Szkola et al. 21 ont développé un microréseau multiplex fiable et pas cher pour la détection simultanée de proteotoxins et de petites toxines, des composés qui pourraient être utilisés dans la guerre biologique. Ils ont détecté des concentrations de toxine ricine, avec une limite de détection de 3 parties par milliard, en moins de 20 min. Récemment, le CYANOCHIP, un biocapteur basé sur un microréseau d&#39;anticorps pour la détection in situ de cyanobactéries toxiques et non toxiques, a été décrite 22. Cette microarray permet l&#39;identification des cyanobactéries blooms potentiels, surtout dans les milieux aquatiques, qui sont difficiles à identifier au microscope. La limite de détection de la puce est de 10 mars 2 à 10 cellules pour la plupart des espèces, en tournant ce biocapteur dans un outil rentable pour la détection multiplex et l&#39; identification des cyanobactéries, même au niveau de l&#39; espèce. Toutes ces propriétés font l&#39;Antibotechnique de microarray dy, et en particulier la méthode présentée dans ce travail, une méthode plus rapide et plus simple par rapport aux techniques mentionnées ci-dessus. Ce travail présente deux exemples d&#39;expériences qui utilisent un biocapteur à base de puces à ADN-anticorps pour détecter la présence de cyanobactéries dans des échantillons de sol et d&#39; eau. Il est une méthode simple et fiable basé sur un format de dosage immunologique en sandwich qui nécessite des volumes d&#39;échantillons très petits et la préparation des échantillons très basique. La méthode nécessite un court laps de temps et peut facilement être effectuée sur le terrain.

Le réchauffement climatique et de l&#39;eutrophisation font certains écosystèmes aquatiques se comportent comme de véritables bioréacteurs qui déclenchent la croissance des cyanobactéries rapide et massive; cela a la santé pertinente et des conséquences économiques. De nombreuses souches de cyanobactéries sont des producteurs de toxines, et seulement quelques cellules sont nécessaires pour induire des dommages irréparables à l&#39;environnement. Par conséquent, les autorités de l&#39;eau du corps et les administrations ont besoin de systèmes d&#39;alerte précoce rapides et efficaces qui fournissent des données fiables pour appuyer leurs décisions préventives ou curatives. Ce manuscrit rapporte un protocole expérimental pour la détection sur le terrain des souches de cyanobactéries produisant des toxines en utilisant une puce de microréseau d&#39;anticorps avec 17 anticorps (ABS) avec résolution taxonomique (de CYANOCHIP). Ici, un fluorescent immunologique sandwich microarray multiplex (FSMI) pour la surveillance simultanée des 17 souches de cyanobactéries trouve fréquemment floraison dans les écosystèmes d&#39;eau douce, certains d&#39;entre eux les producteurs de toxines, est décrite. Ils sont ensuite filtrés (5-20 um) pour éliminer la matière grossière, et le filtrat est incubé avec Abs. Immunoréactions sont révélés par une incubation finale avec un mélange de 17 Abs marqué par fluorescence et sont lues par un détecteur de fluorescence portatif.
