Le choix d&#39;un modèle animal d&#39;épilepsie possible est essentiel pour chaque but expérimental. L&#39;un de nos objectifs est d&#39;élucider les effets de EA sur la suppression de l&#39;épilepsie. EA est une médecine alternative qui peut présenter un effet thérapeutique dans l&#39;épilepsie et a été documentée dans la littérature chinoise ancienne. L&#39;épilepsie est un trouble neurologique commun dans lequel des crises récurrentes se produisent tout au long de la durée de vie d&#39;un patient. La plupart des récurrences d&#39;épilepsie peuvent être bien contrôlés par les médicaments anti-épileptiques (DEA). Toutefois, environ 30% des patients atteints d&#39; épilepsie réfractaire , l&#39; épilepsie développent 1. causes de l&#39;épilepsie, troubles du sommeil qui peut encore aggraver la récidive. La preuve démontre que l&#39; épilepsie peut soit perturber le sommeil la nuit ou peut provoquer une somnolence diurne excessive 2,3. Nos études antérieures indiquent en outre que l&#39; épilepsie se produisant à zeitgeber temps (ZT) 0, soit le début de la période de lumière à la lumière: cycle d&#39; obscurité, diminue le sommeil; ceci est médiée par l&#39;hormone de libération de la corticotropine (CRH), un facteur homéostatique. L&#39;épilepsie chez ZT13 (le début de la période d&#39;obscurité) augmente l&#39;expression d&#39;un autre facteur homéostatique, l&#39;interleukine-1 (IL-1), ce qui augmente le sommeil. Sommeil rythmes circadiens sont modifiés lorsque l&#39;épilepsie survient à ZT6, au milieu de lapériode de lumière 4,5. D&#39;autre part, les problèmes de sommeil aggravent encore la progression et la récurrence de l&#39; épilepsie 6. Sur la base des éléments de preuve ci-dessus, nous essayons de révéler une méthode thérapeutique optimale pour contrôler simultanément l&#39;épilepsie et prévenir les perturbations du sommeil chez les patients épileptiques. Nous avons déjà constaté que électroacupuncture (EA) avec une fréquence de stimulation de 10 Hz, dans lequel une certaine quantité de courant est délivré dans le acupoint à travers une aiguille en acier inoxydable, électrocorticogramme supprime avec succès (EcoG) activités d&#39; épilepsie et des troubles du sommeil induit par l&#39; épilepsie-7 . EA avec une fréquence de stimulation de 100 Hz se détériore davantage les activités d&#39; épilepsie et les troubles du sommeil chez le rat 8,9. Cette expérience réussie dépend de trois facteurs: d&#39;une part, un modèle animal d&#39;épilepsie possible; en second lieu, un procédé d&#39;enregistrement et d&#39;analyse du sommeil chez les rongeurs; et troisièmement, la performance exacte de l&#39;acupuncture et la précision de la locati acupointons. L&#39;épilepsie a été classé en deux grands types: épilepsie focale et l&#39;épilepsie généralisée. Nous sommes intéressés à focale épilepsie temporale de lobe (TLE), épilepsie généralisée, état de mal épileptique (SE), et la récurrence de l&#39;épilepsie généralisée spontanée. Par conséquent, les différentes manipulations sont appliquées pour créer des modèles d&#39;épilepsie appropriés pour nos expériences. Pour établir TLE focale, une faible dose de pilocarpine est administré dans le noyau central de l&#39;amygdale gauche (ACÉ). Pour vérifier ce modèle, six électrodes ECoG sont implantées sur le frontal (F1 et F2), pariétal (P1 et P2), et occipital (O1 et O2) lobes dans les deux hémisphères gauche et droit, et deux électrodes de référence (R1 &amp; R2) sont placés sur le cervelet, dans les deux hémisphères. Un montant supplémentaire de guide de microinjection canule est implanté chirurgicalement dans le aCÉ gauche (AP, 2,8 mm de bregma; ML, 4.2 mm, DV, 7,8 mm par rapport à bregma). Les coordonnées sont adaptées de la Paxinos et Watson atlas de rat 10. Si le TLE focal est induite avec succès, seul l&#39;enregistrement à partir de l&#39;électrode à gauche cortex pariétal (P1), qui est près de la aCÉ gauche, devrait acquérir les ECoGs épileptiformes dominantes, sans ECoGs épileptiformes significatives enregistrées à partir des autres électrodes ECoG. Intrapéritonéale (IP) des injections de pilocarpine dans les