Polymorphkernigen Leukozyten (PMNs, die auch als Granulozyten) und Monozyten sind wichtige zelluläre Komponenten des angeborenen Immunsystems im Blut 1,2. Sie tragen zur primäre Abwehr von Krankheitserregern sowie zur Aktivierung des erworbenen Immunsystems und die Einleitung einer systemischen Entzündungsreaktion 2-4. Doch vor allem Neutrophile, die am häufigsten vorkommende Art von Granulozyten und Monozyten tragen auch wesentlich zur Regulierung und die Beendigung der akuten entzündlichen Ereignisse 5. Daher können diese Zellen auch eine wichtige Rolle bei chronisch - entzündlichen Erkrankungen wie rheumatoider Arthritis 6,7 spielen. In der Tat, Asthma, eine chronische entzündliche Erkrankung der Atemwege, wird durch eine beeinträchtigte Apoptose von Eosinophilen gekennzeichnet, die zweithäufigste Granulozyten Typ im Blut 8. Doch die Apoptose von Granulozyten und deren schnelle Beseitigung durch Makrophagen sind zwei wesentliche Schritte, die während der zellulären BeendigungEntzündungs ​​11.09. In den genannten Immunzellen zwei eng verwandte Enzyme, nämlich Myeloperoxidase (MPO, Neutrophilen und Monozyten) und eosinophile Peroxidase (EPO, Eosinophilen) können 12,13 gefunden werden. Diese Häm Peroxidasen klassisch sind mit der humoralen Immunantwort , wie sie im Zusammenhang mit zwei elektronisch zu oxidieren (pseudo-) -halogenide zu dem entsprechenden hypo (pseudo) halogenhaltige Säuren , die für ihre bakterizide Eigenschaften 14-16 bekannt sind. Unter physiologischen Bedingungen MPO hauptsächlich Formen Hypochlorsäure (HOCl) und hypothiocyanite (- OSCN) , während die letztere und Hypobromsäure (HOBr) durch EPO 17-19 gebildet werden. Neue Ergebnisse deuten darauf hin , dass diese (pseudo-) Halogenierung Enzymaktivität auch für die Regulierung der Entzündungsreaktionen und zur Beendigung von Immunreaktionen 20,21 beitragen. In der Tat wurden die HOCl Produktion von MPO und daraus abgeleitete Produkte gezeigt T - Zell-basierte adaptive Immunantwort 22-2 zu unterdrücken4. Um weitere Einblicke in die immunologischen Rolle der Leukozyten aus dem angeborenen Immunsystems bei chronischen Entzündungskrankheiten zu gewinnen und den Beitrag von MPO und EPO dieser physiologischen Funktion zu bestimmen, haben wir ein Verfahren, um schnell Leukozyten aus kleinen Blutproben für einen nachfolgenden spezifischen reichern Bestimmung der Halogenierung Peroxidase-Aktivität in diesen Zellen. Für Erythrozyten-Depletion haben wir ein standardisiertes Verfahren mit zwei nachfolgenden hypotone Lyse Schritte mit destilliertem Wasser ausgewählt, die bei niedrigen Materialkosten zu einer schnellen Leukozyten-Anreicherung führt. Für die anschließende Bestimmung der Halogenierung MPO und Aktivität EPO die HOCl- und HOBr-spezifischen Farbstoff aminophenyl Fluorescein (APF) wurde 25-27 verwendet. Im Gegensatz zu der Anwendung von unspezifischen Peroxidase Färbemethoden 28,29, ermöglicht dieser Ansatz die selektive Erfassung der Halogenierung Peroxidase - Aktivität, die häufig bei schweren infl beeinträchtigt wirdammation 30,31.

