Es gibt eine steigende Nachfrage viele biologischen Proben für die Krankheit Verständnis und zur Entdeckung von Biomarkern zu analysieren. Globale cPILOT können auf Proben angewendet werden, die von Zellen stammen, Gewebe, Körperflüssigkeiten oder ganze Organismen und gibt Informationen über relative Proteinhäufigkeiten in verschiedenen Probenbedingungen. cPILOT arbeitet durch 1) Puffer mit niedrigeren pH Bedingungen unter Verwendung von Bedingungen , um selektiv dimethylate Peptid N-Termini und 2) mit hohem pH - Puffer mit im Handel erhältlichen Reagenzien isobaren primäre Amine der Lysinreste zu kennzeichnen (siehe Tabelle der Materialien / Reagenzien). Der Grad derProbe Multiplexen verfügbar ist abhängig von der Anzahl von Vorläufern Etikett verwendet, und das isobare Markierungsreagens. Hier stellen wir eine 12-Plex-Analyse unter Verwendung von leichten und schweren Dimethylierung kombiniert mit sechs-plex isobaren Reagenzien 12 Proben von Geweben der Maus in einer einzigen Analyse zu analysieren. Verbessern Multiplexen ist hilfreich zur Reduzierung experimentelle Zeit und Kosten, und was noch wichtiger ist, so dass in vielen Vergleichsprobenbedingungen (biologische Replikaten, Krankheitsstadium, medikamentöse Behandlung, Genotypen oder longitudinale Zeitpunkt) mit weniger experimentellen Bias und Irrtum. cPILOT verwendet Aminbasis Chemie zu Label-Peptide am N-Terminus chemisch und Lysinreste und verbessert Probe Multiplexing-Funktionen.
