De optimale MSC voor gebruik in sommige onderzoek en klinische toepassingen moet sterk worden geactiveerd om hun voordeel te maximaliseren, en bij voorkeur opgesteld onder chemisch gedefinieerde XF voorwaarden voor de afgifte van potentiële antigenen van xenogene mediumbestanddelen zoals FBS minimaliseren. Mesenchymale stamcellen / stromale cellen (MSC) hebben laten zien een groot potentieel voor diverse regeneratieve geneeskunde benaderingen. MSCs werden aanvankelijk geïsoleerd als een stromale component van beenmerg maar zijn inmiddels verkregen zoveel andere volwassen weefsels, zoals vetweefsel 1, 2, 3. Interessant is dat de belangrijkste isolatiewerkwijze omarmt de opmerkelijke eigenschap van MSCs strak hechten aan weefselkweek kunststof in de aanwezigheid van foetaal runderserum (FBS). Hoewel dit traditionele isolatietechniek maakt gemakkelijke en snelle expansie van MSCs in tweedimensionale (2D) cultuur, is ook erg kunstmatige en voorbij betekenis van de natieve driedimensionale (3D) omgeving waarvoor eventueel verlies van belangrijke cellulaire elementen 4, 5 , 6. Daarom is de studie van MSCs in 3D culturen, diezijn meer fysiologische dan de traditionele 2D-culturen, is ontstaan ​​in de zoektocht naar &quot;verloren / verminderd&quot; MSC kenmerken. Bovendien heeft grote belangstelling gestegen tot xeno-vrij (XF) chemisch welbepaalde voorwaarden te identificeren voor MSC cultuur en activering, en zodoende de cellen meer vatbaar voor klinische toepassingen. Veel studies zijn gepubliceerd waaruit de 3D cultuur van MSC&#39;s zowel in biomaterialen en als bolvormige aggregaten of sferoïden. MSCs in biomaterialen werden oorspronkelijk ontworpen voor weefselengineering om beschadigde weefsels te vervangen cel geplaatste steigers, terwijl sferoïde culturen van MSC&#39;s werden als manier om MSC gedrag in vivo na toediening van de cellen voor therapieën in preklinische of klinische proeven begrijpen 4, 5, 7. Interessant is dat MSC vorm bolletjes spontaan bij hechting aan weefselkweek plastic is niet toegestaan8, 9, 10. Traditioneel werd celaggregatie vergemakkelijkt door centrifugekolf methoden of vloeibare overlaytechnieken, die aanvankelijk zijn gebruikt in kankerbiologie bij pogingen om te proberen de tumor micro-omgeving nabootsen. Meer recent zijn andere methoden opgedoken die aantonen celaggregatie in kweekschalen vooraf gecoat met specifieke chemicaliën om cel-adhesie kunststof 4, 5, 6 voorkomen. Een van de eenvoudigste en goedkoopste methode MSC sferoïden te genereren is om ze in cultuur opknoping druppels, een techniek die vaak werd gebruikt om embryoiden organen uit embryonale stamcellen. Met opknoping druppel cultuur techniek wordt celhechting de weefselkweekkunststof voorkomen door suspenderen van de cellen in een medium druppel op de onderkant van een weefselkweekplaat deksel en waardoor zwaartekracht cel vergemakkelijken aggregatie in de top van de druppel. De sferoïde grootte kan gemakkelijk worden gemanipuleerd door het veranderen van de celconcentratie of druppelvolume, waardoor hangende druppel culturen bijzonder eenvoudig te bedienen. Vroege studies op de 3D cultuur van MSC&#39;s aangetoond radicale verschillen in de eigenschappen van de cellen in 3D vergelijking met hun tegenhangers 2D 6, 8, 9. Tegelijkertijd, rapporten aangetoond dat de gunstige effecten van MSC&#39;s in vivo gesteund op hun vermogen om geactiveerd door micro-omgevingsfactoren en in reactie worden, produceren anti-inflammatoire en immunomodulerende factoren 11. Interessant, veel van deze factoren, zoals prostaglandine E2 (PGE2), tumornecrose-factor gestimuleerde gen 6 (TSG6) en hepatocyt groeifactor (HGF) werden geproduceerd in veel grotere hoeveelheden door MSC sferoïden dan traditionele 2D MSC weg vrijmaakt voor de idee vangebruiken 3D culturen naar cellen 8, 12, 13 te activeren. Bovendien genactivering in 3D culturen bleek mechanismen herhalen, ten minste gedeeltelijk, van celactivering na injectie in muizen 12. Door het activeren van MSCs vóór hun gebruik in experimenten kunnen effecten van de cellen verlengd en prominent als de traditionele MSC effect in vivo vaak vertraagd en voorbijgaand en kan worden omschreven als &quot;hit and run&quot;. Gedurende de afgelopen jaren belangrijke onderbouwd met MSC sferoïden hebben aangetoond dat zij ontstekingsreacties kunnen onderdrukken en moduleren immuniteit in vivo door beïnvloeding effectorcellen zoals macrofagen, dendritische cellen, neutrofielen, en T-cellen die sferoïden een aantrekkelijke vorm van geprimede MSCs 2 , 3. Bovendien productie van anti-kanker moleculen, zoals interleukin-24 (IL-24) en tumor necrosis factor-gerelateerde apoptose inducerende ligand (TRAIL), zijn verhoogd bij 3D culturen van MSC&#39;s ten opzichte van MSC monolaag, een fenomeen dat kan worden benut voor gerichte kankertherapieën 8, 10, 14. Aangezien de traditionele cultuur MSC vereist niet alleen het gebruik van weefselkweek plastic maar FBS, andere hindernis MSC sferoïden ontvankelijker te maken voor klinisch gebruik moesten worden overwonnen. Om deze hindernis te pakken, hebben we onlangs toonde vorming van MSC sferoïden onder specifieke omstandigheden chemisch gedefinieerde XF en vastgesteld dat de verkregen MSC sferoïden werden geactiveerd met dezelfde anti-inflammatoire en anti-kanker moleculen als sferoïden gegenereerd onder omstandigheden met FBS 14 produceren. Hier worden deze bevindingen in een aantal gedetailleerde protocollen die de generatie van de pre-geactiveerde MSC&#39;s te tonen in 3D culturen met behulp XFmedia. Daarnaast zijn protocollen geleverd dat manieren om het activeringsniveaus van de MSC&#39;s met betrekking tot hun ontstekingsremmende, immunomodulerende en anti-kanker effecten te evalueren, alsmede een praktische methode om de intacte bolletjes geven in muizen beschreven.

Mesenchymale stamcellen / stromale cellen (MSC) houdt een grote belofte in biotechnologie en regeneratieve geneeskunde. MSC&#39;s kunnen uit verschillende volwassen weefsels worden geïsoleerd door hun sterke hechting aan weefselkweek plastic en vervolgens verder uitgebreid in vitro, meestal via foetaal runderserum (FBS). Omdat FBS kunnen veroorzaken MSCs op immunogene worden, zijn aanwezigheid in MSC kweken beperkt zowel klinische en experimentele toepassingen van de cellen. Daarom is het in dienst hebben studies chemisch gedefinieerde xeno-vrij (XF) media voor MSC culturen zijn zeer waardevol.
