cPILOT voorziet in de gelijktijdige meting van meer dan 12 unieke monsters. enhanced sample multiplexing of tissues using combined precursor isotopic labeling and isobaric tagging (cpilot)

Er is een toenemende vraag naar vele biologische monsters analyseren voor de ziekte van begrip en het ontdekken van biomarkers. Kwantitatieve proteomics strategieën waarmee gelijktijdige meting van meerdere monsters zijn wijdverspreid en experimentele kosten en tijd sterk verminderen. Ons laboratorium ontwikkelde een techniek genaamd gecombineerde precursor isotopische labeling en isobare tagging (cPILOT), welk monster multiplexen van traditionele isotopische labeling of isobare tagging benadering verbetert. Global cPILOT kan worden toegepast op monsters afkomstig uit cellen, weefsels, lichaamsvloeistoffen of hele organismen en geeft informatie over de relatieve abundanties eiwit in verschillende monstercondities. cPILOT werkt door 1) het gebruik van lage pH buffer condities selectief dimethylate peptide N-termini en 2) met behulp van hoge pH buffer omstandigheden primaire aminen lysineresiduen labelen met commercieel verkrijgbare reagentia isobare (zie Tabel Materialen / Reagentia). De mate vansample multiplexen verkrijgbaar is afhankelijk van het aantal gebruikte etiketten precursor en isobare tagging reagens. Hier presenteren we een 12-plex analyse die lichte en zware dimethylering gecombineerd met zes-plex isobare reagentia om 12 monsters van muizenweefsels analyseren één analyse. Verbeterde multiplexing is nuttig voor experimentele tijd en kosten en nog belangrijker verminderen, waardoor vergelijking tussen verschillende monstercondities (biologische replicaten, ziektestadium, drugs, genotypen, of longitudinale tijdstippen) met minder experimentele vertekeningen en fouten. In dit werk wordt de globale cPILOT aanpak waarbij de hersenen, het hart en de lever weefsels door biologische replica&#39;s te analyseren vanuit een ziekte van Alzheimer muismodel en wild-type controles. Global cPILOT kan worden toegepast op andere biologische processen te bestuderen en ingericht om sample multiplexen stijgen tot meer dan 20 monsters.

cPILOT gebruikt op amine gebaseerde chemie label peptiden aan de N-terminus en lysine- resten chemisch en verbetert monster multiplexing mogelijkheden. Figuur 2 toont representatieve MS-gegevens die worden verkregen uit een 12-plex cPILOT analyse van hersenen, hart en leverweefsel van een Alzheimer muismodel en wildtype controles.