rats induisent l&#39;épilepsie et SE généralisés, mais cela peut être fatal. Cinq injections IP de pentylenetetrazol (PTZ) avec un intervalle d&#39;une journée entre chaque injection induisent avec succès l&#39;épilepsie généralisée spontanée chez la souris et assurent également la survie des souris. Deux électrodes de fil ECoG sont implantés dans les frontales et pariétales corticales chez les souris pour recevoir des signaux ECoG et de vérifier l&#39;épilepsie spontanément récurrente. Polysomnographie (PSG) est une méthode complète pour enregistrer les changements physiologiques qui se produisent pendant le sommeil, et il peut objectivement classer le sommeil en différentes étapes du mouvement non-rapide oeil (NREM) et rmouvements oculaires apid (REM) du sommeil. PSG enregistre les paramètres de fonctionnement de l&#39;organisme, y compris les ondes cérébrales (électroencéphalogramme, EEG), les mouvements oculaires (de électrooculogramme, EOG), des tons de muscle squelettique (de électromyogramme, EMG), les rythmes cardiaques (électrocardiogramme, ECG), et les niveaux d&#39;oxygène dans le sang et les paramètres respiratoires. Chez les rats, nous enregistrons ECoGs, EMGs, température corticale, et l&#39;activité locomotrice de classer les états de vigilance en éveil, le sommeil lent et le sommeil paradoxal. analyse du sommeil chez la souris est effectuée en utilisant ECoGs, EMGs, et les résultats de l&#39;activité locomotrice. Les rats sont implantés chirurgicalement avec trois EcoG vis électrodes au frontal, pariétal et corticales cérébelleux controlatéral par la chirurgie stéréotaxique. détermination post-acquisition des états de vigilance (de l&#39;éveil, le sommeil lent et le sommeil paradoxal) est conduite selon les paramètres acquis des ECoGs, EMG, la température du cerveau, et l&#39;activité locomotrice. Les rats et les souris ont besoin d&#39;être anesthésiés avec une faible dose de Zoletil (25 mg / kg), soit la moitié de la dose d&#39;anesthésique normalement administrés pendant la chirurgie stéréotaxique, avant d&#39;effectuer l&#39;acupuncture manuelle ou EA. Ce dosage permet aux animaux de se réveiller 30 à 35 minutes après l&#39;injection. Soit l&#39;acupuncture manuelle ou EA est effectuée au début de la période d&#39;obscurité, avec un intervalle de temps constant de 30 minutes, et chaque animal est consécutivement traités pendant deux à trois jours. Stimuler les courants EA sont livrés dans un acupoint particulier à travers une aiguille en acier inoxydable qui est inséré dans l&#39;acupoint. Le courant de stimulus est un train d&#39;impulsions carrées biphasique, dans lequel la durée d&#39;impulsion est de 150 ms et l&#39;intensité de stimulation est de 1 mA. Si une aiguille sèche est utilisée pour l&#39;acupuncture manuelle, l&#39;aiguille insérée dans les acupoints est crispa 10 fois toutes les 5 min. La partie difficile de l&#39;acupuncture manuelle ou EA est de localiser les points d&#39;acuponcture chez les rongeurs. Le location des acupoints chez des rats ou des souris est similaire à leur localisation anatomique chez les humains. Par exemple, les acupoints bilatéraux Fengchi sont situés 3 mm de la ligne médiane postérieure sur le cou, entre les deux oreilles, ce qui est similaire à son emplacement anatomique chez les humains 11. En outre, les points d&#39;acupuncture avec une faible impédance sur la peau peuvent être confirmées. la manipulation de l&#39;acupuncture ou fictive EA Sham est nécessaire pour l&#39;acupuncture ou des expériences EA. Ancient Chinese literature has documented that acupuncture possesses efficient therapeutic effects on epilepsy and insomnia. There is, however, little research to reveal the possible mechanisms behind these effects. To investigate the effect of acupuncture on epilepsy and sleep, several issues need to be addressed. The first is to identify the acupoints, which correspond between humans, rats, and mice. Furthermore, the depth of insertion of the acupuncture needle, the degree of needle twist in manual needle acupuncture, and the stimulation parameters for electroacupuncture (EA) need to be determined. To