Målet med denne studien var å bestemme nivået av miRNA uttrykket i brystkreft vev ved hjelp av miRNA in situ hybridisering. Det må bemerkes at i dette eksperimentet, ble alle betingelser optimalisert for brystkreft vev. Videre optimalisering kan være nødvendig for andre typer vev. Alle oppløsninger må gjøres med DEPC vann og alle containere som skal brukes skylles med DEPC behandlet vann og deretter autoklavert. Brystkreft er blant de vanligste kreftformer som påvirker den kvinnelige befolkningen over hele verden. Over 1,3 millioner tilfeller av brystkreft er rapportert hvert år på verdensbasis 1,2. Selv om tumorcellene har tradisjonelt vært betraktet som biologisk homogent og meget proliferativ, har det blitt tydelig at brystkreft er genetisk og klinisk heterogen. Motstand mot utbredte kreft narkotika er et kjennetegn på avanserte brystkreft, som fører til dødelighet hos de fleste pasienter gjennom sin tilrettelegging av kreft progresjon og avstand metastase tre. MicroRNAs (mirnas) er en klasse av små RNA-molekyler ca. 22 nukleotider lange som regulerer genuttrykk ved å blokkere mRNA oversettelse eller formidling av mRNA degradering 4. Mer enn 2000 forskjellige mirnas har hittil blitt identifisert hos mennesker, som er knyttet til menneskelige sykdommer 5. Et økende rekke studier have viste at mirnas kan fungere som onkogener og tumorundertrykkere, og er ofte dysregulerte i tumorer 6. mirnas sterkt påvirke alle de primære tumordannelse ved å styre de store regulatorer av cellesyklusprogresjon, begynnende alderdom, apoptose og autophagy, sammen med de av tumorcelle motilitet, invasjon og metastase 7. Avvikende miRNA uttrykket har også blitt assosiert med kliniske implikasjoner som scene, differensiering, prognose og respons på adjuvant behandling 8. Spesielt økte Mir-146 uttrykk er korrelert med dårlig prognose i lungekreftpasienter 9. En annen studie har vist at mistet ekspresjon av miRNA-let7b er knyttet til ondartede fenotyper og følgelig dårlig overlevelse 6. Forstå typer celler og strukturer som uttrykker mirnas er en viktig del av å forstå mekanikken gjennom hvilke endringer i miRNA uttrykket innflytelse celle ogvev fenotyper 10. Visse mirnas funnet å bli utskilt i stromale celler er tatt av epitelceller og spesielt handle på sine mål. I samme ånd, er MIR-212 og MIR-132 utskilt av stroma og regulere epithelial-stromal interaksjoner som kreves for ductal utvekst under brystkjertelen utvikling 11. Det overordnede målet med denne artikkelen er å forklare en detaljert protokoll for å oppdage miRNA uttrykket i brystkreft vev gjennom miRNA in situ hybridisering. Vi har optimalisert alle forutsetninger for å analysere nivået av miRNA-489 uttrykk i brystkreftpasientprøver. For dette formål er det benyttet en DIG merket låst nukleinsyre (LNA) probe i vårt eksperiment. Noen av nukleotider hadde en LNA modifikasjon, det vil si, en metylenbro som forbinder de to &#39;oksygenatom og 4&#39; karbonatom i ryggraden ribose som fester nukleotid slik at den forblir &quot;er låst på plass&quot; etter hybridisering. Som et resultat, er det mye vanskeligerefor den hybridiserte komplisert å denaturere 12,13.

clinicopathological analysis of mirna expression in breast cancer tissues by using mirna in situ hybridization

In this article, we describe a detailed protocol for miRNA detection in breast cancer tissue using in situ hybridization with a digoxigenin-labelled LNA (Locked Nucleic Acid) probe. The probe was recognized by anti-DIG alkaline phosphatase antibodies and later developed using alkaline phosphatase substrate producing fluorescence signals. Here we utilized miRNA in situ hybridization (MISH) technique to analyze expression of miR-489 in tissues from breast cancer patients. This technique can detect the localization of miRNA of interest in individual tissue samples. This technique can be used to compare the expression of desired miRNA in tumor tissue with that in adjacent normal tissue and to identify the specific structures responsible for expressing this miRNA. This technique can be very useful in answering certain clinical questions, such as role of specific miRNA in the development of cancer. Our results indicate that mammary epithelial cells express significantly higher levels of miR-489 than adjacent tumor cells.
