschreven werkwijze maakt succesvolle solubilisatie en zuivering van (His6) gemerkte recombinant Shadoo eiwit. Het is belangrijk, zoals in de titel, dat bij expressie in een prokaryotische bacterie E. Shadoo coli accumuleert in inclusielichamen. Prion neurodegeneratieve ziekten, die bovine spongiforme encefalopathie bij runderen, scrapie bij schapen en ziekte van Creutzfeldt-Jakob bij mensen omvatten fataal en ongeneeslijk. Prion ziekten worden gekenmerkt door conformatieveranderingen cellulaire prioneiwit voornamelijk uit α-helices, in de cross-β-sheet amyloid verrijkte conformer 1,2. Cross-β-structuren bevatten dicht opeengepakte en sterk geordende kristallijne-achtige β-sheets gestabiliseerd met waterstofbruggen 3,4. Prion amyloids kan zichzelf repliceren vanwege de β-strengen aan de groeiende rand die een sjabloon voor werving verschaffen en het omzetten van een monomeer eiwit unit. Doppel een relatief stijve constructie gestabiliseerd met di-sulfide bruggen en niet aan zichzelf polymeriseren en aggregaat om amyloïde fibrillen 9. In tegenstelling, vergelijkbaar met prion eiwitten, Shadoo een β-sheet-verrijkte structuur en zelf-associëren in amyloïde fibril structuren aannemen. Er werd aangetoond dat Shadoo kan fibrilleren onder natuurlijke condities of na binding negatief geladen membranen 10,11. Omzetting van Shadoo in amyloïde-achtige vezels kunnen worden geassocieerd met pathologieën. Inderdaad zijn de mechanismen die amyloïde structuren ziekte veroorzaken slecht begrepen. Shadoo draagt ​​een hydrofoob domein (HD) en een reeks tandem Arg / Gly herhaalt vergelijkbare to de N-terminale deel van prion eiwit (Figuur 1A). Het plasmide werd gebruikt voor het transformeren van E. coli voor productie van N-terminale His6-fusie-eiwit Shadoo. Het expressiesysteem is goed ingeburgerd in ons laboratorium en wordt vaak gebruikt in onze lopende projecten 6,15,16. Een belangrijk punt voor de expressie van Shadoo is de keuze van de E. coli stam. Terwijl bevoegde bacteriestam BL21 wordt gebruikt voor expressie van recombinant proeiwitten Shadoo werd succesvol tot expressie gebracht en gezuiverd uit alleen de getransformeerde SoluBL21 bacteriestam. SoluBL21 competente E. coli is een verbeterde mutant van BL21 gastheerstam ontwikkeld voor de productie van eiwitten waarvan de expressie in de bovenliggende BL21 gaf geen detecteerbaar oplosbaar product. Hoog niveau expressie van Shadoo in SoluBL21 E. coli leidt tot ophoping van het eiwit in inclusielichamen. Een algemeen kenmerk, wanneer een niet-natief eiwit sterk tot expressie gebracht in E. coli, dit eiwit accumuleert in onoplosbare inclusielichamen. Shadoo waarschijnlijk aggregaten via niet-covalente hydrofobe of ionische interacties (of een combinatie van beide) tot zeer verrijkt dynamische structuren gevormd door het gevouwen eiwit in verschillende mate. Bijgevolg, het zuiveren ten minste twee fasen: (i) een selectieve scheiding van het proteïne uit andere biomoleculen in denaturatie medium, en (ii) een renaturatie van het gezuiverde eiwit via in vitro vouwingtechnieken. De selectieve scheiding van Shadoo werd in een bufferoplossing met 8M ureum (of alternatief 6M guanidine HCl) bereikt. De ureum verwijderen en renaturatie van het eiwit kan door het toepassen van verschillende protocollen: (i) renaturatie in een zure pH-oplossing een ongestructureerde monomeer Shadoo eiwit, of (ii) renaturatie verkrijgen van een oplossing met pH ≥ 7 tot Shadoo gepolymeriseerde verkrijgen in stabiele amyloïde vezels met karakteristieke cross-β-sheet motieven.

De Escherichia coli expressiesysteem is een krachtig hulpmiddel voor de productie van recombinante eukaryotische proteïnen. We gebruiken het om Shadoo een eiwit behorend tot de familie prion produceren. Een chromatografische werkwijze voor het zuiveren van (His) 6 gemerkte recombinant Shadoo uitgedrukt als inclusielichamen wordt beschreven.