evaluate the effects of acupuncture on epilepsy and sleep, a feasible model of epilepsy in rodents is required. We administer pilocarpine into the left central nucleus of the amygdala (CeA) to simulate focal temporal lobe epilepsy (TLE) in rats. Intraperitoneal (IP) injection of pilocarpine induces generalized epilepsy and status epilepticus (SE) in rats. Five IP injections of pentylenetetrazol (PTZ) with a one-day interval between each injection successfully induces spontaneous generalized epilepsy in mice. Recordings of electrocorticograms (ECoGs), electromyograms (EMGs), brain temperature, and locomotor activity are used for sleep analysis in rats, while ECoGs, EMGs, and locomotor activity are employed for sleep analysis in mice. ECoG electrodes are implanted into the frontal, parietal, and contralateral occipital cortices, and a thermistor is implanted above the cerebral cortex by stereotactic surgery. EMG electrodes are implanted into the neck muscles, and an infrared detector determines locomotor activity. The criteria for categorizing vigilance stages, including wakefulness, rapid eye movement (REM) sleep, and non-REM (NREM) sleep are based on information from ECoGs, EMGs, brain temperature, and locomotor activity. Manipulation of Epileptiform Electrocorticograms ECoGs and Sleep in Rats and Mice by Acupuncture

This paper demonstrates the performance of acupuncture, epilepsy models, and the analysis of sleep in rodents. The acupuncture procedure and the identification of acupoints are described. Pilocarpine or pentylenetetrazol (PTZ) is used to induce epilepsy. Electrocorticogram (ECoG), electromyogram (EMG), brain temperature, and locomotor activity recordings are employed for sleep analysis.

The overall goal of this procedure is to assess epilepsy, sleep and the effects of acupuncture in an animal model. This method can help answer key questions in neuroscience field about how to establish epilepsy, analyze sleep and perform acupuncture in a small animal model. The main advantages of this technique are that the lateral cortical and the lateral myogram electrodes in that semester can be implanted quickly and easily by stereotaxic surgery.Demonstrating this procedure will be Yi-Jou Wu, a graduate student from my laboratories. After confirming the proper depth of sedation by a lack of response to a toe pinch, apply eye ointment to a six-to eight-week-old 250 to 350 gram Spregg Dolly rat from the first group. Use clippers to remove the hair from the top of the skull.Disinfect the skin with sequential applications of povidone iodine solution and 75 percent ethanol, followed by antibiotic injection to prevent infection. Next, mount the animal onto a stereotaxis machine and then place an ear bar into the ear canal. Using a scalpel, make an approximately two centimeter midline caudal incision along the skull between the ears.Then use hemostats to clip the skin flaps, exposing the skull, and use a scalpel to remove the tissue. When the skull is clean, use a rotary tool to drill eight approximately 0.7 millimeter diameter holes into the frontal, parietal, and occipital lobes and the cerebellum in both the left and right hemispheres of the exposed bone. Then screw eight electrocorticogram electrodes into the holes for focal epilepsy detection.In a separate group of rats, drill three holes and place two screw electroencephalogram electrodes over the right frontal and parietal lobes of the cortices with the same coordinates as in the first group. Next, place a third electroencephalogram electrode over the left cerebellum to ground the animal and reduce the signal artifacts. Separate the neck skin and muscle and insert two electromyogram electrodes into the neck muscle.Drill another hole on the skull and place a microinjection guide canula into the left central nucleus of the amygdala. Then, drill a 1.6 millimeter hole in the skull and insert a calibrated 30 kilovolt thermoster on the surface of the parietal cortex to monitor the cortical temperature. Route the insulated electrocortico and electromyogram electrode leads to a pedestal.Using dental acrylic to