Helicobacter pylori (H. pylori) is een belangrijke oorzaak van gastritis en maagzweren. Deze bacterie is ook geclassificeerd als een kankerverwekkende stof bij de mens door het Internationaal Agentschap voor Kankeronderzoek, onderdeel van de World Health Organization, in 1994. Er wordt geschat dat de prevalentie van H. pylori infectie 70% in de ontwikkelingslanden en 30-40% in de geïndustrialiseerde landen 1. Hoewel de infectie tarief van H. pylori afneemt in de geïndustrialiseerde landen, de infectie tarief van H. pylori in de ontwikkelingslanden een hoog 2. De standaard behandeling om H. uit te roeien pylori-infectie bestaat uit de toediening van een protonpompremmer, PPI, en twee antibiotica, claritromycine plus amoxicilline of metronidazol 3. Echter, de opkomst van de antibioticaresistentie in H. pylori gerelateerde zweren therapie dringt er bij de ontwikkeling van nieuwe strategieën ter voorkoming van or genezen van de infectie. Ontwikkeling van preventieve en / of therapeutische vaccinatie tegen H. pylori kan een alternatieve benadering H. beheersen pylori-infectie. Helicobacter pylori-neutrofiel activerend eiwit (HP-NAP), een belangrijke virulentie factor H pylori, werd voor het eerst geïdentificeerd in water extracten van H. pylori met het vermogen om neutrofielen te activeren om zich te houden aan endotheelcellen en produceren van reactieve zuurstofsoorten (ROS) 4. Zo kan HP-NAP een pathologische rol spelen door het activeren van neutrofielen aan maagontsteking, die verder veroorzaakt maagzweer of H. induceren pylori-geassocieerde gastrische aandoeningen. Toch HP-NAP is een potentiële kandidaat voor klinische toepassingen 5,6. Door de immunogene en immunomodulerende progenschappen van HP-NAP, kan dit eiwit worden gebruikt om vaccins, therapeutische middelen en diagnostische instrumenten te ontwikkelen. Een klinisch onderzoek werd uitgevoerd voor het gebruik van recombinant HP-NAP als een van de bestanddelen van een eiwit vaccin tegen H. pylori. Dit vaccin bestaat uit recombinant HP-NAP, cytotoxine-geassocieerde gen A (CagA) en vacuolating A cytotoxine (VacA) proteïnen geformuleerd met aluminiumhydroxide en is verder aangetoond veilig en immunogeen bij mensen 7 te zijn. Ook HP-NAP werkt als een krachtige immunomodulator triggert T helper type 1 (Th1) -polarized immuunresponsen voor kankertherapie 8 en omlaag reguleren Th2-gemedieerde immuunresponsen opgewekt door allergische reacties en parasitaire infecties 9,10. Zoals voor diagnostiek, heeft recombinant HP-NAP-gebaseerde ELISA werd toegepast op serum antilichamen tegen HP-NAP in H. pylori geïnfecteerde patiënten 11. Een studie toonde aan dat het niveau van HP-NAP-specifieke antilichamen in sera van H. pylori geïnfecteerde patiënten met maagkanker significant hoger dan die van patiënten met chronische gastritis 12. Een andere studie toonde ook aan dat serum antilichamen tegen HP-NAP zijn geassocieerd met de aanwezigheid van niet-cardia adenocarcinoom van de maag 13. Aldus kan recombinant HP-NAP-gebaseerde ELISA toegepast om antilichamen in serum tegen HP-NAP detecteren prognose maagkanker H. pylori geïnfecteerde patiënten. Samen genomen, kon het gezuiverde HP-NAP verder worden gebruikt voor de preventie, behandeling en prognose van H. pylori-geassocieerde ziekten en kanker. Onder de verschillende methoden voor zuivering van recombinant HP-NAP expressie gebracht in Escherichia coli (E. coli) in de natieve vorm gemeld tot nu toe, een tweede zuiveringsstap waarbij gelfiltratiechromatografie nodig om zeer zuivere HP-NAP 14-16 vinden . Hier, een methode waarbij negatieve modus batch chromatografie metdiethylaminoethyl (DEAE) ionenwisselaarharsen beschreven voor zuivering van HP-NAP overexpressie gebracht in E. coli met hoge opbrengst en een hoge zuiverheid. Deze zuiveringstechniek is gebaseerd op de binding van eiwitten van de gastheercel en / of dan HP-NAP aan de hars verontreinigingen. Bij pH 8,0, zijn bijna geen andere eiwitten, behalve HP-NAP hersteld van de ongebonden fractie. Indien een eiwit gewenst voor zuivering geschikt voor ionenuitwisselingschromatografie als negatief, kunnen de beschreven protocol worden aangepast als uitgangspunt voor de ontwikkeling van een zuiveringswerkwijze.

one-step negative chromatographic purification of helicobacter pylori neutrophil-activating protein overexpressed in escherichia coli in batch mode

Helicobacter pylori-neutrofiel activerend eiwit (HP-NAP) is een belangrijke virulentiefactor Helicobacter pylori (H. pylori). Het speelt een cruciale rol in H. pylori geïnduceerde maagontsteking door activering van verschillende aangeboren leukocyten zoals neutrofielen, monocyten en mestcellen. De immunogeen is en immunomodulerende eigenschappen van HP-NAP maken het een potentiële diagnostische en kandidaat-vaccin voor H. pylori en een nieuw kandidaat-geneesmiddel voor de behandeling van kanker. Om grote hoeveelheden gezuiverde HP-NAP voor de klinische toepassingen te verkrijgen, een efficiënte werkwijze voor het zuiveren van dit eiwit met hoge opbrengst en zuiverheid tot stand worden gebracht. In dit protocol beschrijven we een werkwijze voor eenstaps negatieve chromatografische zuivering van recombinant HP-NAP overexpressie gebracht in Escherichia coli (E. coli) met behulp diethylaminoethyl (DEAE) ionenwisselaarharsen (bijv. Sephadex) in batch mode. Recombinant HP-NAP vormt bijna 70% van het totale eiwit in E. coli en bijna volledig hersteld in de oplosbare fractie na cellyse bij pH 9,0. Bij de optimale conditie bij pH 8,0, wordt het merendeel van HP-NAP gewonnen in de ongebonden fractie terwijl de endogene eiwitten van E. coli worden efficiënt verwijderd door de hars. Deze zuiveringsmethode met behulp van negatieve modus batch chromatografie met DEAE ionenwisselaars levert functionele HP-NAP uit E. coli in de natieve vorm met hoge opbrengst en zuiverheid. Het gezuiverde HP-NAP kan verder worden gebruikt voor de preventie, behandeling en prognose van H. pylori-geassocieerde ziekten en kanker.

Het schema van de experimentele procedure negatieve zuivering van recombinant HP-NAP expressie in E. coli door DEAE ionenuitwisselingsharsen in batchmodus is getoond in figuur 1.
