Although we have previously reported a similar procedure to capture live images of the gastrulation process in Drosophilla1, the method we describe here is detailed and easy to trace endogenous cadherin expression and thus is quite useful for genetic screening of key factors involved in gastrulation. Gastrulação é o primeiro conjunto de eventos morfologicamente dinâmicas que ocorrem durante o desenvolvimento embrionário de animais multicelulares, tais como Drosophila 1,2. Curiosamente, a evidência emergente sugere que este processo é regulado através da interação entre os mecanismos mecânicos e moleculares 3. Além disso, o epitelial para mesenquimal (EMT), que é um processo crucial para a gastrulação, também está implicado em processos de doenças humanas, tais como metástases de cancro 4-8. Como tal, muitos genes que controlam a constrição apical também são conhecidos por serem factores fundamentais para o EMT observado na metástase do câncer 9. Gastrulação é o primeiro conjunto de eventos morfologicamente dinâmicas que ocorrem durante o desenvolvimento embrionário de animais multicelulares, tais como Drosophila. Esta alteração morfológica é também reconhecido como epitelial para mesenquimal (EMT). A desregulação de EMT está associada com fibrose e metástases do cancro. Há uma evidência emergente de que EMT é controlado por um número de mecanismos moleculares. Como tal, muitos genes-chave que controlam a constrição apical também são conhecidos por serem fatores importantes na EMT observado na metástase do câncer. Como EMT durante Drosophila gastrulação, as células epiteliais podem ser induzidas a alterar a sua forma e ser reprogramadas de modo a redireccionar o destino celular no sentido de vários outros tipos de células. Aqui nós fornecemos um método de imagem robusta de Drosophila gastrulação para ensaiar o início de movimentos celulares morfogenéticos e identificação destino celular durante esta fase do desenvolvimento embrionário. Usando este método, identificamos cEll rearranjo no momento da gastrulação e demonstrar a importância de constrição apical durante a gastrulação utilizando GFP marcado DE-caderina.
