Importance IR est l&#39;une des principales causes des ulcères de pression de la peau. Les premiers stades (I et II) des escarres de décubitus décrivent l&#39;état de la peau humaine (par rapport aux tissus sous-cutanés sous-jacents et les muscles). Cependant, la compréhension de l&#39;étiologie immunologique fait encore défaut. Ischémie- reperfusion (IRI) se produit lorsqu&#39;il y a une hypoxie transitoire due à l&#39;obstruction de la circulation sanguine (ischémie), suivie d&#39;une ré-oxygénation subséquente des tissus (reperfusion). Dans la peau, l&#39;ischémie-reperfusion (IR) est considéré comme l&#39;un des facteurs qui contribuent à la physiopathologie des ulcères de pression, où alitement prolongé prédispose les patients hospitalisés à long terme à des blessures. Chez ces patients, la peau et les muscles sous - jacents sont constamment exposés à la pression du poids exercé sur les zones de proéminence osseuse, entraînant des blessures localisées qui, l&#39; absence de traitement, peuvent devenir nécrotiques 1. Les dommages-intérêts impliqués dans un IRI sont doubles. Au cours de l&#39;ischémie, l&#39;occlusion des vaisseaux sanguins conduit à une baisse drastique de l&#39;apport d&#39;oxygène aux tissus. Cela se traduit par une diminution de l&#39;ATP et du pH, qui inactive les ATPases impliqués dans le métabolisme cellulaire. La libération des contenus intracellulaires ou des motifs moléculaires de dommages associés (DAMP), comme la HMGB1, contribue à la réponse inflammatoire 3. La seconde insulte se produit lors de la reperfusion. Bien que les niveaux d&#39;oxygène et de pH sont restaurées au cours de la reperfusion, cela se traduit par la génération d&#39;espèces réactives de l&#39;oxygène (ROS), ce qui conduit à l&#39;oxydation des lipides intracellulaires, l&#39;ADN et les protéines. Par conséquent, les médiateurs pro-inflammatoires sont activés, ce qui déclenche une réponse inflammatoire secondaire qui comprend le recrutement de cellules immunitaires vers le site inflammatoire 2. Alors que la cascade d&#39;événements biochimiques conduisant à la réponse inflammatoire a été bien décrite, la régulation spatiale et temporelle des activités de cellules immunitaires ne sont pas bien compris. Ici, nous décrivons un modèle IR robuste sur la peau de l&#39;oreille de souris à l&#39;aide de serrage simple aimant. Couplé avec l&#39;imagerie multiphotonique intravitale (MP-IVM), nousmis en place un modèle pour étudier les réponses inflammatoires in vivo qui se produisent après la reperfusion a lieu. La raison derrière le développement et l&#39;utilisation de cette technique est d&#39;essayer de comprendre comment les cellules interstitielles et infiltrant répondent à IR en temps réel. Les modèles existants de IR en utilisant la technique de serrage sur le flanc de la peau sont très envahissantes, car elles nécessitent l&#39;implantation chirurgicale de plaques d&#39;acier dans le flanc de la peau, ce qui les rend moins-que-idéal pour les études immunologiques 4. Une technique de serrage non invasive similaire a été décrite dans le flanc de souris 5,6 de la peau. Toutefois, en raison de l&#39;incorporation de la composante d&#39;imagerie intravitale dans cette méthode, nous avons choisi à la place de la peau de l&#39; oreille que le site IR ciblé, car il évite les mouvements dus à la respiration et offre la stabilité lors de l&#39; imagerie 7,8. Par ailleurs, les sous-ensembles de leucocytes qui enjambent interstitiel sont identiques entre la peau de l&#39;oreille et le flanc de la peau, même si lales nombres et les proportions peuvent varier légèrement 9. Ainsi, la peau de l&#39;oreille représente un site d&#39;imagerie idéal. En outre, la plupart des données récupérées à partir de ces modèles IRI sont limités à des évaluations macroscopiques (de classement des ulcères) et les analyses microscopiques des indicateurs inflammatoires d&#39;extrémité 10. En utilisant ce modèle, la visualisation en temps réel de la réponse cellulaire des neutrophiles après la reperfusion dans la peau d&#39;une souris rapporteur fluorescent est activé. Un modèle d&#39;imagerie de l&#39; oreille intravitale précédemment publié est utilisé 8 avec des modifications supplémentaires (figures 1, 2).

inducing ischemia-reperfusion injury in the mouse ear skin for intravital multiphoton imaging of immune responses

Ischemia-reperfusion injury (IRI) occurs when there is transient hypoxia due to the obstruction of blood flow (ischemia) followed by a subsequent re-oxygenation of the tissues (reperfusion). In the skin, ischemia-reperfusion (IR) is the main contributing factor to the pathophysiology of pressure ulcers. While the cascade of events leading up to the inflammatory response has been well studied, the spatial and temporal responses of the different subsets of immune cells to an IR injury are not well understood. Existing models of IR using the clamping technique on the skin flank are highly invasive and unsuitable for studying immune responses to injury, while similar non-invasive magnet clamping studies in the skin flank are less-than-ideal for intravital imaging studies. In this protocol, we describe a robust model of non-invasive IR developed on mouse ear skin, where we aim to visualize in real-time the cellular response of immune cells after reperfusion via multiphoton intravital imaging (MP-IVM).
