We presenteren een protocol voor de isolatie en flow cytometrische karakterisatie van small-intestinale lymfocyten, met inbegrip van de LPLs, IELs en lymfocyten in de PP. Voor diegenen die geïnteresseerd zijn in de evaluatie van hoe veranderingen in de microbiota van invloed op de dunne darm immuunsysteem, we detail de eenvoudige stappen die betrokken zijn bij de horizontale overdracht van organismen tussen de muizen die verschillende microbiotas. De belangrijkste taak van de dunne darm wordt vaak beschouwd als de vertering en opname van voedingsstoffen 1 zijn. Hoewel deze metabole functie duidelijk essentieel, heeft de dunne darm een even belangrijke rol in de gastheer te beschermen tegen de voortdurende versperring van antigenen uit de omgeving binnen het lumen 2. Het darmkanaal scheidt de buitenwereld (bv., Luminale antigenen) uit de interne omgeving van de gastheer met een epitheellaag dat slechts één cel laag dik. Als zodanig, de dunne darm immuunsysteem de enorme taak van een sluitende drempel reactiviteit, waardoor vreemde antigenen uit de voeding en commensale bacteriën aan het slijmvlies voeren met minimale of geen immuunrespons tijdens het monteren een robuuste respons tegen binnendringende pathogenen en andere &quot;schadelijk&quot; antigenen. Overmatige of ongepaste immuunreacties op deze antigenen kunnen leiden tot een pathologische aandoening (bv., Inflammatory darmziekte, type I diabetes, multiple sclerose), en moet worden vermeden 3-6. In het algemeen wordt het maagdarmkanaal beschouwd als de grootste immuun orgaan in het lichaam vertegenwoordigen, met meer dan 70% van antilichaam uitscheidende cellen 7. De kleine-intestinale immuunsysteem is opgebouwd uit 3 hoofdvakken - de lamina propria (LP), de intra-epitheliale laag en plaques van Peyer (PPS) - die elk bevat een opvallende groep van lymfocyten 2. De LP lymfocyten (LPLs) primair TCRαβ + T-cellen met ~ 20% B-cellen; intra-epitheliale lymfocyten (IELs) bevatten zeer weinig B-cellen meer TCRγδ + T cellen dan TCRαβ + T-cellen; en PP, die secundaire lymfoïde organen ingebed in de kleine darmwand zijn, bevatten ~ 80% B-cellen. Hoewel elk van deze anatomische gebieden heeft iets verschillende functies en ontologische bases, functioneren ze in aharmonized mode naar de host van pathogene beledigingen beschermen. Verder groeit de waardering die de microbiota een bepalende factor voor de ontwikkeling van de intestinale immuunsysteem, met toenemende erkenning van de verwante relatie tussen specifieke microben en de ontogenie van bepaalde cellijnen 8,9. Aangezien bovendien vorming van de intestinale immuunsysteem beïnvloedt immuunresponsen in anatomisch afstand gelegen plaatsen (bv. Artritis, multiple sclerose, pneumonie), is gebleken dat de ontwikkeling van de intestinale immuunsysteem meer ziekteprocessen relevant is dan eerder opgenomen 10 -12. Als zodanig heeft interesse in kwantitatief beoordelen van de intestinale immuunsysteem verlengd tot na gastheer-pathogeen interacties om nu ook gastheer-commensaal interacties en de pathogenese van vele systemische ziekten ook. Gezien de verschillen in de huidige methodenisolatie van intestinale lymfocyten, een methode die is geoptimaliseerd voor opbrengst, levensvatbaarheid en consistentie waarbij een evenwicht de benodigde tijd steeds belangrijker. Protocollen die Percoll gradiënten betrekken zijn tijd en arbeidsintensief en mogelijk meer kans op menselijke fouten, wat leidt tot variabele opbrengst en uitvoerbaarheid 13. Hierin geven we een geoptimaliseerd protocol voor de isolatie en karakterisering van lymfocyten uit alle 3 dunne darm immune compartimenten. Daarbij komt steeds meer belangstelling microbe-geïnduceerde veranderingen in het mucosale immuunsysteem, nemen we stappen die kunnen worden gebruikt om de horizontale overdracht van micro-organismen tussen muizen om te bepalen hoe deze veranderingen kwantitatief beïnvloeden de intestinale immuunsysteem.

De ingewanden - die het grootste aantal immuuncellen van elk orgaan in het lichaam te bevatten - worden voortdurend blootgesteld aan vreemde antigenen, zowel microbiële en dieet. Gegeven een beter begrip dat deze luminale antigenen meehelpen aan de immuunreactie en vorming van immuuncellen in de darm is essentieel voor een aantal systemische aandoeningen, is er toegenomen belangstelling voor het karakteriseren van de intestinale immuunsysteem. Echter, vele gepubliceerde protocollen lastig en tijdrovend. We presenteren hier een eenvoudige protocol voor de isolatie van lymfocyten uit de dunne darm lamina propria, intra-epitheliale laag en plaques van Peyer die snel, reproduceerbaar en vereist geen moeizame Percoll gradiënten. Hoewel het protocol staat de dunne darm, maar ook voor de analyse van de dikke darm. Bovendien zetten we enkele aspecten die extra optimalisatie kan nodig zijn afhankelijk van de specifieke wetenschappelijke wachtrijentie. Deze benadering resulteert in de isolatie van grote aantallen levensvatbare lymfocyten die vervolgens kan worden gebruikt voor flowcytometrische analyse of alternatieve wijze van karakterisering.

Flow cytometrische analyse van enkelvoudige celsuspensies van dunne darm lymfocyten zou een discrete populatie van cellen die soortgelijke voorwaartse en zijwaartse verstrooiing karakteristieken als splenocyten (Figuren 1A en 1B) hebben verkregen.
