restenose Volgende cardiovasculaire procedures (bypass operatie, angioplastie of stenting) is een significant probleem waardoor de duurzaamheid van deze procedures wordt verminderd. Alle revascularisatieprocedures worden geplaagd door restenose. Huidige strategieën ter voorkoming van restenose (drug-eluerende stents en geneesmiddelcoated ballonnen) remmen zowel vasculaire gladde spiercellen (VSMC) en endotheliale cel proliferatie (EC). Bijgevolg voorkomen deze interventies VSMC gemedieerde restenose, maar voorkomen ook de regeneratie van het endothelium. Zonder een intact endothelium, zijn patiënten verplicht te zijn op krachtige antiplatelet middelen om het risico op in situ trombose te verminderen met het risico op bloedingen complicaties. Een ideale therapie zou VSMC proliferatie remmen, terwijl het bevorderen van de regeneratie van het endothelium. Zo is er een noodzaak om tegelijkertijd de VSMC proliferatie- en endotheliale barrièrefunctie in n vivo te bestuderen. Momenteel zijn er ernstigeAl muismodellen van restenose 1 . Deze modellen omvatten carotid ligatie en femorale arterie draad verwonding 2 . Aortische modellen omvatten stentplaatsing 3 , ballonbesering 4 en aorta-allograft 5 . Alle huidige modellen zijn beperkt. Carotidligatie genereert een doorstroming gemedieerde neointimale laesie en heeft geen endotheel letsel. Bovendien hebben zowel carotide- als femorale slagaders veel vallen minder cellagen dan menselijke vaten, waardoor hun translatiewaarde wordt beperkt. De aorta van de muis die ongeveer 1,3 mm in diameter is, is het enige vat dat een klinisch relevante (coronaire) menselijke slagader benadert (3). Ondanks het translatiepotentiaal van muizen aorta modellen van ziekte, hebben huidige modellen beperkingen. Deze modellen vereisen geavanceerde microchirurgische vaardigheden en gespecialiseerde apparatuur zoals angioplastiekballonnen en stents. Hierin presenteren weEen nieuwe, reproduceerbare techniek om tegelijkertijd VSMC proliferatie te induceren en endotheliale barrièrefunctie te verstoren.

murine aortic crush injury: an efficient in vivo model of smooth muscle cell proliferation and endothelial function

Arteriële reconstructie, ofwel angioplastiek of bypass-operatie, omvat iatrogene trauma waardoor endotheliale verstoring en vasculaire gladde spiercellen (VSMC) proliferatie veroorzaakt worden. Gemeenschappelijke muizenmodellen bestuderen kleine vaten, zoals de carotis- en femorale slagaders. Hierin beschrijven we een in vivo systeem waarin zowel VSMC proliferatie als endotheliale barrière functie gelijktijdig in een groot vat kunnen worden beoordeeld. We bestudeerden het infrarenale aorta respons op letsel in C57BL / 6 muizen. De aorta werd gewond van de linker nierader naar de aorta bifurcatie door 30 transmurale verpletteringen van 5 seconden duur met een katoenstippel applicator. Morfologische veranderingen werden beoordeeld met conventionele histologie. Aorta wanddikte werd gemeten van het luminale oppervlak naar de adventitia. EdU-integratie en tellerverf met DAPI en alfa-actine werd gebruikt om VSMC proliferatie te demonstreren. Activatie van ERK1 / 2, een bekende moderator van intimale hyperplasievorming, was afschrikkelijkOntgonnen door Western Blot analyse. Het effect van ontsteking werd bepaald door immunohistochemie voor B-cellen, T-cellen en macrofagen . En gezichtsafdelingen van endothelium werden gesynchroniseerd met scan-elektronenmicroscopie (SEM). Endotheliale barriere functie werd bepaald met Evans Blue staining. Transmurale letsel leidde tot verdikking van de aorta wand. Deze verwonding veroorzaakte VSMC proliferatie, meest prominente bij 3 dagen na verwonding, en vroege activatie van ERK1 / 2 en verminderde p27 kip1 expressie. Schade leidde niet tot verhoogde B-cellen, T-cellen, of macrofagen infiltratie in de vat van het vat. Schade veroorzaakte deelement van de endotheliale cel en verlies van cel-celcontact. Schade resulteerde in een significant verlies van de endotheliale barrière functie, die na zeven dagen terug naar de basislijn kwam. Het murine transmurale stomp aorta schade model biedt een efficiënt systeem om zowel VSMC proliferatie als endotheliale barrière functie in een groot vat te bestuderen.
