Het chromosoom voorbereiding experiment is uitgevoerd met behulp van de jonge wortels van granen behoren tot het gras familie (Poaceae). Alle geanalyseerde soorten hebben 14 relatief lange mitotische metafasechromosomen (11-15 pm) in het diploïde genoom set en behoren tot grote genoom-soorten (5,1-7,9 GBP). Fluorescentie in situ hybridisatie (FISH) is een doeltreffend instrument voor de fysieke kaart brengen van enkele en hoog kopie sequenties op het chromosomaal niveau. Voorwaarde is de bereiding van hoge kwaliteit chromosoom spreads. Er is geen algemene chromosoom voorbereiding protocol dat even geschikt voor dier- en plantencellen zou zijn. Bereiding van plantaardige chromosomen is bijzonder uitdagend te wijten aan de starre celwand en diverse cytoplasma consistentie binnen verschillende soorten. Eén van de gunstige werkwijzen voor de bereiding van plantaardige chromosomen is een zogenaamd druppel techniek ook wel drop strooitechniek en air-droogtechniek 1,2. Deze werkwijze werd eerst geïntroduceerd in 1958 door rothfels en Siminovitch in vitro gekweekt zoogdiercellen 3. Later Martin et al. 4 en Kato et al. 5 aangepaste methode voor planten. Recenter een werkwijze genaamd &quot;SteamDrop &amp;# 39; ontwikkeld dat waterdamp voor de bereiding van niet-overlappende chromosomen 6. Hoewel de positieve invloed van een hoge luchtvochtigheid was eerder 7 waargenomen, &#39;SteamDrop&#39; levert een gecontroleerde workflow van hoogwaardige chromosoom voorbereiding 6. De stoombehandeling veroorzaakt rekken chromosomen waarschijnlijk verbonden met een aantal wijzigingen van chromosomale eiwitten. De kwaliteit van de verkregen metafase smeersels is zeer hoog, hoewel behoud van voldoende totale metafase spreads volgende FISH experimenten vereist technische expertise. Hier presenteren we een protocol voor de voorbereiding van de mitotische granen chromosomen die geschikt zijn voor de FISH detectie van enkele en hoog kopie sondes 5,8. Deze werkwijze is een verbeterde variant van de luchtdroge dropping beschreven door Kato 9 uitgevoerd onder relatieve vochtigheid van 50% -55% (figuur 1). Dit protocol omvat een gereduceerd aantalvan wasstappen maken van de toepassing ervan eenvoudig, efficiënt en reproduceerbaar. Met behulp van dit protocol verkregen wij hoogwaardige chromosoom spreads en FISH resultaten voor Hordeum vulgare, H. bulbosum, H. marinum, H. murinum, H. pubiflorum en Secale cereale.

Bereiding van chromosoom spreads is een voorwaarde voor de succesvolle uitvoering van fluorescentie in situ hybridisatie (FISH). Voorbereiding van hoogwaardige plantaardige chromosoom spreads is een uitdaging te wijten aan de starre celwand. Eén van de erkende werkwijzen voor de bereiding van plantaardige chromosomen is een zogenaamd druppel preparaat, ook wel drop verspreiding of lucht-droogtechniek. Hier presenteren we een protocol voor de snelle bereiding van mitotische chromosoom spreads geschikt voor de FISH detectie van enkele en hoog kopie DNA-sondes. Deze werkwijze is een verbeterde variant van de luchtdroge neerzetten uitgevoerd onder een relatieve vochtigheid van 50% -55%. Dit protocol omvat een gereduceerd aantal wasstappen van zijn aanvraag eenvoudig, efficiënt en reproduceerbaar. Duidelijke voordelen van deze aanpak zijn goed gespreid, onbeschadigd en tal metafasechromosomen dienen als een perfecte voorwaarde voor een succesvolle FISH analyse. Met behulp van dit protocol verkregen wij hoogwaardige chromOsome spreads en reproduceerbare FISH resultaten voor Hordeum vulgare, H. bulbosum, H. marinum, H. murinum, H. pubiflorum en Secale cereale.

Microscopische objectglaasjes met metafase mitotische smeersels werden bereid met een snelle drogen dropping chromosoom bereidingsmethode beschreven (Suppl. Figuur 1). FISH analyse werd uitgevoerd met zowel repetitief en single-copy sequenties.
