Während der Schwangerschaft, die Plazenta Cytotrophoblastenzellen, die einkernigen Stammzellen sind, rasch vermehren und in entweder Villi oder extravillösen Cytotrophoblastenzellen unterscheiden. Extravillösen Cytotrophoblasten eindringen und die Spiralarterien der Uteruswand umbauen. Villöse Zytotrophoblasten, andererseits weiter zu vermehren, differenzieren und Sicherung kernige Synzytiotrophoblasten (Synzytium) 1 zu bilden. Die Aufrechterhaltung der Villi trophoblast Homöostase ist von wesentlicher Bedeutung für die fetale Wohlbefinden und gesunde Schwangerschaft. In der Tat erlauben villous Trophoblasten Mutter und Fötus Austausch von Sauerstoff und Nährstoffen und produzieren wesentliche Hormone für die Schwangerschaft. Diese klassische Technik, basierend auf sequentiellen enzymatischen Aufschluss mit Trypsin und DNase, gefolgt von der Isolierung in Dichtezentrifugation Medien (kolloidalen Silicapartikel durch Polyvinylpyrrolidon beschichtet oder Percoll) wird nun als Goldstandard zur Isolierung von Villi Cytotrophoblastenzellen erkannt. Die Technik kann durch eine magnetische Immun optimiert werden, ein Verfahren, das villöse Zytotrophoblasten aus nicht-trophoblastischen Zellen basierend auf der differentiellen Expression spezifischer Antigene auf den Oberflächen dieser Zellen trennt. Wir wählten das Human-Leukozyten-Antigen-ABC (HLA-ABC) wegen des Fehlens seiner Expression auf der Zell trophoblastic membrane 7,8. Die Plazenta ist ein Organ, das dramatische Veränderungen in der Sauerstoffspiegel während der Schwangerschaft erfährt. Im ersten Trimester ist die Sauerstoffanreicherungsverhältnis physiologisch sehr gering (2% O 2) , jedoch erhöht sich auf milde Ebenen der Oxygenierung (8% O 2) in der zweiten und dritten Trimester. Tuuli et al. 9 beschrieben , dass die In - vitro - Vermehrung des Trophoblasten Umgebung innerhalb des Plazentazotten ist eine Herausforderung und Variationen der Sauerstoffversorgung Ebenen sogar zu phänotypischen Veränderungen führen kann. Diese Bedingungen führen zu einer abnormen Erhöhung des oxidativen Stress, der Plazenta und fetale Lebensfähigkeit 13,14 kompromittiert. Die Veränderungen , die Trophoblasten - Zellen in vivo während Episoden von Hypoxie / Reoxygenierung laufen kann in vitro nachgeahmt werden , wie folgt: villöse Zytotrophoblasten unter normoxischen Bedingungen (8% O 2) gehalten werden , bis sie in Synzytiotrophoblasten unterscheiden. Sie werden dann in hypoxischen Bedingungen (0,5% O 2) für 4 Stunden, gefolgt von einer weiteren 18 h Normoxie (Reoxygenierung) unterzogen. Mit dieser Hypoxie / Reoxygenierung Ansatz, Trophoblasten exhibit dereguliert Redoxzustand und erhöhte Spiegel von intrinsischen Apoptose 8, wie in bestimmten Komplikationen während der Schwangerschaft beobachtet. Daher ist dies ein in vitro - Modell nützliche neue präventive und therapeutische Ansätze zu bewerten Schwangerschaftskomplikationen mit plazentalem Hypoxie / Reoxygenierung verbunden zu bekämpfen. Plazentalen Zellen produzieren Melatonin, das mehrere wichtige Funktionen, wie beispielsweise eine Fähigkeit , 15 oxidativem Stress und Plazentadysfunktion zu vermeiden. human primary trophoblast cell culture model to study the protective effects of melatonin against hypoxia/reoxygenation-induced disruption

This protocol describes how villous cytotrophoblast cells are isolated from placentas at term by successive enzymatic digestions, followed by density centrifugation, media gradient isolation and immunomagnetic purification. As observed in vivo, mononucleated villous cytotrophoblast cells in primary culture differentiate into multinucleated syncytiotrophoblast cells after 72 hr. Compared to normoxia (8% O2), villous cytotrophoblast cells that undergo hypoxia/reoxygenation (0.5% / 8% O2) undergo increased oxidative stress and intrinsic apoptosis, similar to that observed in vivo in pregnancy complications such as preeclampsia, preterm birth, and intrauterine growth restriction. In this context, primary villous trophoblasts cultured under hypoxia/reoxygenation conditions represent a unique experimental system to better understand the mechanisms and signalling pathways that are altered in human placenta and facilitate the search for effective drugs that protect against certain pregnancy disorders. Human villous trophoblasts produce melatonin and express its synthesizing enzymes and receptors. Melatonin has been suggested as a treatment for preeclampsia and intrauterine growth restriction because of its protective antioxidant effects. In the primary villous cytotrophoblast cell model described in this paper, melatonin has no effect on trophoblast cells in normoxic state but restores the redox balance of syncytiotrophoblast cells disrupted by hypoxia/reoxygenation. Thus, human villous trophoblast cells in primary culture are an excellent approach to study the mechanisms behind the protective effects of melatonin on placental function during hypoxia/reoxygenation.
