In dit protocol tonen we nieuwe werkwijzen voor micro-injectie van genetisch materiaal, hetzij cDNA of shRNA, in de 2-PN muizenembryo&#39;s om de rol van neogenin begin celdifferentiatie staand tijdens embryogenese van de muis. Preimplantatie embryonale ontwikkeling kan worden onderverdeeld in verschillende afzonderlijke fasen, van de ene cel stadium de morula en blastocyst. Veel bewijs suggereert dat de polariteit en positionele signalen een rol spelen in het begin van lot van de cel bepaling in de kaart spelen trophectoderm (TE) en de binnenste celmassa (ICM). De aard van de signalen, hoe ze worden getransduceerd in cellulaire signalen en de fysiologische contexten waarin zulke signalen kunnen initiëren cellijn differentiatie niet bekend. Deze gedifferentieerde cellen vervolgens aan specialisatie, beginnend op kenmerkende structuren en functies voor embryo vorming en groei van de placenta 1-3 te nemen. Pre-implantatie embryo&#39;s zijn bijzonder vatbaar voor manipulatie door directe injectie van genetisch gemodificeerde materialen zoals siRNA of cDNA doelwit en kan dus worden gebruikt om specifieke doelmoleculen bestuderen. Er is een groeiend besef dat de genetischeanalyse van gewervelde embryo&#39;s is van cruciaal belang voor onze kennis van de embryonale ontwikkeling 4. Preciezer van een wildtype gen of een mutante vorm tot een embryo in een geschikte context te bereiken gain of verlies van functie van het gen aanzienlijk vergemakkelijkt de studie van ontwikkeling gereguleerde genen. Verlies- en gain-of-function via micro-injectie van genetisch materiaal in afzonderlijke niet-zoogdieren en zoogdieren embryo begin relatief eenvoudig vanwege hun grote omvang en grote ontvankelijkheid 5. Micro-injectie wordt meestal uitgevoerd met behulp van niet-virale vectoren. Bijzonder voordeel is gehouden met het gemak van het gebruik van niet-virale vectoren in virale vectoren en transgenen. Een snelle verlies- of gain-of-function expressiesysteem in muizenembryo&#39;s is ontwikkeld dat ons in staat stelt te ontleden en te begrijpen genfuncties in gewervelde embryologie 6,7. TL-etikettering van embryo&#39;s zou zeer de selectie van MICR vergemakkelijkenoinjected of genetisch gemanipuleerde embryo en tegelijkertijd verlenen middel indirect kwantificeren van het expressieniveau van microgeïnjecteerde siRNA of cDNA gebaseerd op fluorescentie-intensiteit 8. Om deze etikettering, fluorescerend eiwit cDNA, GFP of RFP te bereiken, zijn co-geïnjecteerd als ofwel fusieconstructen of afzonderlijk. De fluorescentie-intensiteit van GFP of RFP onthult de mate van expressie van siRNA of vreemd DNA dat verliesgevende zorgen of gain-of-function is bereikt. Hier presenteren we een micromanipulatie protocol voor verlies- en gain-of-functie techniek die ons toeliet om neogenin identificeren als een receptor die extracellulaire signalen belangrijk in het begin van celdifferentiatie relais in pre-implantatie muizenembryo&#39;s.

loss- and gain-of-function approach to investigate early cell fate determinants in preimplantation mouse embryos

Gene silencing and overexpression techniques are instrumental for the identification of genes involved in embryonic development. Direct target gene modification in preimplantation embryos provides a means to study the underlying mechanisms of genes implicated in, for instance, cellular differentiation into the trophectoderm (TE) and the inner cell mass (ICM). Here, we describe a protocol that examines the role of neogenin as an authentic receptor for initial cell fate determination in preimplantation mouse embryos. First, we discuss the experimental manipulations that were used to produce gain and loss of neogenin function by microinjecting neogenin cDNA and shRNA; the effectiveness of this approach was confirmed by a strong correlation between the pair-wise expression levels of either red fluorescent protein (RFP) or green fluorescent protein (GFP) and the immunocytochemical quantification of neogenin expression. Secondly, overexpression of neogenin in preimplantation mouse embryos leads to normal ICM development while neogenin knockdown causes the ICM to develop abnormally, implying that neogenin could be a receptor that relays extracellular cues to drive blastomeres to early cell fates. Given the success of this detailed protocol in investigating the function of a novel embryonic developmental stage-specific receptor, we propose that it has the potential to aid in exploration and identification of other stage-specific genes during embryogenesis.
