In het bovenstaande protocol, hebben we aangetoond een doelmatige interventie om neuroanatomisch tekorten na neonatale blootstelling aan alcohol te redden. Kippen in verschillende omgevingen is lang bekend dat veranderingen in verschillende maatregelen van neurologische wellness veroorzaken. Veel studies kijken naar de positieve effecten van het grootbrengen in een complexe omgeving (EG) te beginnen met baanbrekend onderzoek van Diamond en Rosenzweig (bv 1, 2) en Greenough (Bijvoorbeeld 3, 4). EG is aangetoond dat een onmiskenbaar positief effect op synaptische en cellulaire veranderingen in de hersenen 5, 6, 7 hebben. EG kan een veelvoud aan hersengebieden beïnvloeden zoals de hippocampus 8, 9 en visuele cortex 10, 11, ventrale striatum 12, 13, enals brain-breed neuro-functie (beoordeeld in 14). Bijzondere belangstelling heeft zich ontwikkeld van de studies hippocampus toen werd aangetoond dat EG de overleving van volwassen geboren granule cellen van de dentate gyrus kan verhogen door dendritische plasticiteit 9, 13. Dit laatste punt is er veel belangstelling verzameld als gevolg van de groeiende hoeveelheid literatuur die aangeeft dat cardiovasculaire training bevordert volwassen neurogenese in zowel de gezonde en beschadigde hersenen 15, 16, 17, 18. Wiel draait (WR) is een eenvoudig te implementeren op vrijwillige basis cardiovasculaire activiteit die is gebleken bij diermodellen van neurologische aandoeningen of veroudering 17, 19, 20 te zijn. WR beïnvloedt de expressie van groeifactoren<html> in zowel het centrale als perifere zenuwstelsel 21, 22, 23. De combinatie van (later) WR en EC in een &quot;superintervention&quot; (WR-EC) (dat wil zeggen 12 dagen van WR, gevolgd door 30 dagen in de EG) biedt een robuuste stijging van de hippocampus volwassen neurogenese en verhoogde overleving van de pas verspreidden cellen 8, de blijkt dat in het diermodel van FASD niet wordt bereikt door individuele componenten (zie hieronder). Aangezien beide onderdelen van de WR-EC gevolgen hebben voor een divers scala aan structuren binnen de hersenen 13 (WR beoordeeld in 22, EC beoordeeld in 24), de implementatie van deze interventie kan eenvoudig worden toegepast op knaagdieren modellen van zowel de ontwikkelings- en het latere leven ontstaan modellen van neurologische nierfunctie (bv neonatale blootstelling aan alcohol, vergrijzing, vroege leven stress). nt &quot;&gt; Integratie van WR-EC in de adolescentie en vroege volwassen periode (dat wil zeggen, postnatale dagen 30-72) kan een aantal van de negatieve effecten van een rat model van foetale alcohol spectrum stoornissen (FASDs) te verbeteren 8 Een verzameling van studies hebben. aangetoond dat knaagdieren blootgesteld aan alcohol van postnatale dag (PD) 4 tot 9 tonen grote tekortkomingen neuroanatomical maatregelen zoals dendritische complexiteit 25, cerebellaire ontwikkeling 26, 27 en neuroimmune responsiviteit 28 en manifestaties van verminderde leren en geheugen 29, 30, 31 . Zelfs een verminderde hoeveelheid alcohol blootstelling binnen dit tijdvenster (dwz PD 7 tot en met 9) kan leiden tot tekorten in het leren en het geheugen bij adolescente en volwassen ratten 32, terwijl sommige structuren niet meer zien sigcante neuroanatomische impairment 27. Veel van deze tekorten - in aanvulling op gedrags impairments in hippocampus-afhankelijke taken - zijn verzacht na blootstelling aan deze WR-EC paradigma 8, 33 of WR alleen 25, 31. Hoewel WR staan een veelgebruikt interventie is de combinatie van WR-EC nog niet toegepast in de literatuur, ondanks het vermogen om de voordelen van WR 8 relatief korte termijn te ondersteunen. Dit artikel zal de uitvoering van de WR-EG-interventie tijdens de adolescentie te bespreken. Hoewel dit paradigma wordt gebruikt in de context van vroege postnatale blootstelling aan alcohol kan worden ingevoerd om verschillende knaagdiermodellen te hersenpotentiaal van neuroplasticiteit van modellen van hersenaandoeningen beoordelen.