In diesem Beitrag wird ein Protokoll für die schnelle und standardisierte Anreicherung von Leukozyten aus kleinen Vollblut-Proben beschrieben. Dieses Verfahren wird auf der hypotonischen Lyse der Erythrozyten basieren und kann als an Blut von nicht-menschlichen Ursprungs sowie für die menschliche Proben angewendet werden. Die geringe anfängliche Probenvolumen von etwa 50 bis 100 &amp; mgr; l macht dieses Verfahren für wiederkehrende Blutentnahme von kleinen Labortieren. Darüber hinaus wird Leukozyten Anreicherung innerhalb von Minuten und mit geringem Material Anstrengungen in Bezug auf Chemikalien und Instrumentierung erreicht, ist dieses Verfahren anwendbar in mehreren Laborumgebungen. Standardisierte Reinigung von Leukozyten wird mit einem hochselektiven Färbeverfahren kombiniert Halogenierung Peroxidaseaktivität von Häm - Peroxidasen, Myeloperoxidase (MPO) und eosinophile Peroxidase (EPO), dh die Bildung von unterchloriger und hypobromige Säure (HOCl und HOBr) auszuwerten. Während MPO stark in neut ausgedrücktrophils, die häufigste Immunzelltyp im menschlichen Blut sowie in Monozyten, die verwandten Enzym EPO wird ausschließlich in Eosinophilen exprimiert wird. Die Halogenierung Aktivität dieser Enzyme wird unter Verwendung der fast HOCl- und HOBr-spezifischen Farbstoff aminophenyl fluorescein (APF) und den primären Peroxidasesubstrat Wasserstoffperoxid behandelt. Bei der anschließenden Durchflusscytometrieanalyse alle Peroxidase-positive Zellen (Neutrophile, Monozyten, Eosinophilen) unterscheidbar und ihre Halogenierung Peroxidase-Aktivität quantifiziert werden können. Da APF Färbung kann mit der Anwendung von Zelloberflächenmarkern kombiniert werden, kann dieses Protokoll speziell verlängert werden, um Leukozyten-Unterfraktionen adressieren. Das Verfahren ist für HOCl und HOBr Produktion sowohl in der Human- und in Nagetier Leukozyten zu erkennen. Angesichts der breiten und mannigfaltig diskutiert immunologische Rolle dieser enzymatischen Produkte in chronisch-entzündlichen Erkrankungen kann dieses Protokoll zu einem besseren Verständnis des beitragenimmunologischer Relevanz von Leucocyten abgeleitetes Häm Peroxidasen.

Wie zuvor berichtet , oben gedreht beschriebene Verfahren geführt anwendbar sowohl für die menschliche zu sein und nicht menschlichen Material 32. Außerdem, wie für die Mäuse, die mit asthmatischen Symptomen der Färbung APF gezeigt ist, kann ein geeignetes Werkzeug sein, um Unterschiede in der systemische proinflammatorische Zustand erkennen. Fast and Specific Assessment of the Halogenating Peroxidase Activity in Leukocyte-enriched Blood Samples

This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. The method can be applied to human and non-human material and may contribute to the evaluation of new inflammatory markers.

The overall goal of this erythrocyte depletion method is to achieve a quick and simple leukoctye-enrichment from a small blood sample. Many advantages of this technique are that it is cheap, reliable, and fast. Moreover, it can be applied to human and non-human blood samples.Before beginning the lysis procedure, adjust the pipette for water administration to two milliliters, and the pipette for PBS to five milliliters. Then place open flasks of water and PBS under a laminar flow hood at room temperature. As the hypotonic lysis is a time-critical process, everything should be prepared previously.Moreover, the lysis proceudre should always be done in the same way, to have compatible results. Next, transfer 100 microliters from each blood sample into a 15 milliliter conical tube. And add two milliliters of distilled water to each sample with vortexing at the medium setting.Rest the cells for 60 seconds, then add five milliliters of PBS. And mix the samples by vortexing. Now, collect the cells by centrifugation, and decant the supernatants.Invert the tubes and tap the openings on a paper towel to remove as much liquid as possible. Then repeat the hypotonic lysis procedure as just demonstrated. After obtaining a dry pellet for the second time, re-suspend each sample in 500 microliters of HBSS, until a homogeneous solution is obtained.And transfer the cells into 1.5 milliliter, micro-centrifuge tubes on ice. To check the peroxidase specificity of the samples after amino phenol fluorescein, or APF staining, first add five microliters of the heme peroxidase inhibitor, 4-ABAH, to the appropriate samples for a 15 minute incubation at 37 degrees Celsius. Next, add five microliters of APF working solution to each tube, and gently vortex the cells on the medium setting.Incubate all of the samples at 37 degrees Celsius for 30 minutes. Then add five microliters of hydrogen peroxide working solution to each sample, and gently vortex the tubes. After 60 minutes at 37 degrees Celsius, centrifuge the cells, and carefully remove the supernatant without disturbing the pellets.Finally, re-suspend the cells in 250 microliters of HBSS, and store the samples in the dark until their analysis, to avoid photo bleaching. Here, a representative example of the leukocyte populations after hypotonic lysis and APF staining as just demonstrated is shown. By plotting cell size versus granularity, the robust depletion of the erythrocytes can be observed, with the red blood cell and debris population accounting for only 22%of the events.Further, the intensity distribution of the APF-derived fluorescence facilitates a clear discrimination of the peroxidase-negative erythrocites and lymphocytes, with a moderate APF oxidation observed, for the peroxidase-positive monocytes and eosinophils, and a robust peroxidase oxidation exhibited by the neutrophils. Once mastered, this technique can be applied to up to 50 blood samples in parallel if it is performed properly. Following this procedure, other methods like antibody staining can be performed to identify the cell subsets of interest.After its development, this technique paved the way for researchers in the field of small animal studies to explore the role of halogenating activity of MPO and EPO in animal models for chronic inflammatory diseases.
