In dem obigen Protokoll haben wir gezeigt, eine sinnvolle Intervention folgende neuroanatomischen Defizite Neugeborenen Alkoholexposition zu retten. Aufzucht in verschiedenen Umgebungen ist seit langem bekannt in verschiedenen Maßnahmen von neurologischen Wellness Veränderungen zu verursachen. Viele Studien Blick auf die positiven Auswirkungen der in einer komplexen Umgebung (EG) Aufzucht mit bahnbrechende Forschung von Diamond und Rosenzweig beginnen (zB 1, 2) und Greenough (Beispielsweise 3, 4). EG hat sich gezeigt , 6 im Gehirn 5, eine unbestreitbare positive Auswirkungen auf die synaptische und zelluläre Veränderungen zu haben, 7. EG kann eine Vielzahl von Hirnregionen beeinflussen , einschließlich der Hippocampus 8, 9 und visuellen Kortex 10, 11, ventralen Striatum 12, 13, sowiewie Gehirn weite Neuroimmun - Funktion (in 14 überprüft). Von besonderem Interesse ist aus den Untersuchungen an Hippocampus entwickelt , wenn es wurde gezeigt , dass die EG die Überlebensrate der erwachsenen geborenen Körnerzellen des Gyrus dentatus durch dendritische Plastizität 9, 13 zu erhöhen. Dieser letzte Punkt hat angibt viel Interesse auf Grund der wachsenden Körper der Literatur gesammelt , dass Ausdauertraining adulten Neurogenese sowohl in der gesunden und geschädigten Gehirn 15, 16, 17, 18 fördert. Radfahr (WR) ist eine einfache Form der freiwilligen kardiovaskuläre Aktivität zu implementieren , die in Nagetiermodellen von neurologischen Erkrankungen als vorteilhaft gezeigt hat, oder Alterungs 17, 19, 20. WR wirkt sich auf die Expression von Wachstumsfaktoren<html> sowohl im zentralen und peripheren Nervensystem 21, 22, 23. Die Kombination von (später) WR und EG in eine &quot;superintervention&quot; (WR-EC) (dh 12 Tagen um 30 Tage in EG des WR) bietet eine robuste Zunahme des Hippocampus - adulten Neurogenese und ein verlängertes Überleben der neu proliferierten Zellen 8, die die Wirkung der einzelnen Komponenten erreicht im Tiermodell von FASD nicht (siehe unten). Da beide Komponenten von WR-EC eine Vielfalt von Strukturen innerhalb des Gehirns 13 beeinflussen (WR in 22 überprüft, überprüft EC in 24), die Umsetzung dieser Maßnahme leicht zu Nagetiermodellen beider Entwicklungs- und späteren Leben Beginn Modelle von neurologischen angewendet werden kann , Beeinträchtigung (zB neonatale Exposition Alkohol, Altern, Stress frühen Leben). nt &quot;&gt; Integration von WR-EC in den Jugendlichen und jungen Erwachsenen Perioden (dh postnatalen Tag 30 bis 72) können einige der negativen Auswirkungen eines Rattenmodell der fetalen Alkohol - Spektrum - Störungen verbessern (FASDs) 8 Eine Sammlung von Studien haben. gezeigt , dass 31 bis Alkohol aus postnatalen Tag (PD) 4 bis 9 Anzeige erhebliche Defizite in neuroanatomischen Maßnahmen wie dendritischen Komplexität 25, des Kleinhirns Entwicklung 26, 27 und Neuroimmun - Ansprechbarkeit 28 sowie Manifestationen von beeinträchtigter Lernen und Gedächtnis 29, 30, ausgesetzt Nagetiere . Viele dieser Defizite - zusätzlich zu Verhaltensstörungen bei Hippocampus-abhängigen Aufgaben - wurden 8, 33 oder WR allein 25, 31 nach Exposition mit diesem WR-EC Paradigma gemildert. Obwohl allein WR eine weit verbreitete Intervention gewesen ist, hat sich die Kombination von WR-EG noch nicht in der Literatur trotz seiner Fähigkeit verwendet worden 8 die relativ kürzer fristigen Vorteile der WR aufrecht zu erhalten. Dieser Artikel wird die Umsetzung des WR-EG-Intervention während der Adoleszenz zu diskutieren. Obwohl dieses Paradigma im Kontext der frühen postnatalen Exposition Alkohol verwendet wird, kann es zu verschiedenen Nagetiermodellen eingeführt werden, um Gehirnpotenziale neuroplasticity in den Modellen von Hirnerkrankungen zu beurteilen.

