I det nuvarande protokollet visar vi nya metoder för mikroinjektion av genetiskt material, antingen cDNA eller shRNA, i embryona 2-PN musen för att utforska den roll neogenin i början av celldifferentiering under mus embryogenes. Preimplantatorisk embryonal utveckling kan delas in i flera olika etapper, från en-cellstadiet till morula och blastocyst. Mycket bevis tyder på att polariteten och positionella signaler spela en roll i tidig determinering in trophectoderm (TE) och den inre cellmassan (ICM). Dock arten av de signaler, hur de är omvandlade till cellulära signaler, och de fysiologiska sammanhang där sådana signaler är i stånd att initiera cellhärstamning differentiering är inte kända. Dessa differentierade celler genomgå sedan specialisering, börjar ta på karakteristiska strukturer och funktioner som behövs för embryo korrekt bildning och tillväxt av moderkakan 1-3. Preimplantatorisk embryon är särskilt mottagliga för manipulation genom direkt injektion av modifierade genetiskt material som siRNA eller mål-cDNA och därför kan användas för att studera specifika målmolekyler. Det finns en växande insikt om att genetiskanalys av ryggradsdjur embryon är avgörande för att öka kunskaperna om embryonal utveckling 4. Exakt införande av en vildtyp-gen eller en muterad form i ett embryo i tid sammanhang att åstadkomma GAIN- eller förlust-av-funktion av genen har i hög grad underlättat studier av utvecklingsmässigt reglerade gener. Förlust- och få-av-funktion via mikroinjektion av genetiskt material till individuella tidiga icke-däggdjur och däggdjurs embryon är relativt enkelt på grund av sin storlek och stor mottaglighet 5. Mikroinjektion utförs typiskt med användning av icke-virala vektorer. Särskild fördel har tagits av den enkla att använda icke-virala vektorer över virala vektorer och transgener. En snabb förlust eller vinst-of-funktionsuttryck systemet musembryon har utvecklats som gör det möjligt för oss att dissekera och förstå genfunktioner i ryggradsdjur embryologi 6,7. Fluorescensmärkning av embryon skulle i hög grad underlätta valet av microinjected eller genetiskt manipulerade embryon och samtidigt ge ett medel för att indirekt kvantifiera uttrycksnivån för mikroinjiceras siRNA eller cDNA baserat på fluorescensintensitet 8. För att åstadkomma detta märkning, fluorescerande protein cDNA, GFP eller RFP, samar injiceras som antingen fusionskonstruktioner eller separat. Fluorescensintensiteten hos GFP eller RFP avslöjar graden av uttryck av siRNA eller främmande DNA för att säkerställa att förlust- eller vinna-of-funktion har uppnåtts. Här presenterar vi en mikromanipulation protokoll för förlust och vinst-of-funktion teknik som tillät oss att identifiera neogenin som en receptor som förmedlar extracellulära signaler viktiga i början av celldifferentiering i embryon preimplantatorisk mus.

loss- and gain-of-function approach to investigate early cell fate determinants in preimplantation mouse embryos

Gene silencing and overexpression techniques are instrumental for the identification of genes involved in embryonic development. Direct target gene modification in preimplantation embryos provides a means to study the underlying mechanisms of genes implicated in, for instance, cellular differentiation into the trophectoderm (TE) and the inner cell mass (ICM). Here, we describe a protocol that examines the role of neogenin as an authentic receptor for initial cell fate determination in preimplantation mouse embryos. First, we discuss the experimental manipulations that were used to produce gain and loss of neogenin function by microinjecting neogenin cDNA and shRNA; the effectiveness of this approach was confirmed by a strong correlation between the pair-wise expression levels of either red fluorescent protein (RFP) or green fluorescent protein (GFP) and the immunocytochemical quantification of neogenin expression. Secondly, overexpression of neogenin in preimplantation mouse embryos leads to normal ICM development while neogenin knockdown causes the ICM to develop abnormally, implying that neogenin could be a receptor that relays extracellular cues to drive blastomeres to early cell fates. Given the success of this detailed protocol in investigating the function of a novel embryonic developmental stage-specific receptor, we propose that it has the potential to aid in exploration and identification of other stage-specific genes during embryogenesis.
