using scaffold liposomes to reconstitute lipid-proximal protein-protein interactions in vitro

시험관 내에서 세포막 단백질 연구가 많이 소수성 막 횡단 도메인의 존재에 의해 복잡하게된다. 또한 이러한 연구를 복잡하게, 리포좀에 세제 - 가용화 된 막 단백질의 reincorporation 단백질 토폴로지를 적용하는 것은 불가능 확률 과정이다. 본 논문은 리포좀 기반의 발판을 이용하여 이러한 도전적인 기술에 대한 대체 방법을 제공합니다. 단백질의 용해성은 경막 도메인의 결실에 의해 강화되며, 이러한 아미노산 같은 그의 태그 같은 더링 잔기로 대체된다. 이것은 테더가 리포솜의 표면에 균일 한 단백질 토폴로지를 적용 (니트릴 로트리 아세트산 그의 태그 된 단백질 (NTA (니켈 2+))에 의해 조정 니켈 2+) 앵커링 그룹과 상호 작용한다. 필수 막 단백질 미토콘드리아 분열 인자 (MFF)와 Dynamin 관련 단백질 1 (Drp1) 사이의 상호 작용이 INVE이었다 상기 예가 제시이 골격 리포솜 법을 이용 stigated. 이 작품에서 우리는 효율적으로는 GTPase 활동을 자극 리포좀의 표면에 용해 Drp1을 모집 MFF의 능력을 증명하고있다. 또한 Drp1 특정 지질의 존재 MFF 장식 지질 템플릿 관 모양 할 수 있었다. 이 예는 구조 및 기능 분석을 사용하여 골격 리포좀의 효과를 보여주고 Drp1 활성을 조절하는데 MFF의 역할을 강조한다.