Aerobic oefening (bv wiel lopen (WR) op grote schaal gebruikt in proefdieronderzoek) een positieve invloed veel maatregelen van neuroplastic potentieel in de hersenen, zoals de tarieven van de volwassen neurogenese, angiogenese, en de expressie van neurotrofe factoren bij knaagdieren. Deze ingreep is ook aangetoond gedrags- en neuroanatomische aspecten van de negatieve effecten van teratogens (dwz ontwikkelingsstoornissen blootstelling aan alcohol) en leeftijdsgebonden neurodegeneratie bij knaagdieren beperken. Omgevingscomplexiteit (EC) is aangetoond dat vele voordelen neuroplastic corticale en subcorticale structuren te produceren en kan worden gekoppeld wiel draait om de proliferatie en overleving van nieuwe cellen in de volwassen hippocampus verhogen. Wij zullen de uitvoering van WR / EG en haar in te besprekenterventions voor gebruik als een krachtigere therapie in ratten met behulp van het diermodel van prenatale blootstelling aan alcohol bij de mens. We zullen ook bespreken welke elementen van de procedures zijn absoluut noodzakelijk zijn voor de interventies en welke kan worden gewijzigd afhankelijk van de vraag of faciliteiten van de experimentator.

The overall goal of this behavioral intervention is to enhance environmental stimulation of laboratory rodents via exposure to voluntary cardiovascular exercise and environmental complexity in a controlled laboratory setting. This technique can help answer key questions in the field of rehabilitation and neuroplasticity. It highlights the importance of social exercise and exploratory behaviors on neurological development in adolescence and health maintenance in adulthood.The main advantage of this procedure is that it combines the exercise and environmental complexity in a synergistic way, and it allows the interpretation of each of the components separately if need be. To begin the experiment, place weaned rat pups into an appropriate cage and make sure that they always have free access to food and water. A week later, on day 30 of their development, move half the rats into exercise cages with running wheels.Each running wheel should have a revolution counter and be freely accessible. On the same day, weigh all the rats. Every following day at the same hour, check the number of revolutions on the running wheels.On day 36, weigh the rats again. Then, leave them in their respective housing condition for 12 days until day 42. Early on developmental day 42, prepare the environmentally complex cages.The 30 by 18 by 36 inch galvanized steel cages should contain multiple levels capable of supporting the weight of multiple rats. These cages must also be capable of supporting standard bedding and multiple food and water dispensers. Place novel colorful objects of variable sizes and shapes in the cage.First, position six large toys in the cage that are large enough for three or more rats to interact with concurrently. Next, place six medium-sized toys in the cage each of which should also be large enough for three or more rats to use concurrently. Then, place at least 20 small toys in the cage that might only occupy one rat at a time.Select toys of different colors and shapes to enhance the complexity. The novelty of each toy is critical to this form of intervention. At the opposite ends of the cage, place food dispensers and attach water bottles so each cage has two of each.Once the cages are prepared, weigh all the animals and relocate them to the environmentally complex cages. Group nine to 12 animals per cage, making certain that they all have unique identifiers. Every two days, replace all the toys.Make sure to have enough toys such that animals rarely encounter the same toy twice during the paradigm. When replacing the toys, also refill the food and water. Every third day, clean the cages.Temporarily house the rats in groups of two or three and replace the bedding, food, and water. Return the same toys to the cage on these days unless cleaning coincides with the toy replacement schedule. The wheel running and complex environmental interventions impacted adult neurogenesis in the dentate gyrus as assessed by BrdU staining.Increases in cell proliferation were observed following the wheel running portion of the intervention in the normally developing early life stressed and alcohol-exposed animals. The complex environment intervention increased the survival of adult-generated cells in the dentate gyrus in animals that were exposed to either stress or alcohol neonatally. Furthermore, the interventions resulted in an increase in cells that differentiate into a neuronal phenotype as seen with increases in adult-born dentate gyrus granule cells despite neonatal exposure to alcohol or intubation stress.Investigating the length of double cort and positive dentrites of dentate gyrus granule cells showed that there was no difference between the alcohol-exposed rats and control rats after wheel running and environmental complexity intervention. After watching this video, you should have a really good understanding of how to appropriately lay out the environmental complexity cage as well as how to house animals in the wheel running cages and environmental complexity cages. When attempting this procedure, it's important to thoroughly clean all of your materials and check up on your animals daily.Following this procedure, the behavioral, neurophysiological, and neuroanatomical measures can be assessed in order to determine the consequences of exposure to wheel running and environmental complexity interventions. After its development, this technique paved the way for researchers in the field of neuroplasticity to explore experience-dependent changes in rodent models of neurological disease.