Aerobic - Übungen (zB Rad läuft (WR) ausgiebig in der Tierforschung) einen positiven Einfluss auf viele Maßnahmen von neuroplastische Potenzial im Gehirn, wie der Zahl der adulten Neurogenese, Angiogenese, und die Expression von neurotrophen Faktoren bei Nagern. Dieser Eingriff wurde auch Verhaltens- und neuroanatomische Aspekte der negativen Auswirkungen von teratogens (dh Entwicklungs Exposition gegenüber Alkohol) und altersbedingte Neurodegeneration bei Nagetieren zu mildern gezeigt. Umwelt Komplexität (EG) wurde mit zahlreichen neuroplastische Vorteile in kortikalen und subkortikalen Strukturen zu erzeugen, dargestellt und können mit Radfahr gekoppelt werden, um die Proliferation und das Überleben von neuen Zellen im erwachsenen Hippocampus zu erhöhen. Die Kombination dieser beiden Maßnahmen bietet eine robuste &quot;superintervention&quot; (WR-EC), die in einem Bereich von Nagetiermodellen von neurologischen Störungen durchgeführt werden kann. Wir werden die Umsetzung von WR / EG und ihren konstituierenden diskutieren inInterventionen für den Einsatz als leistungsfähigeren therapeutische Intervention bei Ratten, die das Tiermodell der pränatalen Exposition gegenüber Alkohol beim Menschen verwendet wird. Wir werden diskutieren, auch, welche Elemente der Verfahren unbedingt notwendig sind für die Eingriffe und welche verändert werden kann, abhängig von der Frage des Experimentators oder Einrichtungen.

Um die Wirkung des Super Intervention beurteilen zu können, müssen wir die Auswirkungen jedes seiner Bestandteile zu finden - WR und EC - auf unserer Maßnahmen von Interesse. The overall goal of this behavioral intervention is to enhance environmental stimulation of laboratory rodents via exposure to voluntary cardiovascular exercise and environmental complexity in a controlled laboratory setting. This technique can help answer key questions in the field of rehabilitation and neuroplasticity. It highlights the importance of social exercise and exploratory behaviors on neurological development in adolescence and health maintenance in adulthood.The main advantage of this procedure is that it combines the exercise and environmental complexity in a synergistic way, and it allows the interpretation of each of the components separately if need be. To begin the experiment, place weaned rat pups into an appropriate cage and make sure that they always have free access to food and water. A week later, on day 30 of their development, move half the rats into exercise cages with running wheels.Each running wheel should have a revolution counter and be freely accessible. On the same day, weigh all the rats. Every following day at the same hour, check the number of revolutions on the running wheels.On day 36, weigh the rats again. Then, leave them in their respective housing condition for 12 days until day 42. Early on developmental day 42, prepare the environmentally complex cages.The 30 by 18 by 36 inch galvanized steel cages should contain multiple levels capable of supporting the weight of multiple rats. These cages must also be capable of supporting standard bedding and multiple food and water dispensers. Place novel colorful objects of variable sizes and shapes in the cage.First, position six large toys in the cage that are large enough for three or more rats to interact with concurrently. Next, place six medium-sized toys in the cage each of which should also be large enough for three or more rats to use concurrently. Then, place at least 20 small toys in the cage that might only occupy one rat at a time.Select toys of different colors and shapes to enhance the complexity. The novelty of each toy is critical to this form of intervention. At the opposite ends of the cage, place food dispensers and attach water bottles so each cage has two of each.Once the cages are prepared, weigh all the animals and relocate them to the environmentally complex cages. Group nine to 12 animals per cage, making certain that they all have unique identifiers. Every two days, replace all the toys.Make sure to have enough toys such that animals rarely encounter the same toy twice during the paradigm. When replacing the toys, also refill the food and water. Every third day, clean the cages.Temporarily house the rats in groups of two or three and replace the bedding, food, and water. Return the same toys to the cage on these days unless cleaning coincides with the toy replacement schedule. The wheel running and complex environmental interventions impacted adult neurogenesis in the dentate gyrus as assessed by BrdU staining.Increases in cell proliferation were observed following the wheel running portion of the intervention in the normally developing early life stressed and alcohol-exposed animals. The complex environment intervention increased the survival of adult-generated cells in the dentate gyrus in animals that were exposed to either stress or alcohol neonatally. Furthermore, the interventions resulted in an increase in cells that differentiate into a neuronal phenotype as seen with increases in adult-born dentate gyrus granule cells despite neonatal exposure to alcohol or intubation stress.Investigating the length of double cort and positive dentrites of dentate gyrus granule cells showed that there was no difference between the alcohol-exposed rats and control rats after wheel running and environmental complexity intervention. After watching this video, you should have a really good understanding of how to appropriately lay out the environmental complexity cage as well as how to house animals in the wheel running cages and environmental complexity cages. When attempting this procedure, it's important to thoroughly clean all of your materials and check up on your animals daily.Following this procedure, the behavioral, neurophysiological, and neuroanatomical measures can be assessed in order to determine the consequences of exposure to wheel running and environmental complexity interventions. After its development, this technique paved the way for researchers in the field of neuroplasticity to explore experience-dependent changes in rodent models of neurological disease.
