Staphylocoques (S. aureus) est connu comme le pathogène le plus important dans le monde responsable de l&#39; infection intra - mammaire (IMI) chez les bovins 1. L&#39;étude menée par Fournier et al. 2 démontré chez les vaches suisses, les études réalisées par Cosandey et al. 3 et Boss et al. 4 vaches de 12 pays européens que S. aureus isolé de bovin IMI est un groupe génétiquement hétérogène. Par amplification par PCR de la région d&#39;espaceur intergénique 16S-23S ARNr (RS-PCR), un total de 17 génotypes ont été initialement détecté dans 101 isolats épidémiologiquement indépendants 2. Génotype B et le génotype C (GTC) étaient les plus fréquentes (80%), tandis que les 15 autres génotypes (SMT) se sont produits rarement, chaque prise de 1 à 4% de tous les isolats. Au niveau européen 3, GTB, GTC, GTF, GTI et GTR étaient les génotypes les plus importants , comprenant 76% de toutes les souches analysées (n = 456). GTB était situé en Europe centrale wconsidérant que les autres génotypes ont été largement diffusés. Les 24% restants des souches inclus 41 génotypes, beaucoup d&#39;entre eux, cependant, ont été observés une seule fois. Les études menées par Fournier et al. 2, Cosandey et al. 3, et Graber et al. 5 ont démontré en outre que les génotypes étaient fortement associés à leur modèle de gène de virulence, ainsi que le suisse au niveau européen. Les autres études ont révélé des différences massives dans la prévalence IMI chez S. aureus GTB, GTC et les autres génotypes 2,5: compte tenu de GTB, jusqu&#39;à 87% des vaches dans un troupeau ont été infectées par ce génotype. Dans le cas du GTC et des autres génotypes, cependant, l&#39;IMI a été trouvé que dans 1 à 2 vaches d&#39;un troupeau. IMI causée par S. aureus, se traduit normalement par une inflammation des glandes mammaires correspondant et une augmentation des cellules inflammatoires dans le lait. En conséquence, la coopération de cellules somatiquesunts dans le lait (SCC), l&#39;indicateur clé de la qualité du lait dans la plupart des pays, est augmenté conduisant à un prix du lait a diminué ou même l&#39;arrêt de la livraison. En plus de l&#39;interface RS-PCR de Fournier et al. 2, d&#39; autres méthodes de typage ont été décrites pour le sous - typage des souches bovines de S. aureus 8-11. Deux plus communs comprennent le typage spa 12 et Multi Locus Sequence Typing (MLST) 13. L&#39;ancien est basée sur le séquençage d&#39; ADN de la région d&#39;espacement variable du gène codant spa pour la protéine A staphylococcique, alors que celui - ci nécessite une séquencement de 7 gènes de ménage. Par rapport à RS-PCR, ces méthodes nécessitent un effort supplémentaire considérable dans le laboratoire résultant en un débit d&#39;échantillons faibles et des coûts élevés. lele débit est particulièrement faible pour les ECP. Compte tenu de la résolution de ces méthodes pour les souches bovines de S. aureus, RS-PCR est au moins aussi bon que spa tapant 4 et mieux que MLST 4 ou ECP 15. Toutes ces méthodes , y compris la dactylographie binaire 13 ont démontré leur efficacité dans l&#39; obtention d&#39; un éclairage supplémentaire sur la pathogenèse de bovins IMI causée par S. aureus, mais à cause des restrictions mentionnées ils sont moins appropriés pour les grandes investigations cliniques. En outre, le génotypage par RS-PCR comme décrit par Fournier et al. 2 permet la prédiction fiable de l&#39;épidémiologie et le potentiel pathogène de S. aureus impliqué dans l&#39; IMI bovine, deux valeurs clés de la médecine vétérinaire clinique.

genotyping of staphylococcus aureus by ribosomal spacer pcr (rs-pcr)

The ribosomal spacer PCR (RS-PCR) is a highly resolving and robust genotyping method for S. aureus that allows a high throughput at moderate costs and is, therefore, suitable to be used for routine purposes. For best resolution, data evaluation and data management, a miniaturized electrophoresis system is required. Together with such an electrophoresis system and the in-house developed software (freely available here) assignment of the pattern of bands to a genotype is standardized and straight forward. DNA extraction is simple (boiling prep), setting-up of the reactions is easy and they can be run on any standard PCR machine. PCR cycling is common except prolonged ramping and elongation times. Compared to spa typing and Multi Locus Sequence Typing (MLST), RS-PCR does not require DNA sequencing what simplifies the analysis considerably and allows a high throughput. Furthermore, the resolution for bovine strains of S. aureus is at least as good as spa typing and better than MLST or pulsed-field gel electrophoresis (PFGE). The RS-PCR data base includes presently a total of 141 genotypes and variants. The method is highly associated with the virulence gene pattern, contagiosity and pathogenicity of S. aureus strains involved in bovine mastitis. S. aureus genotype B (GTB) is contagious and causes herds problems causing large costs in the Switzerland and other European countries. All the other genotypes observed in Switzerland infect individual cows and quarters. Genotyping by RS-PCR allows the reliable prediction of the epidemiological and the pathogenic potential of S. aureus involved in bovine intramammary infection (IMI), two key factors for clinical veterinary medicine. Because of these beneficial properties together with moderate costs and a high sample throughput the goal of this publication is to give a detailed, step-by-step protocol for easily establishing and running RS-PCR for genotyping S. aureus in other laboratories.
