Ultra-strukturelle og sub-organelle detaljer om cellene har blitt hovedsakelig avslørt av transmisjonselektronmikroskopi (TEM) studier fra vev eller celle pellets 1,2. Solid biter av vev kan lett utnyttes for elektronmikroskopi studier. Men analyser TEM 1,3 på celler i suspensjon fortsatt utfordrende og nødvendig høye celletall, dvs. millioner av celler. På grunn av dette, ultra-strukturelle analyser av sjeldne cellepopulasjoner i suspensjon, f.eks, hematopoetiske stamceller (HSCs), er ikke lett vurderes. Flere forsøk for TEM-analyse fra knappe eksempler ved å bruke cytospin eller monolagsfremgangsmåter ikke klarer å få vesentlig kvantitative data fra et begrenset antall celler. Dermed blir kravet om et høyt celle nummer begrenser bruken av dette kraftige verktøyet for å forstå subcellulære ultra detaljer om sjeldne celle populasjoner. En viktig begrensning for TEM-studier med et begrenset antall celles i suspensjon er lokalisering av cellene for behandling og dermed TEM-studier fra begrensede celler med særlig små størrelser forbli utfordrende. Flere alternative tilnærminger har blitt vedtatt for å motvirke denne begrensningen: BSA / bisacrylamide (BSA-BA) mediert polymerisasjon av cellesuspensjon, farging av celler med et fargestoff for å gjøre dem synlige på en tynn film-støtte, inkludert Dekk, og TEM analyser fra Cytospin forberedelser 4-6. Men svært begrenset suksess ble oppnådd, som svært få celler ble funnet i avsnittene etter ultra-seksjonering. Hovedutfordringen med å identifisere spredte celler vedvarer fordi cellemonolaget forblir stort sett usynlig i størknet gel for seksjonering. Videre kan cytospin fremstilling av celler forandre deres cellulære organisasjon på grunn av celle spredning og skjørheten av cellestrukturen. Derfor er disse iboende ulemper garanterer en ny tilnærming for å utføre TEM studier av sjeldne cellepopulasjoner med merkonsistens. For å bøte på dette problemet, har vi beskrevet en ny og alternativ prøveopparbeidelse metode for TEM-studier fra sjeldne celle populasjoner 7. Her rapporterer vi en effektiv prøveopparbeidelse protokollen for å utføre TEM fra knappe biologiske prøver med konsekvent kvalitative og kvantitative resultater. Evans blue farging ble utført etter fiksering for å lokalisere en liten celle pellet fra lavt antall celler, dvs. 10 000 benmarg blodkreft stilk og stamceller som ellers ville ha forblitt usynlig, og pelleten ble innebygd i agarose før dehydrering og harpiks innebygging prosesser. Denne fremgangsmåte klynger cellene sammen og muliggjør effektiv analyse av ultra og subcellulære organisering av hematopoetiske stamceller (identifisert som Lin - Sca-1 + c-kit + Flt3 - CD34 -; HSC), en sjelden 0,2 til 0,5%-cellepopulasjonen i benmargen. Dette eksperimentell proprotokollen kan være nyttig å utføre ultrastrukturstudier og få kvantitative resultater på mange sjeldne, men svært viktige bestander.

Transmisjonselektronmikroskopi (TEM) gir detaljer om den cellulære organisasjon og ultrastruktur. Men TEM analyse av sjeldne celle populasjoner, spesielt celler i suspensjon som blodkreft stamceller (HSCs), fortsatt begrenset på grunn av kravet om et høyt celle nummer under tillagingen. Det er noen cytospin eller monolags tilnærminger for TEM-analyse fra knappe prøver, men disse metodene ikke klarer å få signifikante kvantitative data fra et begrenset antall celler. Her er en alternativ og ny tilnærming for prøveopparbeidelse i TEM studier beskrevet for sjeldne celle populasjoner som gjør at kvantitativ analyse. En forholdsvis lav cellenummer, dvs. 10000 HSCs, ble med hell anvendt for TEM-analyse i forhold til de millioner av celler som vanligvis brukes for TEM-studier. Spesielt ble Evans blå flekker utført etter paraformaldehyde-glutaraldehyd (PFA-GA) fiksering for å visualisere den lille cellen pella, som tilrettelagt embedding i agarose. Klynger av mange celler ble observert i ultra-tynne snitt. Cellene hadde et godt bevart morfologi, og det ultra-strukturelle detaljer av Golgi-komplekset og flere mitokondrier var synlig. Dette effektiv, enkel og reproduserbar protokollen tillater prøvepreparat fra en lav cellenummer, og kan brukes til kvalitativ og kvantitativ analyse TEM på sjeldne cellepopulasjoner fra begrensede biologiske prøver.

En effektiv og konsistent protokoll for prøvepreparering for å utføre TEM-analyse fra lavt antall celler er beskrevet. Post-fiksering farging med Evans blå og overføring av cellene til et 0,5 ml mikrosentrifugerør hjalp visualisere det lille cellepelleten 7.