cement the pedestal, guide the canula to the skull and then connect the thermoster to the tether. Then, treat the incision topically with polysporin to prevent infection and give the animals water supplemented with ibuprofin and penicillin G for one week after surgery. To induce focal temporal lobe epilepsy, use a microinjection pump to administer 0.5 microliters of pilocarpine into the left central nucleus of the amygdala through the injection guide canula at 0.2 microliters per minute.To induce generalized epilepsy with recurrent status epilepticus, inject 300 milligrams per kilogram of pilocarpine IP.For acupuncture, first manually locate the acupoints. For example, the feng chi acupoints are located three millimeters away from the posterior median line between the ears on the neck. After confirming a low skin impedance at the acupoints of interest, insert stainless steel needles into the acupoints at a two millimeter depth.Then, using a functional electrical stimulator, deliver a 10 hertz train of 150 millisecond byphasic pulses with a one milliamp intensity to the acupoints through the needle. Here, a predominant epileptiform electrocorticogram acquired from the electrocorticogram electrode on the parietal lobe of the left hemisphere is shown. After pilocarpine administration, rare epileptic activities are picked up from the rest of the electrocorticogram electrodes, indicating a successful focal temporal lobe epilepsy induction.Within five minutes, severe cholinergic effects on behavior occur, gradually increasing in severity until an electrocorticogram generalized seizure occurs followed by a status epilepticus event. No epileptiform electrocorticograms are observed during the dark period after the first pentilene tetrasol injection, or during the second dark period on an off-day for injection until the fifth pentilene tetrasol injection, along with spontaneously and recurrently generalized epilepsy. Wakefulness exhibits a high amplitude electromyogram and an abundance of locomotor activity, with a gradual increase in the cortical temperature as the vigilant state transits from either non-REM or REM sleep to wakefulness.Non-REM sleep is characterized by synchronized electrocorticograms with a high amplitude and low frequency, with a gradual electromyogram amplitude decline and no locomotor activity, as well as a decrease in the cortical temperature as the vigilant state transits from wakefulness into NREM sleep. During REM sleep, the electrocorticogram wave is desynchronized with a reduced amplitude. The predominant electroencaphalogram power density occurs within the theta frequency.The electromyogram activity is the lowest, phasic body twitch is observed, and the cortical temperature rapidly increases. Administration of 100 hertz of electroacupuncture into the bilateral feng chi acupoints exacerbates the pilocarpine induced epileptiform electrocorticograms, while 10 hertz of electroacupuncture suppresses the epileptiform electrocorticograms. Further, pilocarpine administration into the left central nucleus of the amygdala also suppresses non-REM sleep during the light period of the 12-hour light/dark cycle.100 hertz of electroacupuncture further deteriorates the pilocarpine induced suppression of non-REM sleep. In contrast, a 10 hertz electroacupuncture into the bilateral feng chi acupoints increases non-REM sleep per se during the dark period, and blocks the pilocarpine-induced reduction of non-REM sleep during the light period. Once mastered, this technique can be completed in one and a half hours if it is performed properly.Before attempting this procedure, it is important to practice the skill of stereotaxic surgery until proficiency. After its development, this technique paves the way for the researchers in the field of neuroscience to explore the effects of acupuncture in epilepsy and sleep disturbance. After watching this video, you should have a good understanding of how to surgically implant electroencephalo and electromyogram electrodes, a guide canula, and a semester to assess sleep patterns and other effects of acupuncture on brain activity.
