the terroir concept interpreted through grape berry metabolomics and transcriptomics

Terroir se réfère à la combinaison de facteurs environnementaux qui influent sur les caractéristiques des cultures telles que la vigne (Vitis vinifera) selon des habitats particuliers et les pratiques de gestion. Cet article montre comment certaines signatures du terroir peuvent être détectés dans le métabolome de baies et de transcriptome du Corvina vigne cultivar en utilisant une analyse statistique multivariée. La méthode nécessite d&#39;abord un plan d&#39;échantillonnage approprié. Dans cette étude de cas, un clone spécifique du cultivar Corvina a été choisi pour minimiser les différences génétiques, et des échantillons ont été prélevés dans sept vignobles représentant trois macro-zones différentes au cours de trois saisons de croissance différents. Une LC-MS approche untargeted métabolomique est recommandé en raison de sa sensibilité élevée, accompagnée d&#39;un traitement efficace des données en utilisant un logiciel MZmine et une stratégie d&#39;identification des métabolites basée sur l&#39;analyse de l&#39;arbre de la fragmentation. l&#39;analyse du transcriptome global peut être réalisé en utilisant microarraysLes sondes contenant couvrant ~ 99% de tous les gènes de la vigne prédites, ce qui permet l&#39;analyse simultanée de tous les gènes exprimés de manière différentielle dans le contexte des différents terroirs. Enfin, l&#39;analyse des données multidimensionnelles sur la base de méthodes de projection peut être utilisé pour surmonter l&#39;effet fort spécifiques vintage, permettant aux métabolomique et les données de transcriptomique à être intégrées et analysées en détail pour identifier les corrélations informatives.

L&#39;étude de cas décrit dans cet article a donné une matrice de données finale comprenant 552 signaux (m / z caractéristiques) , y compris des ions moléculaires ainsi que leurs isotopes, adduits et quelques fragments, relativement quantifiés parmi 189 échantillons (7 vignobles x 3 étapes de maturation x 3 saisons de x croissance 3 répétitions biologiques).
